The methods and instrumentation for carrying out precise measurements of the equivalent electrical parameters of piezoelectric crystals have been developed. An accuracy exceeding 0.05% has been achieved with low power dissipation in the crystal permitting the use of this system at low temperatures and particularly in applications concerning the detection of gravitational radiation.  相似文献

A quartz resonator thin film monitor of exceptionally high accuracy and resolution has been developed. It is capable of providing measurements with an effective resolution of 0.001 nm and a corresponding relative accuracy of about 1% for a 0.100-nm-thick film of gold. A miniature resonance absorption spectrometer is able to perform a routine monitoring in space of the line profile and intensity of the He i resonance line from the solar disk with a time resolution of about 1 min and a spectral resolution of 10(5). It consists of a predispersive spectrometer incorporating a helium gas absorption cell. The instrument has been designed to meet a small size and weight. It was tested on a rocket flight in June 1976. The construction, principles of operation, and laboratory calibration of the instrument are presented. Its capability to resolve details of the order of 0.3 pm FWHM and its sensitivity to the intensity of line wings three order of magnitudes fainter than the peak of the line have been established by laboratory investigation.  相似文献

Commercial preparations of Candida rugosa lipase (CRL) are mixtures of lipase isoforms used for the hydrolysis and synthesis of various esters. The presence of variable isoforms and the amount of lipolytic protein in the crude lipase preparations lead to a lack of reproducibility of biocatalytic reactions. Purification of crude CRL improve their substrate specificity, enantioselectivity, stability, and specific activities. The expression of the isoforms is governed by culture or fermentation conditions. Unfortunately, the nonsporogenic yeast C. rugosa does not utilize the universal codon CTG for leucine; therefore, most of the CTG codons were converted to universal serine triplets by site-directed mutagenesis to gain expression of functional lipase in heterologous hosts. Recombinant expressions by multiple-site mutagenesis or complete synthesis of the lipase gene are other possible ways of obtaining pure and different CRL isoforms, in addition to culture engineering. Protein engineering of purified CRL isoforms allows the tailoring of enzyme function. This involves computer modeling based on available 3-D structures of lipase isoforms. Lid swapping and DNA shuffling techniques can be used to improve the enantioselectivity, thermostability, and substrate specificity of CRL isoforms and increase their biotechnological applications. Lid swapping can result in chimera proteins with new functions. The sequence of the lid can affect the activity and specificity of recombinant CRL isoforms. Candida rugosa lipase is toxicologically safe for food applications. Protein engineering through lid swapping and rationally designed site-directed mutagenesis will continue to lead to the production of CRL isoforms with improved catalytic power, thermostability, enantioselectivity, and substrate specificity, while providing evidence for the mechanisms of actions of the various isoforms.  相似文献

Clinical adverse experience (AE) data are routinely evaluated using between group P values for every AE encountered within each of several body systems. If the P values are reported and interpreted without multiplicity considerations, there is a potential for an excess of false positive findings. Procedures based on confidence interval estimates of treatment effects have the same potential for false positive findings as P value methods. Excess false positive findings can needlessly complicate the safety profile of a safe drug or vaccine. Accordingly, we propose a novel method for addressing multiplicity in the evaluation of adverse experience data arising in clinical trial settings. The method involves a two-step application of adjusted P values based on the Benjamini and Hochberg false discovery rate (FDR). Data from three moderate to large vaccine trials are used to illustrate our proposed 'Double FDR' approach, and to reinforce the potential impact of failing to account for multiplicity. This work was in collaboration with the late Professor John W. Tukey who coined the term 'Double FDR'.  相似文献

Effects of organic solutes properties, including Henry's law constant (H), molecular weight (M), molar volume at normal boiling point (Vb), and solubility (S), on the usefulness of rate estimation by reference to a reference substance (i.e., reference substance concept) and on the evaporation rate were investigated by measuring the volatilization rate constant of organic solutes under different environmental conditions, including mixing and surfactants. It was found that if benzene was used as a reference substance, the ratio of the solute rate to that of benzene (F) becomes insensitive to water mixing, whether the solute possessed high or low Henry's law constant. In the presence of surfactants, however, the F value changes sharply as the solute solubility decreases. For benzene, toluene, ethylbenzene, and xylenes (BTEX), the F values are essentially constant irrespective of the presence of other chemicals (including organic compounds, surfactants, and salinity) and of the variation in temperature. On the other hand, the volatilization rates are closely related to molecular weight (solubility) in the existence of mixing (surfactants). Three different approaches, i.e., mass-transport theory, modified Knudsen equation, and reaction rate concept, were used to evaluate the dependence of solute volatilization rates on solute properties. It was concluded that the interaction between solute properties and environmental parameters might play a key role in the volatilization process of organic solutes under different environmental conditions.  相似文献

