A multiplex polymerase chain reaction (M-PCR) assay for Haemophilus ducreyi, Treponema pallidum, and herpes simplex virus (HSV) was compared with clinical and standard laboratory methods for the diagnosis of genital ulcer disease (GUD) in 105 patients; 36% were human immunodeficiency virus (HIV)-seropositive. Chancroid (80%), syphilis (8%), and genital herpes (8%) were the most frequent diagnoses. H. ducreyi and HSV were isolated from ulcers of 43% and 18% of patients, respectively; in 35%, all cultures were negative and the laboratory diagnosis indeterminate. M-PCR detected H. ducreyi, T. pallidum, and HSV in 56%, 23%, and 26% of patients, respectively; (no definitive diagnosis, 6%). The proportion of patients with more than one agent was 4% by culture and 17% by M-PCR (P = .002). Resolved sensitivities of M-PCR for H. ducreyi and HSV cultures were 95% and 93%, respectively. The sensitivities of H. ducreyi and HSV cultures were 75% and 60%, respectively. HSV, detected in 47% of specimens from HIV-infected versus 16% from HIV-uninfected patients (P < .001), may be emerging as a more frequent cause of GUD. 相似文献

A strategy for the structural characterization of the four major NaBH4-reduced peptidoglycan monomers derived from muramidase-digested peptidoglycan from the cyanelles of the flagellate Cyanophora paradoxa Korschikoff is described. Initial molecular weight determination of these glycopeptides was performed by positive and negative ion plasma desorption mass spectrometry. Due to the presence of two pairs of disaccharide tripeptide and disaccharide tetrapeptide monomers differing in mass by 112 units, respectively, an as yet unknown peptidoglycan modification either at the carbohydrate or at the peptide moiety was assumed. beta-Elimination of the disaccharide unit from the unreduced peptidoglycan monomers yielded the corresponding (modified) N1-lactyltripeptides and -tetrapeptides, respectively. These peptides, N-terminally blocked with lactic acid, unambiguously showed the modification to be located on the peptide moiety. By positive ion fast atom bombardment/hybrid tandem mass spectrometry of the reduced peptidoglycan monomers as well as of the corresponding deglycosylated monomers (= N1-lactylpeptides) the modification was determined to be linked to the glutamic acid moiety. Based on combined data from plasma desorption mass spectrometry, tandem mass spectrometry, accurate mass measurement and amino acid analysis of the acid hydrolysate after derivatization with o-phthaldialdehyde by high-performance liquid chromatography we could establish the structure of the modification as N-acetylputrescine. Finally, the confirmation of the linkage of the glutamic acid to diaminopimelic acid via the gamma-COOH was based on the presence of a-type peptide backbone fragment ions in the positive ion plasma desorption mass spectra of the modified N1-lactylpeptides. 相似文献

We propose an on-line learning algorithm for Multi Layered Perceptrons (MLP) with an Orthogonal Weight Estimator (OWE) architecture. Such an architecture allows to dynamically and efficiently estimate the weights of a MLP in context dependent behaviour problems. Osmotic pressure measurements were satisfactorily used to compute the effective charge of poly(acrylic) acid (PAA) at different pHs and with different monovalent and divalent counterions. This experimental method is sensitive to the osmotically active species (polymer and counterions). After correction for the polymer contribution to the total osmotic pressure (solvency and excluded volume), the remaining pressure can be attributed to the polyelectrolyte counterions and processed with Donnan and equation of state in order to compute the effective charge number per polymeric chain (Zeff). The behavior of Zeff against the chain concentration and pH was investigated after neutralization of PAA with LiOH, NaOH, and TMAOH. The results clearly indicate that the nature of the monovalent counterion has no effect on Zeff leading to the conclusion that the interaction between monovalent counterions and the acrylate functionality is purely electrostatic in agreement with conductimetric and potentiometric results reported in the literature. The behavior of Zeff against the degree of ionization of the polymer and its concentration is also in good agreement with the theoretical expectations of the theory of ionic condensation. Osmotic measurements were also used in order to understand the influence of divalent cations (Mg2+, Ca2+, Ba2+) on Zeff of the sodium salt of PAA at pH 9 and at different divalent/acrylate molar ratio. All the divalent cations depress Zeff each of one at different degrees, confirming a specific divalent/polymer interaction. The energy of hydration of cations can explain most of the observed results with divalents. 相似文献

Exposure to foreign particles sometimes causes inflammatory reactions through production of cytokines and chemoattractants by phagocytic cells. In this study, we focused on macrophage migration inhibitory factor (MIF) to evaluate its pathophysiological role in the phagocytic process. Immunohistochemical analysis of human pseudosynovial tissues retrieved at revision of total hip arthroplasty showed that infiltrating mononuclear and multinuclear cells were positively stained by both an anti-CD68 antibody and anti-human MIF antibody. For in vitro study, MIF was released from murine macrophage-like cells (RAW 264.7) in response to phagocytosis of fluorescent-latex beads in a particle dose-dependent manner. Northern blot analysis showed marked elevation of the MIF mRNA level in the phagocytic macrophage-like cells. Moreover, pretreatment of RAW 264.7 cells with rat recombinant MIF increased the extent of phagocytosis by 1.6-fold compared with the control. Taken together, these results suggest that MIF plays an important role by activating macrophages in autocrine and paracrine fashion to phagocytose foreign particles. 相似文献

Infrared and UV–vis studies of metallocene immobilization on silica are reported here. The results have indicated changes in the Zr coordination sphere of metallocene depending on the immobilization route used. The reaction of [(Cp)2ZrCl2] with silica formed [(Cp)2ZrCl]+[SiO]− species. The same metallocene, reacting with TMA modified silica, formed monomethylated and dimethylated species by the substitution of chloro for methyl ligands, stabilized on the surface by interaction with “MAO-like” species (methylaluminoxane, MAO). These monomethylated and dimethylated cationic zirconium species are the active centers for the polymerization reaction. Different order of TMA addition in the silica modification step generated surface species of a similar nature, differing in their relative quantities. The highest amount of these active species was obtained when the support was added to the TMA solution rather than adding the TMA solution to the silica support. This was the most significant parameter affecting catalytic activity in ethylene polymerization. 相似文献

The dynamic interaction, through the underlying or surrounding soil, between three-dimensional structures has been investigated, by coupling finite and boundary elements. The structures are discretized by finite elements, and the infinite soil is represented by boundary elements. A harmonic force is applied on both mass-lumped structures and mass-distributed structures. On the basis of our numerical studies, we further indicate the effects of distance, direction of alignment between two foundations, bedrock, natural frequency of the system and the location of load on the dynamic responses of structure-soil-structure interaction systems. 相似文献
