Microscopic compression tests (micropillar tests) are typically used to obtain stiffness and strength properties of materials at small length scales. In this work it is shown that structural effects, in particular instabilities, have implications on the resulting load–displacement diagram. Care has to be taken when the measured load–displacement path of a micropillar is interpreted as a stress–strain path of the material. In the era of personalized medicine, insights into the molecular mechanisms that differentially contribute to disease phenotypes, such as asthma phenotypes including obesity-associated asthma, are urgently needed. Peripheral blood was drawn from 10 obese, non-atopic asthmatic adults with a high body mass index (BMI; 36.67 ± 6.90); 10 non-obese, non-atopic asthmatic adults with normal BMI (23.88 ± 2.73); and 10 healthy controls with normal BMI (23.62 ± 3.74). All asthmatic patients were considered to represent a low type-2 asthma phenotype according to selective clinical parameters. RNA sequencing (RNA-Seq) was conducted on peripheral blood CD4+ T cells. Thousands of differentially expressed genes were identified in both asthma groups compared with heathy controls. The expression of interferon (IFN)-stimulated genes associated with IFN-related signaling pathways was specifically affected in obese asthmatics, while the gap junction and G protein-coupled receptor (GPCR) ligand binding pathways were enriched in both asthma groups. Furthermore, obesity gene markers were also upregulated in CD4+ T cells from obese asthmatics compared with the two other groups. Additionally, the enriched genes of the three abovementioned pathways showed a unique correlation pattern with various laboratory and clinical parameters. The specific activation of IFN-related signaling and viral infection pathways might provide a novel view of the molecular mechanisms associated with the development of the low type-2 obesity-associated asthma phenotype, which is a step ahead in the development of new stratified therapeutic approaches. 相似文献

Osteo-inductive materials give rise to ectopic bone formation in vivo either in muscles or in subcutaneous tissue. However, the underlying molecular mechanism is totally unclear. To investigate the expression pattern of bone related genes in osteo-inductive materials, we performed quantitative PCR (qPCR) to detect the expressions of type I collagen, alkaline phosphatase (ALP) and bone morphogenetic protein 4 (BMP-4) in biphasic calcium phosphate (BCP) ceramics implanted in dorsal muscle of dogs. Bone formation in mandibular alveolus defects served as controls showing the expression patterns of these genes in natural healing bones. Histological examinations were performed to show the bone formation in osteo-inductive BCP implants. Data of qPCR indicated that all tested genes had a similar expression pattern with two peaks during the bone formation either in BCP implants or natural healing bones. Type I collagen and ALP were expressed at lower levels with delayed peak in BCP implant than that in natural healing bone. Higher BMP-4 expression level was detected in BCP ceramic implant than in natural healing bone at all the time points. These results demonstrated that expression patterns of bone-related genes in the inductive bone formation are similar to that of natural healing bone formation. As these three genes are important parameters for osteoblast activity in bone formation, our data provide clue to uncover the molecular mechanism of bone formation in osteo-inductive materials. 相似文献

Sewage pumping stations represent an element of the sewer system, which is directly responsible for affecting serviceability; i.e., failing pumps may result in combined sewer overflows or flooding. However, failures of sewage pumps are not yet incorporated in sewer assessments due to lack of data. This paper presents the analysis of pump failure data provided by two sewer management authorities in The Netherlands. Pump failures have been studied accounting for the nature of the failures, the operation and maintenance procedures of the management authority, the aging of the pumps, and the changes in the environment of pumps. The analysis shows that sewage pumps fail relatively often due to the composition of sewage and the discontinuous operation of the pumps. The interarrival time and the duration of failures are highly variable and independent of the specific function of the pump. Resulting pump failure characteristics are applied in a Monte Carlo simulation to calculate the impact of failures on combined sewer overflow volumes. The results indicate that the serviceability of sewer systems is significantly affected by failing pumps. Therefore, including pump availability in sewer system assessments should be considered. 相似文献

When administered intracerebroventricularly to mice performing various learning tasks involving either short-term or long-term memory, secreted forms of the beta-amyloid precursor protein (APPs751 and APPs695) have potent memory-enhancing effects and block learning deficits induced by scopolamine. The memory-enhancing effects of APPs were observed over a wide range of extremely low doses (0.05-5,000 pg intracerebroventricularly), blocked by anti-APPs antisera, and observed when APPs was administered either after the first training session in a visual discrimination or a lever-press learning task or before the acquisition trial in an object recognition task. APPs had no effect on motor performance or exploratory activity. APPs695 and APPs751 were equally effective in the object recognition task, suggesting that the memory-enhancing effect of APPs does not require the Kunitz protease inhibitor domain. The preparation and evaluation of peat/No. 2 fuel oil mixtures (POM) and peat/methanol mixtures (PAM) is described. POM and PAM prepared using North Carolina peat and having varied peat loadings, peat moisture contents, and peat particle sizes have been studied by measuring slurry sedimentation ratios and drain times from sedimentation tubes. The peat moisture content was particularly crucial in forming stable slurries. The effect of a variety of additives at 0.5–1.0 wt.% on sedimentation ratios, drain times, and viscosities was studied. Calorimetric studies of several PAM and POM slurries as well as preliminary combustion tests of POM slurries in a salamander burner are also reported. 相似文献

The aim of this study was to develop matrix-type transdermal systems (TDSs) containing the highly lipophilic (log P = 5.82) antiestrogen (AE) and the permeation enhancers propylene glycol and lauric acid. For that purpose, permeation of AE from various adhesive matrices through excised skin of hairless mice was evaluated. It was found that pretreatment of the skin with permeation enhancers raised the transdermal flux of subsequently applied antiestrogen. Highest steady-state transdermal fluxes (1.1 µg cm-2 h-1) were obtained from Gelva®, polyacrylate adhesive, followed by 0.55 µg cm-2 h-1 from Oppanol® polyisobutylene, 0.31 µg cm-2 h-1 from BIO-PSA® silicone, and 0.12 µg cm-2 h-1 from Sekisui polyacrylate matrices. In order to develop TDS with high content of fluid permeation enhancer propylene glycol, two different strategies were investigated. One strategy was the addition of hydroxypropyl cellulose (HPC) as thickening agent to Gelva matrices. This allowed for propylene glycol loading levels of up to 30%, resulting in transdermal AE fluxes of 0.09 µg cm-2 h-1. On the other hand, a fleece-laminated backing foil was loaded with the described permeation enhancer formulation and laminated with polyacrylate adhesive layer, resulting in transdermal AE fluxes of 0.06 µg cm-2 h-1. However, application of these TDSs on skin pretreated with permeation enhancers raised the fluxes to 2.6 µg cm-2 h-1 from Gelva/HPC and 0.46 µg cm-2 h-1 from fleece/Sekisui. 相似文献
