A novel method for measuring an adhesion force of single yeast cell is proposed based on a nanorobotic manipulation system inside an environmental scanning electron microscope (ESEM). The effect of ambient humidity on a single yeast cell adhesion force was studied. Ambient humidity was controlled by adjusting the chamber pressure and temperature inside the ESEM. It has been demonstrated that a thicker water film was formed at a higher humidity condition. The adhesion force between an atomic force microscopy (AFM) cantilever and a tungsten probe which later on known as a substrate was evaluated at various humidity conditions. A micro-puller was fabricated from an AFM cantilever by use of focused ion beam (FIB) etching. The adhesion force of a single yeast cell (W303) to the substrate was measured using the micro-puller at the three humidity conditions: 100%, 70%, and 40%. The results showed that the adhesion force between the single yeast cell and the substrate is much smaller at higher humidity condition. The yeast cells were still alive after being observed and manipulated inside ESEM based on the result obtained from the re-culturing of the single yeast cell. The results from this work would help us to understand the ESEM system better and its potential benefit to the single cell analysis research.  相似文献

Commercial medium density fiberboard (MDF) was produced using rubberwood and oil palm empty fruit bunch (OPEFB) based on oven dried weight. Relative humidities of 65%, 93% with fixed temperature at 20°C, and normal condition was used for storage of the boards. The effects of storage time and relative humidity of MDF were assessed by measuring the changes of the board mechanical and physical properties for 10 weeks at 2 week intervals. The mechanical tests involved were internal bonding and flexural test, whereas physical tests were moisture content, thickness swelling, water absorption, and scanning electron microscope. The result showed that at 65% humidity, MDF exhibited a lower effect on the mechanical and physical properties of the panel, whereas at 93% humidity, the panel exhibited a decrease in the dimensional stability and mechanical properties of the board. Possible mechanisms behind this reduction are discussed.  相似文献

The present study investigated the wax deposition tendencies of a light Malaysian crude oil(42.4° API), and the wax inhibiting potential of some surfactants and their blends with nanoparticles. With the knowledge that the majority of the wax inhibition research revolved around polymeric wax inhibitors, which cause environmental issues, we highlighted the potential of surfactants and their blend with SiO_2 nanoparticles as wax deposition inhibitors. Different surfactants including oil-based, silane-based, Gemini and bio-surfactants were considered as primary surfactants. The primary surfactants and their respective blends at a concentration of 400 ppm were screened as wax inhibitor candidates using cold finger apparatus. The screening results showed a significant influence on the paraffin inhibition efficiency on wax deposition by using 400 ppm of silane-based surfactant, which decreased the wax deposition up to 53.9% as compared to that of the untreated crude oil. The inhibition efficiency among the silane-based surfactant(highest) and bio-surfactant(lowest)revealed an appreciable difference up to 36.5%. Furthermore, the wax from the treated sample was found to deposit in a thin gel-like form, which adhered inadequately to the surface of the cold finger. A further investigation by blending the 400 ppm silane-based surfactant with a 400 ppm SiO_2 nanoparticle suspension in a load ratio of 3:1 found that the wax inhibition decreased up to 81% as compared to the scenario when they were not added. However, we have shown that the synergy between the silane-based surfactant and the nanoparticles is influenced by the concentration and load ratio of surfactant and nanoparticles, residence time, differential temperature and rotation rate.  相似文献
