The identification of foodborne microorganisms and their endospores in food products are important for food safety. The present work compares Bacillus (Bacillus licheniformis, Bacillus circulans and Bacillus subtilis) and Micrococcus (Micrococcus luteus) species with Fourier transform infrared (FTIR) spectroscopy. Our results show that there are several characteristic peaks belonging to both the Micrococcus and Bacillus species which can be used for the identification of these foodborne bacteria and their endospores. For Micrococcus species, a new band was observed at 1338 cm−1 which may be due to acetate oxidation via the carboxylic acid cycle. The bands at 1313 cm−1 and 1256 cm−1 can be explained by an exopolymer formation and the other bands at 1074 cm−1 and 550 cm−1, may be due to the glycogen-like storage material in Micrococcus spp. There are also characteristic peaks at 993 cm−1 and 801 cm−1 for these bacterial species. Different Bacillus species also showed characteristic peaks at 1000–500 cm−1 region. Dipicolinic acid (DPA) bands at ∼728 cm−1 and ∼703 cm−1 seen only in B. circulans were the marker of an endospore formation.  相似文献

Four different types of mould‐ripened Civil cheese were manufactured. A defined (nontoxigenic) strain of a Penicillium roqueforti (SC 509) was used as the secondary starter with and without addition of the whey cheese (Lor); in parallel, secondary starter‐free counterparts were manufactured. Chemical composition, microbiology and proteolysis were studied during the ripening. The incorporation of whey cheese in the manufacture of mould‐ripened Civil cheese altered the gross composition and adversely affected proteolysis in the cheeses. The inoculated P. roqueforti moulds appeared to grow slowly on those cheeses, and little proteolysis was evident in all cheese treatments during the first 90 days of ripening. However, sharp increases in the soluble nitrogen fractions were observed in all cheeses after 90 days. Microbiological analysis showed that the microbial counts in the cheeses were at high levels at the beginning of ripening, while their counts decreased approximately 1–2 log cfu/g towards the end of ripening.  相似文献

Ion-exchange experiments were performed to evaluate the formation of the uranium-citrate and uranium-iron-citrate complexes over a wide concentration range; i.e., environmentally relevant concentrations (e.g., 10(-6) M in metal and ligand) and concentrations useful for spectroscopic investigations (e.g., 10(-4) M in metal and ligand). The stability of the well-known uranium-citrate complex was determined to validate the computational and experimental methods applied to the more complex system. Values of the conditional stability constants for these species were obtained using a chemical equilibrium model in FITEQL. At a pH of 4.0, the stability constant for uranium-citrate complex (log beta1,1) was determined to be 8.71+/-0.6 at I = 0. Analysis of the results of ion-exchange experiments for the U-Fe-citric acid system indicates the formation of the 1:1:1 and 1:1:2 ternary species with stability constants (log beta) of 17.10+/-0.41 and 20.47+/-0.31, respectively, at I= 0.  相似文献

The objective of this study is to characterise the gross chemical and microbiological status and identify the volatile compounds of mouldy Civil cheeses. A total of forty‐one samples were surveyed, and gross compositional status of the cheeses was (as mean values): 6.5 for pH, 6.2% for fat‐in‐dry matter, 51.8% for moisture and 15.3% for water‐soluble nitrogen (as% of total nitrogen). Chemical composition of the cheese samples varied widely. Mouldy Civil cheese has similar pH values and moisture contents when compared with blue‐type cheeses, but it has distinct feature for fat contents. The microbiological counts of the samples were found to be high and some samples contained coliform bacteria. A total of 95 volatiles, including esters (28), acids (6), ketones (12), aldehydes (3), alcohols (15), terpenes (10), sulphur compounds (3) and miscellaneous (18), were identified in the volatile fractions of the cheeses, and principal volatile groups were esters, alcohols and ketones.  相似文献

The excitation energy-dependent nature of Raman scattering spectrum, vibration, electronic or both, has been studied using different excitation sources on as-grown and annealed n- and p-type modulation-doped Ga1 − xInxNyAs1 − y/GaAs quantum well structures. The samples were grown by molecular beam technique with different N concentrations (y = 0%, 0.9%, 1.2%, 1.7%) at the same In concentration of 32%. Micro-Raman measurements have been carried out using 532 and 758 nm lines of diode lasers, and the 1064 nm line of the Nd-YAG laser has been used for Fourier transform-Raman scattering measurements. Raman scattering measurements with different excitation sources have revealed that the excitation energy is the decisive mechanism on the nature of the Raman scattering spectrum. When the excitation energy is close to the electronic band gap energy of any constituent semiconductor materials in the sample, electronic transition dominates the spectrum, leading to a very broad peak. In the condition that the excitation energy is much higher than the band gap energy, only vibrational modes contribute to the Raman scattering spectrum of the samples. Line shapes of the Raman scattering spectrum with the 785 and 1064 nm lines of lasers have been observed to be very broad peaks, whose absolute peak energy values are in good agreement with the ones obtained from photoluminescence measurements. On the other hand, Raman scattering spectrum with the 532 nm line has exhibited only vibrational modes. As a complementary tool of Raman scattering measurements with the excitation source of 532 nm, which shows weak vibrational transitions, attenuated total reflectance infrared spectroscopy has been also carried out. The results exhibited that the nature of the Raman scattering spectrum is strongly excitation energy-dependent, and with suitable excitation energy, electronic and/or vibrational transitions can be investigated.  相似文献

