Pooled serum samples from 3802 Australian residents were analyzed for four perfluoroalkylsulfonates, seven perfluoroalkylcarboxylates, and perfluorooctanesulfonamide (PFOSA). Serum was collected from men and women of five different age groups and from rural and urban regions in Australia. The highest mean concentration was obtained for perfluorooctane sulfonate (PFOS, 20.8 ng/mL) followed by perfluorooctanoic acid (PFOA, 7.6 ng/mL), perfluorohexane sulfonate (PFHxS, 6.2 ng/mL), perfluorononanoic acid (PFNA, 1.1 ng/mL), and PFOSA (0.71 ng/mL). Additional four PFCs were detected in 5-18% of the samples at concentrations near the detection limits (0.1-0.5 ng/mL). An increase in PFOS concentration with increasing age in both regions and genders was observed. The male pool levels of some of the age groups compared to females were higherfor PFOS, PFOA, and PFHxS. In contrast, PFNA concentrations were higher in the female pools. No substantial difference was found in levels of PFCs between the urban and rural regions. The levels are equal or higher than previously reported serum levels in Europe and Asia but lower compared to the U.S.A. These results suggest that emissions from production in the Northern Hemisphere are of less importance for human exposure.  相似文献

Different particulate matter (PM) samples were investigated by on-line single-particle aerosol time-of-flight mass spectrometry (ATOFMS). The samples consist of soot particulates made by a diffusion flame soot generator (combustion aerosol standard, CAST), industrially produced soot material (printex), soot from a diesel passenger car as well as ambient particulates (urban dust (NIST) and road tunnel dust). Five different CAST soot particle samples were generated with different elemental carbon (EC) and organic carbon (OC) content. The samples were reaerosolized and on-line analyzed by ATOFMS, as well as precipitated on quartz filters for conventional EC/OC analysis. For each sample ca. 1000 ATOFMS single-particle mass spectra were recorded and averaged. A typical averaged soot ATOFMS mass spectrum shows characteristic carbon cluster peak progressions (Cn+) as well as hydrogen-poor carbon cluster peaks (CnH(1-3)+). These peaks are originated predominately from the elemental carbon (EC) content of the particles. Often additional peaks, which are not due to carbon clusters, are observed, which either are originated from organic compounds (OC-organic carbon), or from the non-carbonaceous inorganic content of the particles. By classification of the mass spectral peaks as elemental carbon (i.e., the carbon cluster progression peaks) or as peaks originated from organic compounds (i.e., molecular and fragment ions), the relative abundance of elemental (EC) and organic carbon (OC) can be determined. The dimensionless TC/EC values, i.e., the ratio of total carbon content (TC, TC = OC + EC) to elemental carbon (EC), were derived from the ATOFMS single-particle aerosol mass spectrometry data. The EC/TC values measured by ATOFMS were compared with the TC/EC values determined by the thermal standard techniques (thermooptical and thermocoulometric method). A good agreement between the EC/TC values obtained by on-line ATOFMS and the offline standard method was found.  相似文献

