Objective

We outline our vision for a 14 Tesla MR system. This comprises a novel whole-body magnet design utilizing high temperature superconductor; a console and associated electronic equipment; an optimized radiofrequency coil setup for proton measurement in the brain, which also has a local shim capability; and a high-performance gradient set.

Research fields

The 14 Tesla system can be considered a ‘mesocope’: a device capable of measuring on biologically relevant scales. In neuroscience the increased spatial resolution will anatomically resolve all layers of the cortex, cerebellum, subcortical structures, and inner nuclei. Spectroscopic imaging will simultaneously measure excitatory and inhibitory activity, characterizing the excitation/inhibition balance of neural circuits. In medical research (including brain disorders) we will visualize fine-grained patterns of structural abnormalities and relate these changes to functional and molecular changes. The significantly increased spectral resolution will make it possible to detect (dynamic changes in) individual metabolites associated with pathological pathways including molecular interactions and dynamic disease processes.

Conclusions

The 14 Tesla system will offer new perspectives in neuroscience and fundamental research. We anticipate that this initiative will usher in a new era of ultra-high-field MR.

In vitro research in the field of type I diabetes is frequently limited by the availability of a functional model for islets of Langerhans. This method shows that by the addition of theophylline to the glucose buffers, mouse insulinoma MIN6 and rat insulinoma INS1E pseudo-islets can serve as a model for islets of Langerhans for in vitro research. The effect of theophylline is dose- and cell line-dependent, resulting in a minimal stimulation index of five followed by a rapid return to baseline insulin secretion by reducing glucose concentrations after a first high glucose stimulation. This protocol solves issues concerning in vitro research for type I diabetes as donors and the availability of primary islets of Langerhans are limited. To avoid the limitations of using human donor material, cell lines represent a valid alternative. Many different β cell lines have been reported, but the lack of reproducible responsiveness to glucose stimulation remains a challenge.  相似文献

Silk fibers combine good stiffness and strength with a very high strain to failure and are as such highly promising to realize composites with high impact resistance. It is shown that to realize this potential it is quite beneficial to employ matrix materials of high strain to failure, particularly thermoplastic matrices. High impact resistance is thus achieved, well above the values for the pure matrices. Below the glass-transition temperature of the thermoplastic matrix, the impact energy absorption decreases. The adhesion between fiber and matrix also plays a significant role; lower adhesion typically increases the low-velocity penetration impact resistance, due to the spread of damage. Finally, the fiber architecture is pivotal; when a woven fabric is used which is unbalanced in strength, the impact resistance reduces in correspondence with the weakest material direction. A quasi-isotropic layup has a lower capacity for deformation than a balanced woven configuration which likely explains the observed lower penetration impact resistance.  相似文献

In this article, we describe the usage of persuasion profiles in a large scale, N = 1,129, field trial. Persuasive technologies—technologies intentionally designed to influence user behavior—are emergent and becoming more and more individualized and ubiquitous. Individual differences in people’s responses to often used persuasion principles—different psychological means by which to influence users—motivate personalization. We describe how, through identification, representation, and measurement, persuasive technologies can personalize their persuasive attempts. Next, we show that dynamically adapting a persuasive technology to the responses of its users increases the effectiveness of the system. Ubiquitous computing systems are, because of their ability to unobtrusively measure user behavior, very well suited for these types of applications.  相似文献

The world runs a serious risk of sliding into catastrophic climate change. It is to all practical purposes impossible to bring the emissions of greenhouse gases to a halt straight away. This article discusses a number of sources of uncertainty and the possibility of bias in climate forecasting. It identifies a specific approach towards rapid decarbonization combined with a systematic reduction in fossil fuel extraction: the manufacture of synthetic hydrocarbon compounds for use as fuel as well as plastics, using carbon dioxide from power stations and cement factories as feedstock during a transitional phase. Policymakers and even researchers working on renewable energy seem to be insufficiently aware of the potential of this mature chemical technology to reduce emissions. The paper also surveys several economic and managerial issues regarding the implementation of the transition to a world using only renewable energy, and provides some information concerning the latest state of the technology of renewable electricity generation.  相似文献