A multistage polymerization protocol, the so‐called “modified seeded polymerization,” was developed for the production of monodisperse‐porous poly(styrene‐co‐divinylbenzene) providing high column efficiency as a packing material in reversed phase high performance liquid chromatography (RPLC). In the first stage of the multistage production, uniform polystyrene seed particles, produced by dispersion polymerization, were swollen by an organic agent (i.e., the diluent) and then by a monomer mixture containing styrene and divinylbenzene. The final porous particles were obtained in the monodisperse form by the polymerization of monomer mixture in the seed particles. By the use of a small size seed latex with low molecular weight and by the selection of the appropriate diluent, relatively small monodisperse‐porous particles with suitable pore structure could be achieved. In the reversed phase separation of alkylbenzenes, under isocratic conditions, theoretical plate numbers up to 40,000 plates/m were achieved by using 5.2 μm porous particles, obtained by a toluene‐dibutyl phthalate mixture as the diluent. No significant decrease in the resolution power was observed by the fourfold increase in the mobile phase flow rate. The column efficiency and the resolution observed with 5.2 μm monodisperse‐porous particles were significantly higher with respect to the currently available polymer based packing materials used in the reversed phase HPLC. © 2005 Wiley Periodicals, Inc. There is currently considerable interest in developing stiff, strong, tough, and heat resistant poly(lactide) (PLA) based materials with improved melt elasticity in response to the increasing demand for sustainable plastics. However, simultaneous optimization of stiffness, strength, and toughness is a challenge for any material, and commercial PLA is well-known to be inherently brittle and temperature-sensitive and to show poor melt elasticity. In this study, we report that high-shear mixing with cellulose nanocrystals (CNC) leads to significant improvements in the toughness, heat resistance, and melt elasticity of PLA while further enhancing its already outstanding room temperature stiffness and strength. This is evidenced by (i) one-fold increase in the elastic modulus (6.48 GPa), (ii) 43% increase in the tensile strength (87.1 MPa), (iii) one-fold increase in the strain at break (∼6%), (iv) two-fold increase in the impact strength (44.2 kJ/m2), (v) 113-fold increase in the storage modulus at 90°C (787.8 MPa), and (vi) 103-fold increase in the melt elasticity at 190°C and 1 rad/s (∼105 Pa) via the addition of 30 wt% CNC. It is hence possible to produce industrially viable, stiff, strong, tough, and heat resistant green materials with improved melt elasticity through high-shear mixing.  相似文献

Over the past 30 years, numerical methods and simulation tools for incompressible flows have been advanced as a subset of the computational fluid dynamics (CFD) discipline. Although incompressible flows are encountered in many areas of engineering, the simulation of compressible flows has driven most of the development of computational algorithms and tools at NASA Ames Research Center. This is due to the stringent requirements for predicting aerodynamic performances of flight vehicles. Conversely, low-speed incompressible flow through or past flow devices did not require the same numerical accuracy. This practice of tolerating relatively low-fidelity solutions in engineering applications has changed, as the design of low-speed flow devices have become more sophisticated, along with more strict efficiency requirements. Accurate and robust CFD tools have become increasingly important in fluid engineering for incompressible and low-speed flow. This paper reviews advances in computational technologies for incompressible flow simulation developed at Ames, and some engineering successes brought about by these advances made during the same period. Additionally, some of the current challenges faced in computing incompressible flows are presented.  相似文献

Induced-charge electro-osmotic (ICEO) flow of polymer-containing electrolyte solution around a cylindrical gold-coated stainless steel rod under AC electric field is measured by micro-particle image velocimetry (micro-PIV) for the first time. The ICEO flows as functions of the amount of non-ionic PEG (polyethylene glycol), cationic PDADMA (polydiallyldimethylammonium chloride), and anionic PVSASS (polyvinylsulfonic acid sodium salt) polymers added into the salt solution, frequency, and strength of the AC electric field are measured. The ICEO flow of polymer-containing fluid around the rod is quadrupolar with four vortices and is proportional to the square of imposed electric field. The ICEO flow velocity exponentially decreases with an increased concentration of neutral PEG. Ionic polyelectrolytes significantly increase ICEO velocities due to the enriched net charge within the induced electric double layer arising from the electrostatic interaction between the polarized rod’s surface and the charged polyelectrolytes in ionic polymer solution. In addition, polymer concentration affects the dependence of the ICEO flow on the frequency of AC electric field.  相似文献
