The authors report a series of experiments in which they use the masked congruence priming paradigm to investigate the processing of masked primes in the manual and verbal response modalities. In the manual response modality, they found that masked incongruent primes produced interference relative to both congruent and neutral primes. This finding, which replicates the standard finding in the masked congruence priming literature, is presumed to reflect the conflict that arises between two incompatible responses and, thus, to index the extent of processing of the masked prime. Somewhat surprisingly, when participants were asked to respond verbally in the same task, masked incongruent primes no longer produced interference, but masked congruent primes produced facilitation. These findings are surprising because they suggest that the processing of nonconsciously perceived primes extends to the response level in the manual, but not verbal, response modality. The authors propose that the modulation of the masked congruence priming effect by response modality is due to verbal, but not manual, responses being mediated by the lexical-phonological production system. A one-dimensional stationary model of biomass gasification in a fixed bed downdraft gasifier is presented in this paper. The model is based on the mass and energy conservation equations and includes the energy exchange between solid and gaseous phases, and the heat transfer by radiation from the solid particles. Different gasification sub-processes are incorporated: biomass drying, pyrolysis, oxidation of char and volatile matter, chemical reduction of H2, CO2 and H2O by char, and hydrocarbon reforming. The model was validated experimentally in a small-scale gasifier by comparing the experimental temperature fields, biomass burning rates and fuel/air equivalence ratios with predicted results. A good agreement between experimental and estimated results was achieved. The model can be used as a tool to study the influence of process parameters, such as biomass particle mean diameter, air flow velocity, gasifier geometry, composition and inlet temperature of the gasifying agent and biomass type, on the process propagation velocity (flame front velocity) and its efficiency. The maximum efficiency was obtained with the smaller particle size and lower air velocity. It was a consequence of the higher fuel/air ratio in the gasifier and so the production of a gas with a higher calorific value. 相似文献

5-Hydroxymethylfurfural (5-HMF) as a product of the Maillard reaction is found in many foods. Estimated intakes range between 4 and 30 mg per person and day, while an intake of up to 350 mg can result from, e.g., beverages made from dried plums. In vitro genotoxicity was positive when the metabolic preconditions for the formation of the reactive metabolite 5-sulphoxymethylfurfural were met. However, so far in vivo genotoxicity was negative. Results obtained in short-term model studies for 5-HMF on the induction of neoplastic changes in the intestinal tract were negative or cannot be reliably interpreted as "carcinogenic". In the only long-term carcinogenicity study in rats and mice no tumours or their precursory stages were induced by 5-HMF aside from liver adenomas in female mice, the relevance of which must be viewed as doubtful. Hence, no relevance for humans concerning carcinogenic and genotoxic effects can be derived. The remaining toxic potential is rather low. Various animal experiments reveal that no adverse effect levels are in the range of 80-100 mg/kg body weight and day. Safety margins are generally sufficient. However, 5-HMF exposure resulting from caramel colours used as food additives should be further evaluated. 相似文献

BACKGROUND: Nitrite is a key ingredient the manufacture of meat products, forming a stable pink color characteristic of cured products, retarding the development of rancidity and off‐odors and flavors during storage, and preventing microbial growth. The negative aspects of nitrite and the demands for healthy foods result in the need to reduce nitrite in cured meat products. Paprika or tomato has been employed as natural pigments in meat products. The objective of this work was to determine the effect of incorporating paprika powder or tomato paste on the texture, rancidity and instrumental and sensory color compensation in nitrite‐reduced meat batters. RESULTS: Addition of tomato paste improved moisture content, resulting in harder but less cohesive samples as compared to control and paprika‐containing meat batters. Color characteristics of reduced nitrite samples obtained higher a* red coloration (8.9 for paprika and 7.7–8.0 for tomato paste), as compared to control samples (5.65). Instrumental color was low in control samples, with high values for tomato paste and paprika samples. Nonetheless, tomato paste used to compensate color in nitrite‐reduced meat batters was ranked closer to the control sample in sensory evaluation. CONCLUSION: Color characteristics—instrumental and sensory—in these kinds of meat products were enhanced by the addition of 2.5–3.0% of tomato paste, presenting results close to the non‐reduced nitrite control. Similarly, antioxidant components of tomato paste or paprika reduced lipid oxidation. Activation of innate immunity and low-grade inflammation contributes to hyperglycemia and an onset of Type 2 Diabetes Mellitus (T2DM). Interleukin-2 (IL-2), leptin, High Mobility Group Box-1 (HMGB-1), and increased glucose concentrations are mediators of these processes also by modulating peripheral blood mononuclear cells (PBMCs) response. The aim of this study was to investigate if HMGB-1 and IL-2 turn on PBMCs and their leptin secretion. In isolated human PBMCs and their subpopulations from healthy individuals and naïve T2DM patients, leptin release, pro-inflammatory response and Toll-like Receptors (TLRs) activation was measured. After treatment with IL-2 and HMGB1, NK (Natural Killer) have the highest amount of leptin secretion, whilst NK-T have the maximal release in basal conditions. TLR4 (TAK242) and/or TLR2 (TLR2-IgA) inhibitors decreased leptin secretion after IL-2 and HMGB1 treatment. A further non-significant increase in leptin secretion was reported in PBMCs of naive T2DM patients in response to IL-2 and HMGB-1 stimulation. Finally, hyperglycemia or hyperinsulinemia might stimulate leptin secretion from PBMCs. The amount of leptin released from PBMCs after the different treatments was enough to stimulate the secretion of IL-1β from monocytes. Targeting leptin sera levels and secretion from PBMCs could represent a new therapeutic strategy to counteract metabolic diseases such as T2DM. 相似文献