Zusammenfassung. Lebensmittel, die in den Verkehr gebracht werden, müssen sicher sein. Dieses Ziel ist in der Verordnung (EG) 178/2002 formuliert. Als nicht sicher gelten unter anderem Lebensmittel, die negative Auswirkungen auf die Gesundheit des Verbrauchers besitzen. Im Mittelpunkt des Interesses stehen damit die Auswirkungen der Lebensmittel und somit die Gesamtheit ihrer Inhaltsstoffe auf die Gesundheit des Menschen. Die Entwicklung von Methoden für die Lebensmittelkontrolle ist zur Zeit vor allem von dem Ziel gepr?gt, bestimmte ausgew?hlte Stoffe, für die in der Regel gesetzlich festgelegte Grenzwerte bzw. H?chstmengen existieren, in immer geringeren Konzentrationen nachzuweisen. W?hrend etwa 80.000 Chemikalien derzeit auf dem EU-Markt erh?ltlich sind und ein Vielfaches an Verbindungen als Neben- und Umbauprodukte in die Umwelt und damit potentiell in die Nahrungskette gelangen k?nnen, wird in der Lebensmittelüberwachung nur ein Bruchteil derartiger Verbindungen analysiert. Substanzen, die in diesem Spektrum nicht enthalten sind, bleiben – unabh?ngig von ihrem Gef?hrdungspotential – au?er Acht. In vorliegendem Artikel werden das Prinzip und die Einsatzbereiche der wirkungsbezogenen Analytik in der Lebensmittelkontrolle dargestellt. Dabei flie?en Erfahrungen aus der ?kotoxikologie ein, in der dieser Untersuchungsansatz bereits seit Jahren eine Rolle spielt. Es werden zwei sich erg?nzende Teilstrategien beschrieben, die beide eine deutliche Verbesserung des vorsorgenden Verbraucherschutzes bedeuten k?nnen und sinnvoll verknüpft werden müssen. Die erste Strategie setzt auf das Screening potentiell sch?dlicher Effekte der Gesamtextrakte durch biologische Wirkungstests; die zweite Strategie geht über die Detektion von Wirkungen hinaus und versucht die stofflichen Ursachen der gemessenen Effekte zu identifizieren. Im Sinne eines umfassenden Verbraucherschutzes – entsprechend der Verordnung (EG) Nr. 178/2002 zur Lebensmittelsicherheit – sollte vom Gesetzgeber angestrebt werden, zus?tzlich zu Grenzwerten für einzelne Kontaminanten auch Grenzwerte für bestimmte Effekte zu etablieren. Lactic acid bacteria (LAB) isolated from marinated or non-marinated, modified atmosphere packaged (MAP) broiler leg products and air samples of a large-scale broiler meat processing plant were identified and analyzed for their phenotypic properties. Previously, these strains had been found to be coccal LAB. However, the use of a 16 and 23S rRNA gene RFLP database had not resulted in species identification because none of the typically meat-associated LAB type strains had clustered together with these strains in the numerical analysis of the RFLP patterns. To establish the taxonomic position of these isolates, 16S rRNA gene sequence analysis, numerical analysis of ribopatterns, and DNA-DNA hybridization experiments were done. The 16S rRNA gene sequences of three isolates possessed the highest similarities (over 99%) with the sequence of S. parauberis type strain. However, in the numerical analysis of HindIII ribopatterns, the type strain did not cluster together with these isolates. Reassociation values between S. parauberis type or reference strain and the strains studied varied from 82 to 97%, confirming that these strains belong to S. parauberis. Unexpectedly, most of the broiler meat-originating strains studied for their phenotypical properties did not utilize lactose at all and the same strains fermented also galactose very weakly, properties considered atypical for S. parauberis. This is, to our knowledge, the first report of lactose negative S. parauberis strains and also the first report associating S. parauberis with broiler slaughter and meat products.  相似文献

We propose a multimedia model-based methodology to evaluate whether a chemical substance qualifies as POP-like based on overall persistence (Pov) and potential for long-range transport (LRTP). It relies upon screening chemicals against the Pov and LRTP characteristics of selected reference chemicals with well-established environmental fates. Results indicate that chemicals of high and low concern in terms of persistence and long-range transport can be consistently identified by eight contemporary multimedia models using the proposed methodology. Model results for three hypothetical chemicals illustrate that the model-based classification of chemicals according to Pov and LRTP is not always consistent with the single-media half-life approach proposed by the UNEP Stockholm Convention and thatthe models provide additional insight into the likely long-term hazards associated with chemicals in the environment. We suggest this model-based classification method be adopted as a complement to screening against defined half-life criteria at the initial stages of tiered assessments designed to identify POP-like chemicals and to prioritize further environmental fate studies for new and existing chemicals.  相似文献

