PGE2 is one of the key molecules in the osteoblast. It is the major prostanoid in the bone, and its production is under the control of both systemic and local factors. PGE2 has been reported to have multiple actions in the osteoblast, such as growth promotion and cell differentiation. To better understand the action of PGE2 in the osteoblast, we determined the PGE receptor subtypes in MC3T3-E1, an osteoblastic cell line derived from the normal mouse calvaria. Northern blot analysis revealed that EP1 and EP4 subtypes are expressed in MC3T3-E1. In contrast, EP3 subtype was not detected by either Northern blot analysis or RT-PCR. The contribution of each subtype was evaluated by studying the effects of subtype-specific analogs on osteoblastic function at confluency and 5 days after confluency. An EP1 agonist, 17-phenyl-omega-trinor PGE2, increased DNA synthesis and decreased alkaline phosphatase activity. 11-Deoxy-PGE1, and EP2 and EP4 agonist, decreased DNA synthesis and increased alkaline phosphatase activity at both stages. Butaprost, an EP2-selective agonist, showed effects similar to those of 11-deoxy-PGE1 only at confluency. Another and more differentiated osteoblastic marker, osteocalcin production, was detectable and was stimulated by 11-deoxy-PGE1 only 5 days after confluency. The exposure of these cells to EP1 agonist changed the cell shape to a more fibroblastic appearance. These results indicate that EP1, EP4, and probably EP2 are present in MC3T3-E1 cells; EP1 promotes cell growth, and EP2 and EP4 mediate differentiation of the osteoblast. Furthermore, the decreased response to EP2-specific agonist 5 days after confluency suggests that the expression of PGE receptor subtype is dependent on the stage of osteoblastic differentiation. This is the first report to determine PGE receptor subtypes in the bone.  相似文献

The relationship of virus load to clinical disease progression in HIV-infected children remains to be elucidated. In this study, HIV-1 proviral DNA load was determined in peripheral blood mononuclear cells (PBMCs) by the quantitative competitive DNA polymerase chain reaction assay (QC-PCR) in 47 HIV-infected children subdivided by age (group I, < or = 2 years; group II, > or = 5 years), who were further categorized to include 12 rapid progressors (RP, age < or = 2 years, Centers for Disease Control [CDC] defined clinical category C and/or immune category 3, or death before age 2 years) and slow progressors (SP, age > or = 5 years, excluding CDC categories C and/or immune category 3). Significantly higher mean proviral copies/10(3) PBMCs were detected in group I versus group II (75.4 +/- 104.3 and 13.0 +/- 17.8 respectively, p < 0.0001) and in RP (158.0 +/- 118.2) as compared to either SP (11.8 +/- 18.8, p < 0.0001) or other age-matched infected children (20.3 +/- 38.8, p < 0.0001). Thus HIV-infected children appear to have a higher cell-associated virus load early in life, especially in association with rapid disease progression.  相似文献

A mathematical model of complex heat exchange in the flame space of the melting zone of a regenerative bath furnace for sheet glass is presented. OBJECTIVE: To find the reasons which determine failures to comply with anti-flu vaccinations, so that these can be corrected and the coverage of this preventive action be increased. DESIGN: Observational crossover study, done by means of a telephone survey of people over 65. A questionnaire with closed questions, composed after a pilot study and validated by Cronbach's alpha. SETTING: Primary Care Centre (PCC). PATIENTS: We calculated a population sample for qualitative variables (_ = 0.05; p = 0.60; e = 0.05) of 294 people over 65, chosen from the PCC records, by means of random sampling (K = 4) stratified for age and discounting the telephone selection bias. MEASUREMENTS AND RESULTS: The proportion of vaccinated patients (60.9%) obtained in our study did not significantly differ from that in the general population. The percentage of patients included in the programme for the first time was 14%. Level of satisfaction among those vaccinated was 89.4%, with 8.9% of problems detected being light. Main causes of non-vaccination were: thinking that they didn't need it (63.5%), ignorance of the campaign (35.7%), fear of the reaction (24.3%), forgetting (10.4%). The main form of access to the campaign information was from the PCC, both through individuals and posters. Lack of information was statistically significant (p < 0.00001) as a determinant of non-vaccination, without other factors (age, sex, associated pathologies...) explaining these differences. CONCLUSIONS: Individualised and on-going health education by the PCC is fundamental. This would enable the identification of the group not vaccinated due to their express refusal and the recovery of non-vaccinated patients.  相似文献

Using power resources of the outer space and its industrialization have become an urgent task in the development of present day civilization. Solar energy is the most suitable basis for power supply for the majority of projects in the near-Earth space. Passing on to the large-scale space-based systems of power supply requires putting into life demonstration experiments in which power transmission by electromagnetic emission is supposed.In the paper the expedience of developing an autonomous power module which provides the possibility of space programs with great power consumption and wireless transmission system running-up is proved. Ukraine can design a solar power satellite of 10–20 kW power on the basis of the research satellite AUOS-SM. Two variants of design solutions for such satellite with solar arrays of great area are presented. Power transmission to the space vehicle-consumer can be conducted by cable as well as in a wireless way. The possibilities of placing microwave and laser energy transmission systems on board the satellite are analysed. It is shown that a power supply system for space vehicles with transmission distance of thousands km can be designed basing on modern lasers.Some experiments making use of great electric power generated by the plant of the satellite under consideration are proposed. The running-up of electric thrusters which are necessary for orientation and distancing is of great interest. Wireless power transmission may be carried on to a small satellite equipped with electric thrusters.  相似文献

For the noninvasive diagnosis of heart disease based on the acoustic and elastic characteristics of the heart muscle, it is necessary to transcutaneously measure small vibration signals, including components with an amplitude of less than 100 μm, from various parts of the heart wall continuously for periods of more than several heartbeats in a wide frequency range up to 1 kHz. Such measurement, however, has not been realized by any ultrasonic diagnostic methods or systems to date. By introducing the constraint least-square approach, this paper proposes a new method for accurately tracking the movement of the heart wall based on both the phase and magnitude of the demodulated signal to determine the instantaneous position of the object so that the vibration velocity of the moving object can be accurately estimated. By this method, small vibrations of the heart wall with small amplitudes less than 100 μm on the motion resulting from a heartbeat with large amplitude of 10 mm can be successfully detected with sufficient reproducibility in the frequency range up to several hundred Hertz continuously for periods of about 10 heartbeats. The resultant small vibration is analyzed not only in the time domain, but also in the frequency domain. As confirmed by the preliminary experiments herein reported, the new method offers potential for research in acoustical diagnosis of heart disease  相似文献

This paper presents a potentially practical treatment of dynamic operator-system interactions. The approach employs a dynamic event tree framework to explicitly address plant dynamics, system indications, crew-plant interactions, time available, crew cognition, errors of commission (mistakes), and multiple planning and diagnosis possibilities. The approach is applied in an analysis of a hypothesize medium break loss of coolant accident for a test reactor that occurs during plant shutdown. This transient was selected on the basis of: a significant cognitive component being present, high consequences being associated with operator actions, and the importance of event timing to scenario progression. The results presented show how quantitative risk predictions are affected by the treatment of dynamics, and demonstrate how non-proceduralized recovery actions and a number of performance shaping factors (e.g., crew experience, stress, and confidence) can be explicitly treated. Insights and lessons learned regarding the performance of a dynamic assessment are also presented.  相似文献