Ingestion of fish parasitized with Anisakis larvae can produce infestation and/or allergy in consumers. Technological and food processing treatments have been applied to parasitized fish in order to kill the larvae and avoid the infestation; however, their influence on allergenicity has not been studied. Four lots of hake (Merluccius merluccius) steaks artificially parasitized with Anisakis larvae were subjected to two storage chilling (5 degrees C +/- 1 degrees C) and freezing (-20 degrees C +/- 1 degrees C) treatments and two food processing treatments of heat (final temperature 86.3 degrees C) and microwave (final temperature 66.9 degrees C) and studied by scanning electron microscopy, environmental scanning electron microscopy (ESEM) (acid [pH = 2] and water preparations), and emission of fluorescence. Anisakis larvae were resistant to acid conditions, remaining alive after treatment. Larvae in the heat- and microwave-treated lots presented coagulated and disrupted zones in the cuticle with release of fluids. The cylindrical shape changed to a dehydrated appearance mainly observed by ESEM. Fluorescence was only noticeable in the frozen larvae. Larvae without apparent changes, together with dehydrated ones, were observed by ESEM in the frozen lot; nevertheless, no disruptions in the cuticle were perceptible. Further studies are needed in order to elucidate if the changes observed in the cuticle reduce the resistance of the parasites to the action of gastric enzymes in the gastrointestinal tract and to determine the release of allergens to the flesh by the live larvae during chilled storage of the fish. 相似文献

Liquid array technology was used to develop a multiplexed assay for the detection of antibodies to viral nonstructural proteins (NSPs), raised in cattle in response to infection with foot-and-mouth disease (FMD) virus. Two assays, one based on recombinant NSPs and the other on synthetically produced peptides, were developed and compared side-by-side. Serum samples from serial bleeds of cattle, each experimentally infected with one of the seven serotypes (C, A, O, Asia, SAT1, SAT2, SAT3) of FMD virus were analyzed. A distinct pattern in the detection of NSP antibodies and a close correlation of the recombinant protein and peptide-based assays were observed. The detection of antibodies to NSPs is a method to differentiate FMD-infected and FMD-vaccinated animals, and a high-throughput assay would be an invaluable tool in the case of an outbreak of FMD in North America, when emergency vaccination may be utilized to spare vaccinated, noninfected animals from slaughter and subsequent disposal. 相似文献

Recent links between computer science and synthetic biology allow for construction of many kinds of algorithmic processes within cells, obtained either by a direct engineered design or by an evolutionary search. In the latter case, horizontal gene transfer and especially transfer of plasmids by conjugation is generally respected as a crucial source of genetic diversity in bacteria. While some previous studies focused on mutations as the crucial principle to obtain diversity for engineered evolution, here we consider conjugation itself as a tool to generate diversity from a pre-determined library of biocircuits basic components. The recent development of CRISPR-Cas9 and its programmable DNA cutting ability makes it a powerful selection tool able to remove nonfunctional biocircuits from a cell population. In this paper, we describe a framework for controlled bacterial evolution of biocircuits based on conjugation and on CRISPR-Cas9, resulting in a direct biological implementation of an evolutionary algorithm.
