Low cardiac output after open heart operations in neonates and infants carries a high mortality. Delayed sternal closure may be life-saving but may prolong hospital stay and increase costs. The phosphorylation of glucose to glucose-6-phosphate, catalyzed by hexokinase, is the first committed step in glucose uptake into skeletal muscle. Two isoforms of hexokinase, HKI and HKII, are expressed in human skeletal muscle, but only HKII expression is regulated by insulin. HKII messenger RNA, protein, and activity are increased after 4 h of insulin infusion; however, glucose uptake is stimulated much more rapidly, occurring within minutes. Studies in rat muscle suggest that changes in the subcellular distribution of HKII may be an important regulatory factor for glucose uptake. The present studies were undertaken to determine if insulin causes an acute redistribution of HKII activity in human skeletal muscle in vivo. Muscle biopsies (vastus lateralis muscle) were performed before and at the end of 30 min insulin infusion, performed using the euglycemic clamp technique. Muscle biopsies were subfractionated into soluble and particulate fractions to determine if insulin acutely changes the subcellular distribution of HKII. Insulin decreased HKII activity in the soluble fraction from 2.20 +/- 0.31 to 1.40 +/- 0.18 pmoles/(min[chempt]micrograms) and increased HKII activity in the particulate fraction from 3.02 +/- 0.46 to 3.45 +/- 0.46 pmoles/(min[chempt]micrograms) (P < 0.01 for both). These changes in HKII activity were correlated with changes in HKII protein, as determined by immunoblot analysis (r = 0.53, P = 0.05). Insulin had no effect on the subcellular distribution of HKII activity, which was primarily restricted to the soluble fraction. These studies are consistent with the conclusion that, in vivo in human skeletal muscle, insulin changes the subcellular distribution of HKII within 30 min. 相似文献
In this paper, a hybrid integrator backstepping controller is proposed for robotic manipulators actuated with brushless DC motors in the presence of arbitrary uncertain inertia parameters of the manipulator and the electrical parameters of the actuators. The main features are: 1) the design, requiring at most the joint velocities and stator currents feedback; 2) the semiglobal asymptotic stability of the controller being established in the Lyapunov sense; and 3) suitability of the scheme for multijoint robots. Particle clogging in the artificial groundwater recharge process is one of the main factors influencing the artificial groundwater recharge efficiency, and particle deposition is the microscopic mechanism of the occurrence and development of particle clogging. Particle deposition in porous media changes the pore structure. The computed tomography (CT) scanning technique is a nondestructive testing method and determines the spatial distribution of pores in porous media. This study combines physical and CT scanning experiments to identify the change process of the pore structure in the artificial groundwater recharge process and compares the pore changes during recharge experiments between two columns containing different media. Porous media changes are observed with the CT scanning technique. The fractal theory is applied in the analysis of CT scan images and physical experiment results. The results of this study indicate that particle deposition can be examined by using CT scan images to obtain pore-related parameters, the internal pore structure of porous media determined through CT scan images can be applied in numerical simulation, and a mathematical model for particle deposition calculation in porous media is established. Compared to the physical experiment measurements, the spatial particle deposition information acquired with the CT scanning technique exhibits a higher accuracy and contains much more relevant data. Not only does this research reveal more clearly the particle clogging mechanism which is based on particle deposition, but also characterize, simulate and predict more accurately the development tendency of particle clogging during artificial groundwater recharge.
