To facilitate efficient drug delivery to tumor tissue, several nanomaterials have been designed, with combined diagnostic and therapeutic properties. In this work, we carried out fundamental in vitro and in vivo experiments to assess the labeling efficacy of our novel theranostic nanoprobe, consisting of glycogen conjugated with a red fluorescent probe and gadolinium. Microscopy and resazurin viability assays were used to study cell labeling and cell viability in human metastatic melanoma cell lines. Fluorescence lifetime correlation spectroscopy (FLCS) was done to investigate nanoprobe stability. Magnetic resonance imaging (MRI) was performed to study T1 relaxivity in vitro, and contrast enhancement in a subcutaneous in vivo tumor model. Efficient cell labeling was demonstrated, while cell viability, cell migration, and cell growth was not affected. FLCS showed that the nanoprobe did not degrade in blood plasma. MRI demonstrated that down to 750 cells/μL of labeled cells in agar phantoms could be detected. In vivo MRI showed that contrast enhancement in tumors was comparable between Omniscan contrast agent and the nanoprobe. In conclusion, we demonstrate for the first time that a non-toxic glycogen-based nanoprobe may effectively visualize tumor cells and tissue, and, in future experiments, we will investigate its therapeutic potential by conjugating therapeutic compounds to the nanoprobe.  相似文献

MS2 bacteriophages removal from surface water, characterized by high natural organic matter (NOM) content, was investigated by inline coagulation/flocculation pretreatment followed by ceramic microfiltration (MF). MS2 and DOC removal increased with lower pH and higher coagulant dose. Lowering the coagulant pH from 6.5 to 5.5 for polyaluminum chloride (PACl), and to 5.0 for iron chloride (FeCl), respectively, along with doubling of the coagulant dose from 2 to 4 mg Al/L, and from 4 to 8 mg Fe/L, respectively, maximized the virus removal, resulting in more than six log unit reductions up to complete virus retention. However, high residual metal concentrations were found under such conditions. Comparison of conventional two‐stage coagulation pretreatment with simple inline coagulation did not show any significant performance differences. Both investigated coagulants showed virus inactivation about two log units after 60 min contact time, which is equivalent to a virus inactivation of 99%. This inactivation was only reversible to a small extend by chemical or physical floc destruction. The detectability of wavelength-modulation (WM) diode-laser spectrometric techniques is frequently limited by various background signals. A new theoretical formalism for WM spectrometry, based on Fourier analysis and therefore capable of handling a variety of phenomena including the characterization and the analysis of analytical as well as background WM signals, was recently presented [Appl. Opt. 38, 5803 (1999)]. We report a detailed characterization of WM background signals from multiple reflections between pairs of surfaces in the optical system that act as etalons and from the associated intensity modulation in terms of this new formalism. The agreement between the background signals from a thin glass plate and those predicted by the formalism is good, which verifies the new Fourier analysis-based formalism.  相似文献

A case–control study was conducted on 204 drivers fatally injured in road traffic accidents in south-eastern Norway during the period 2003–2008. Cases from single vehicle accidents (N = 68) were assessed separately. As controls, 10 540 drivers selected in a roadside survey in the same geographical area during 2005–2006 were used. Blood samples were collected from the cases and oral fluid (saliva) samples from the controls. Samples were analysed for alcohol, amphetamines, cannabis, cocaine, opioid analgesics, hypnotics, sedatives and a muscle relaxant; altogether 22 psychoactive substances. Equivalent cutoff concentrations for blood and oral fluid were used. The risk for fatal injury in a road traffic accident was estimated using logistic regression adjusting for gender, age, season of the year, and time of the week. The odds for involvement in fatal road traffic accidents for different substances or combination of substances were in increasing order: single drug < multiple drugs < alcohol only < alcohol + drugs. For single substance use: medicinal drug or THC < amphetamine/methamphetamine < alcohol. For most substances, higher ORs were found when studying drivers involved in single vehicle accidents than for those involved in multiple vehicle accidents, but confidence intervals were wider.  相似文献

This study investigated the relationship between premorbid and current cognitive function with respect to the clinical features of patients with various types of neurodegeneration in the form of Alzheimer's disease (AD), mild cognitive impairment (MCI), and subjective cognitive impairment (SCI), as compared with a healthy control group (C). Clinical features (MMSE, cognitive and depressive symptoms), genetics (apolipoprotein E; APOE) and measures of neurodegeneration (Aβ42, t-tau, and p-tau) were examined, as well as present cognitive function. Various methods of assessing premorbid cognitive function were compared, including a Swedish NART-analogous test (Irregularly Spelled Words; ISW), a Swedish lexical decision test (SLDT), a Hold test (Information in WAIS-R), Best current performance test, and combined demographic characteristics. Results showed that cognitive decline (premorbid minus current cognitive function) based on SLDT and ISW was a significant predictor for MMSE and Aβ42, whereas corresponding associations for present cognitive function and decline measures based on other methods were less powerful. Results also showed that specific verbal abilities (e.g., SLDT and ISW) were insensitive to AD and that these abilities indicated premorbid cognitive function in retrospect. Free‐radical suspension polymerization was used to synthesize thermally expandable microspheres (TEMS); in this process, a poly(acrylonitrile‐co‐methacrylonitrile) shell encapsulated isooctane. Different amounts of dimethacrylate, diacrylate, or divinyl ether functional crosslinker were added to investigate the effects on the crosslinking density of the polymer and the expansion properties of the TEMS. The optimum amount of crosslinker was found to be approximately 0.05–0.1 mol %. However, a significantly better expansion could be obtained with 1,4‐butanediol dimethacrylate as a crosslinker, compared to 1,4‐butanediol divinyl ether or 1,4‐butanediol diacrylate. From monitoring the conversion of monofunctional analogues by gas chromatography, we suggest that the differences in expansion obtained with different crosslinkers, originated from the difference in the reactivity of the radicals in the system toward the vinyl functionalities of the crosslinkers. This regulated the incorporation of the crosslinker into the polymer and, thereby, the mechanical properties of the microsphere shell. © 2010 Wiley Periodicals, Inc. J Appl Polym Sci, 2010  相似文献

The properties of full meet contraction are investigated, andit is shown to be a useful building-block in the constructionof composite contraction operators. This is followed by a detailedinvestigation of specified meet contraction, i.e. the operation÷ such that K ÷ p = K f(p), where is full meetcontraction and f is a function from sentences to sentences.A number of realistic examples of belief contraction are discussed,and it is shown how the properties of contraction differ accordingto how the sentence to be contracted is situated in the structureof the belief state.