The development of immune protective islet encapsulation devices could allow for islet transplantation in the absence of immunosuppression. However, the immune protective membrane / barrier introduced there could also impose limitations in transport of oxygen and nutrients to the encapsulated cells resulting to limited islet viability. In the last years, it is well understood that achieving prevascularization of the device in vitro could facilitate its connection to the host vasculature after implantation, and therefore could provide sufficient blood supply and oxygenation to the encapsulated islets. However, the microvascular networks created in vitro need to mimic well the highly organized vasculature of the native tissue. In earlier study, we developed a functional macroencapsulation device consisting of two polyethersulfone/polyvinylpyrrolidone (PES/PVP) membranes, where a bottom microwell membrane provides good separation of encapsulated islets and the top flat membrane acts as a lid. In this work, we investigate the possibility of creating early microvascular networks on the lid of this device by combining novel membrane microfabrication with co-culture of human umbilical vein endothelial cell (HUVEC) and fibroblasts. We create thin porous microstructured PES/PVP membranes with solid and intermittent line-patterns and investigate the effect of cell alignment and cell interconnectivity as a first step towards the development of a stable prevascularized layer in vitro. Our results show that, in contrast to non-patterned membranes where HUVECs form unorganized HUVEC branch-like structures, for the micropatterned membranes, we can achieve cell alignment and the co-culture of HUVECs on a monolayer of fibroblasts attached on the membranes with intermittent line-pattern allows for the creation of HUVEC branch-like structures over the membrane surface. This important step towards creating early microvascular networks was achieved without the addition of hydrogels, often used in angiogenesis assays, as gels could block the pores of the membrane and limit the transport properties of the islet encapsulation device.  相似文献

Extrahepatic transplantation of islets of Langerhans could aid in better survival of islets after transplantation. When islets are transfused into the liver 60-70% of them are lost immediately after transplantation. An important factor for a successful extrahepatic transplantation is a well-vascularized tissue surrounding the implant. There are many strategies known for enhancing vessel formation such as adding cells with endothelial potential, the combination with angiogenic factors and / or applying surface topography at the exposed surface of the device. Previously we developed porous, micropatterned membranes which can be applied as a lid for an islet encapsulation device and we showed that the surface topography induces human umbilical vein endothelial cell (HUVEC) alignment and interconnection. This was achieved without the addition of hydrogels, often used in angiogenesis assays. In this work, we went one step further towards clinical implementation of the device by combining this micropatterned lid with Mesenchymal Stem Cells (MSCs) to facilitate prevascularization in vivo. As for HUVECs, the micropatterned membranes induced MSC alignment and organization in vitro, an important contributor to vessel formation, whereas in vivo (subcutaneous rat model) they contributed to improved implant prevascularization. In fact, the combination of MSCs seeded on the micropatterned membrane induced the highest vessel formation score in 80% of the sections.  相似文献

Software product lines are increasingly built using components from specialized suppliers. A company that is in the middle of a supply chain has to integrate components from its suppliers and offer (partially configured) products to its customers. To satisfy both the variability required by each customer and the variability required to satisfy different customers’ needs, it may be necessary for such a company to use components from different suppliers, partly offering the same feature set. This article presents three new approaches to prove termination of rewrite systems with the Knuth–Bendix order efficiently. The constraints for the weight function and for the precedence are encoded in (pseudo-)propositional logic or linear arithmetic and the resulting formula is tested for satisfiability using dedicated solvers. Any satisfying assignment represents a weight function and a precedence such that the induced Knuth–Bendix order orients the rules of the encoded rewrite system from left to right. This means that in contrast to the dedicated methods our approach does not directly solve the problem but transforms it to equivalent formulations for which sophisticated back-ends exist. In order to make all approaches complete we present a method to compute upper bounds on the weights. Furthermore, our encodings take dependency pairs into account to increase the applicability of the order. This paper discusses vectorization schemes that take advantage of the streaming‐SIMD‐extensions in commonly used floating‐point MIN and MAX reductions.
