A novel, comprehensive two-dimensional asymmetric field-flow fractionation-liquid chromatographic system is described (AsFlFFF-RPLC). The interface is based on a switching valve, and the whole sample is analyzed in both dimensions. The system proved to be repeatable and quantitative in the characterization of egg white proteins. Four peaks at 4, 5.5-6.0, 7.5-8.0, and 10.0-11.0 nm, and corresponding to lysozyme, ovalbumin, transferrin, and a dimer of transferrin, were obtained in the AsFlFFF first-dimension system. Lysozyme also produced an additional peak, which overlapped with ovalbumin. Twelve compounds were separated in the LC second-dimension system. Identifications were made with the help of standards (ovalbumin, ovotransferrin, lysozyme) and by comparison of the peak areas, particle sizes, and retention data with values given in the literature. The effect of heat on egg white denaturation was studied, and the unfolding of peptide bonds of the protein was found to be pronounced when the sample was heated in phosphate solution.  相似文献

Aerodynamic flow control effected by interactions of surface-mounted synthetic (zero net mass flux) jet actuators with a local cross flow is reviewed. These jets are formed by the advection and interactions of trains of discrete vortical structures that are formed entirely from the fluid of the embedding flow system, and thus transfer momentum to the cross flow without net mass injection across the flow boundary. Traditional approaches to active flow control have focused, to a large extent, on control of separation on stalled aerofoils by means of quasi-steady actuation within two distinct regimes that are characterized by the actuation time scales. When the characteristic actuation period is commensurate with the time scale of the inherent instabilities of the base flow, the jets can effect significant quasi-steady global modifications on spatial scales that are one to two orders of magnitude larger than the scale of the jets. However, when the actuation frequency is sufficiently high to be decoupled from global instabilities of the base flow, changes in the aerodynamic forces are attained by leveraging the generation and regulation of 'trapped' vorticity concentrations near the surface to alter its aerodynamic shape. Some examples of the utility of this approach for aerodynamic flow control of separated flows on bluff bodies and fully attached flows on lifting surfaces are also discussed.  相似文献

Excitations of disordered systems such as glasses are of fundamental and practical interest but computationally very expensive to solve. Here, we introduce a technique for modeling these excitations in an infinite disordered medium with a reasonable computational cost. The technique relies on a discrete atomic model to simulate the low‐energy behavior of an atomic lattice with molecular impurities. The interaction between different atoms is approximated using a spring‐like interaction based on the Lennard‐Jones potential, but the method can be easily adapted to other potentials. The technique allows to solve a statistically representative number of samples with low computational expense and uses a Monte Carlo approach to achieve a state corresponding to any given temperature. Adsorption characteristics of three clay adsorbents – sepiolite, kaolin and synthetic talc – were investigated to determine their applicability to remove an anionic reactive dye – Reactive Yellow 138:1 – from a water stream. Results showed that synthetic talc had the highest maximum monolayer adsorption capacity of 10.07 mg/g, followed by kaolin (3.73 mg/g) and sepiolite (3.23 mg/g), for the removal of Reactive Yellow 138:1. Synthetic talc and sepiolite removed dyes from aqueous solutions mainly through physical adsorption depending on the surface area. By contrast, the adsorption mechanism of kaolin was strongly influenced by the anion exchange mechanism. Zeta potential analysis revealed that the adsorption capacity of clay adsorbents increased under acidic rather than alkaline conditions, owing to protonation of the clay surface. Moreover, synthetic talc was successfully regenerated by heating at more than 400°C, effectively restoring its original adsorption capacity.  相似文献