In vitro short-term storage of boar semen for up to 72 h before insemination negatively affects fertility, but this often remains undetected during semen quality assessment. One important sperm function is the ability to form the functional sperm reservoir in the oviduct. In the present study, we used the modified oviductal explant assay to study sperm binding to oviductal epithelium in vitro in diluted boar semen stored for 24 or 72 h. First, we determined the kinetics of in vitro sperm binding to oviductal epithelium in relation to co-incubation time of sperm and oviductal tissue pieces. Then, we studied how the binding of sperm to oviductal epithelium was affected by in vitro semen storage and by differences among individual boars. Sperm binding after different incubation times was significantly higher when semen was stored 24 h than after 72-h storage (P < 0.05), and peaked at 30-90 min of incubation. Sperm binding differed between boars (n = 44), and was negatively correlated to the percentage of sperm with cytoplasmic droplets (R = -0.51, P < 0.001). There were no significant changes in motility, acrosome integrity and propidium iodide stainability during the 72-h storage period. However, sperm-binding indices were significantly lower after 72 h in vitro storage than after 24-h storage in sperm from boars with normal semen quality (P < 0.05); in contrast, the binding capacity of sperm from boars with higher percentages of morphologically altered sperm remained at a low level. The sperm-binding capacity of sperm from four of the five boars with known subfertility was lower than the mean binding index minus one standard deviation of the boar population studied here. It is concluded that changes in the plasma membrane associated with in vitro ageing reduce the ability of stored boar sperm to bind to the oviductal epithelium. This study shows the potential of sperm-oviduct binding as a tool to assess both male fertility and changes in sperm function associated with in vitro ageing.  相似文献

Chemical speciation determines Se solubility and therefore its bioavailability and potential for transport in the environment. In this study we investigated the speciation of Se in soil developed on reclaimed mine sites in the U.S. Western Phosphate Resource Area (WPRA) using micro-X-ray absorption near-edge structure (micro-XANES) spectroscopy and micro-X-ray fluorescence (micro-XRF) mapping. Selenium was nonuniformly distributed in the soils and positively correlated with Fe, Mn, Cu, Zn, and Ni. Sixteen points of interest (POI) from three soil samples were analyzed with micro-XANES spectroscopy. The XANES data indicated that Se is present in the soils in at least three oxidation states, Se(-II, 0), Se(IV), and Se(VI). Selenides or elemental Se dominated 7 of the 16 POI. Selenate was the dominant species at only one of the POI. The remaining eight POI were composed of both Se(IV) and Se(VI), with minor Se(-II, 0) contributions. The results of this research suggest that the reduced Se species in the soil parent material are oxidizing to Se(VI), one of the more mobile species of Se in the environment. This information can be used to better predict and manage Se availability in soils.  相似文献

Group II (non-proteolytic) Clostridium botulinum poses a safety hazard in modern food processing, which consists of mild pasteurization treatments, anaerobic packaging, extended shelf lives and chilled storage. The high risk is reflected in the relatively large number of botulism cases due to group II C. botulinum in commercially produced foods during the past decades. Because of the high prevalence of group II C. botulinum in the environment, food raw materials may carry spores. Although group II spores are less heat-resistant than group I (proteolytic) spores, they can tolerate the heat treatments employed in the chilled food industry. Some food components may actually provide spores with protection from heat. Spore heat resistance should therefore be investigated for each food in order to determine the efficiency of industrial heat treatments. Group II strains are psychrotrophic and thus they are able to grow at refrigeration temperatures. Anaerobic packages and extended shelf lives provide C. botulinum with favourable conditions for growth and toxin formation. As the use of salt and other preservatives in these foods is limited, microbiological safety relies mainly on refrigerated storage. This sets great challenges on the production of chilled packaged foods. To ensure the safety of these foods, more than one factor should safeguard against botulinal growth and toxin production.  相似文献

The aim of this study was to determine the tocopherol contents of wine by-products including grape seed, pomace (seed, skin and stem) and bagasse (skin and stem) by using two different extraction methods in order to evaluate the possibilities of them as natural non-polar antioxidants. Cold extraction appeared to give more tocopherol contents in seeds than hot extraction. But hot extraction was found more suitable for pomace and bagasse. α-Tocopherol was the most abundant tocopherol in the grape seed, pomace and bagasse. Although γ- and δ-tocopherols were found in the samples with low concentrations, β-tocopherol was not detected in the samples. Kalecik karası had the highest α and total tocopherol compared to the other cultivars. On the other hand, hot extracted bagasse in all cultivars gave the highest tocopherol contents. As a conclusion, wine by-products, a large scale wastes, can be evaluated both to get natural tocopherol source and to obtain economical gain.  相似文献