Listeria monocytogenes has been isolated from condensate-forming surfaces in food processing plants. The objective of this research was to observe the behavior of L. monocytogenes on condensate-covered stainless steel with a Pseudomonas putida biofilm. L. monocytogenes-containing biofilms, either with or without added chicken protein, were incubated in a high humidity chamber at 12 degrees C to allow formation of condensate. Samples were analyzed for attached and unattached L. monocytogenes and total plate count periodically for 35 days. Samples were also taken for microscopic observation of Listeria and bacterial extracellular polymeric substances (EPS). L. monocytogenes attached in significantly greater numbers (> 3-log difference) to surfaces with preexisting P. putida biofilms than to Pseudomonas-free surfaces. L. monocytogenes survived in the presence or absence of P. putida with no added nutrients for 35 days, with numbers of survivors in the range of 3 to 4 log CFU/cm2 in the presence of P. putida and less than 2.9 log CFU/cm2 in pure culture. Attached and unattached L. monocytogenes were at similar levels throughout the incubation under all conditions studied. The addition of protein to the biofilms allowed growth of L. monocytogenes in pure culture during the first 7 days of incubation. Numbers of L. monocytogenes were not affected by the presence of P. putida when protein was present. Unattached L. monocytogenes were at levels of 3.6 to 6.7 log CFU/cm2 on the protein-containing surfaces. Microscopic observation of the condensate-covered biofilms indicated that L. monocytogenes formed microcolonies embedded within an EPS matrix over a 28-day period. This research demonstrates that L. monocytogenes can survive on condensate-forming stainless steel in low and high nutrient conditions, with or without the presence of Pseudomonas biofilm. The Listeria can detach and, therefore, have the potential to contaminate product.  相似文献

Total mixed rations containing conventional forage sorghum, brown midrib (bmr)-6 forage sorghum, bmr-18 forage sorghum, or corn silage were fed to Holstein dairy cows to determine the effect on lactation, ruminal fermentation, and total tract nutrient digestion. Sixteen multiparous cows (4 ruminally fistulated; 124 d in milk) were assigned to 1 of 4 diets in a replicated Latin square design with 4-wk periods (21-d adaptation and 7 d of collection). Diets consisted of 40% test silage, 10% alfalfa silage, and 50% concentrate mix (dry basis). Acid detergent lignin concentration was reduced by 21 and 13%, respectively, for the bmr-6 and bmr-18 sorghum silages when compared with the conventional sorghum. Dry matter intake was not affected by diet. Production of 4% fat-corrected milk was greatest for cows fed bmr-6 (33.7 kg/d) and corn silage (33.3 kg/d), was least for cows fed the conventional sorghum (29.1 kg/d), and was intermediate for cows fed the bmr-18 sorghum (31.2 kg/d), which did not differ from any other diet. Total tract neutral detergent fiber (NDF) digestibility was greatest for the bmr-6 sorghum (54.4%) and corn silage (54.1%) diets and was lower for the conventional (40.8%) and bmr-18 sorghum (47.9%) diets. In situ extent of NDF digestion was greatest for the bmr-6 sorghum (76.4%) and corn silage (79.0%) diets, least for the conventional sorghum diet (70.4%), and intermediate for the bmr-18 sorghum silage diet (73.1%), which was not different from the other diets. Results of this study indicate that the bmr-6 sorghum hybrid outperformed the conventional sorghum hybrid; the bmr-18 sorghum was intermediate between conventional and bmr-6 in most cases. Additionally, the bmr-6 hybrid resulted in lactational performance equivalent to the corn hybrid used in this study. There are important compositional differences among bmr forage sorghum hybrids that need to be characterized to predict animal response accurately.  相似文献