Magnesium is important for both bone growth and cartilage formation. However, the postoperative intake of antibiotics such as quinolones may cause a reduction in magnesium levels in tissue. The addition of magnesium to scaffolds may therefore be beneficial for the regeneration of osteochondral defects. In this study, porous composite scaffolds were produced by gas foaming of poly(d ,l ‐lactide‐co‐glycolide) (PLGA) rods with magnesium‐containing bioresorbable glasses and magnesium hydroxide as fillers. The in vitro hydrolytical degradation of the composite scaffolds in Tris buffer was followed over a 10‐week period. Mg2+ was released in a controlled manner from the scaffolds with varying release profiles between the different materials. Higher glass content resulted in a reduced mass loss compared to scaffolds with lower glass content. As a result of the foaming method, the scaffolds shrank initially, without evidence that the addition of hydrophilic fillers would decrease the initial shrinkage. © 2015 Wiley Periodicals, Inc. J. Synthetic macromolecules that can bind and co-assemble with proteins are important for the future development of biohybrid materials. Active systems are further required to create materials that can respond and change their behavior in response to external stimuli. Here we report that stimuli-responsive linear-branched diblock copolymers consisting of a cationic multivalent dendron with a linear thermoresponsive polymer tail at the focal point, can bind and complex Pyrococcus furiosus ferritin protein cages into crystalline arrays. The multivalent dendron structure utilizes cationic spermine units to bind electrostatically on the surface of the negatively charged ferritin cage and the in situ polymerized poly(di(ethylene glycol) methyl ether methacrylate) linear block enables control with temperature. Cloud point of the final product was determined with dynamic light scattering (DLS), and it was shown to be approximately 31 °C at a concentration of 150 mg/L. Complexation of the polymer binder and apoferritin was studied with DLS, small-angle X-ray scattering, and transmission electron microscopy, which showed the presence of crystalline arrays of ferritin cages with a face-centered cubic (fcc Fm3¯m) Bravais lattice where lattice parameter a = 18.6 nm. The complexation process was not temperature dependent but the final complexes had thermoresponsive characteristics with negative thermal expansion.  相似文献

 In order to determine the best extraction procedure for volatile oil composition studies in wormwood (Artemisia absinthium L.), several extraction techniques have been investigated. In pentane extracts obtained using microwave or ultrasound sources of radiation, no thermic degradations occurred, but high amounts of heavy oils and waxes resulted, causing trouble in the gas chromatographic analysis. Headspace extraction provided the most volatile constituents, but high molecular weight compounds were not extracted. Simultaneous distillation-extraction resulted in the most adequate extraction procedure for volatile oil composition studies since the required compounds were extracted with minimum degradations when the distillation time was no longer than 30 min. Oncolytic virotherapy is a rapidly progressing field that uses oncolytic viruses (OVs) to selectively infect malignant cells and cause an antitumor response through direct oncolysis and stimulation of the immune system. Despite demonstrated pre-clinical efficacy of OVs in many cancer types and some favorable clinical results in glioblastoma (GBM) trials, durable increases in overall survival have remained elusive. Recent evidence has emerged that tumor-associated macrophage/microglia (TAM) involvement is likely an important factor contributing to OV treatment failure. It is prudent to note that the relationship between TAMs and OV therapy failures is complex. Canonically activated TAMs (i.e., M1) drive an antitumor response while also inhibiting OV replication and spread. Meanwhile, M2 activated TAMs facilitate an immunosuppressive microenvironment thereby indirectly promoting tumor growth. In this focused review, we discuss the complicated interplay between TAMs and OV therapies in GBM. We review past studies that aimed to maximize effectiveness through immune system modulation—both immunostimulatory and immunosuppressant—and suggest future directions to maximize OV efficacy.  相似文献

The same origin ray set (SORS) is a computational primitive which can be used by ray tracing, radiosity and multiple pass illumination simulation algorithms for realistic image synthesis. A SORS consists of a set of rays emanating from the same point in space. The SORS query computes the first object intersected by each ray and the intersection point. In this paper we present an efficient projection algorithm for computing a SORS query for polygonal scenes. The algorithm achieves its efficiency by separating ray-polygon intersection detection from the computation of the intersection point between the ray and the polygon's plane. The algorithm can be integrated with all current illumination acceleration schemes. We analyse the projection algorithm and compare it to the alternative of computing the SORS query one ray at a time. The analysis' results are expressed in terms of a few intuitive parameters, measuring the success of the acceleration scheme in culling irrelevant objects and the concentration of the ray set. The projection algorithm can be up to five times more efficient, depending on these parameters and the quality of the image. The relative advantage of the projection increases with image quality.  相似文献
