Repair of parenchyma by stem/progenitor cells is seen as a possible alternative to cure acute and chronic renal failure in future. To learn about this therapeutic purpose, the formation of nephrons during organ growth is under focus of present research. This process is triggered by numerous morphogenetic interactions between epithelial and mesenchymal cells within the renal stem/progenitor cell niche. Recent data demonstrate that an astonishingly wide interstitial interface separates both types of stem/progenitor cells probably controlling coordinated cell-to-cell communication. Since conventional fixation by glutaraldehyde (GA) does not declare in transmission electron microscopy the spatial separation, improved contrasting procedures were applied. As a consequence, the embryonic cortex of neonatal rabbit kidneys was fixed in solutions containing glutaraldehyde in combination with cupromeronic blue, ruthenium red or tannic acid. To obtain a comparable view to the renal stem/progenitor cell niche, the specimens had to be orientated along the cortico-medullary axis of lining collecting ducts. Analysis of tissue samples fixed with GA, in combination with cupromeronic blue, demonstrates demasked extracellular matrix. Numerous braces of proteoglycans cover, as well, the basal lamina of epithelial stem/progenitor cells as projections of mesenchymal stem/progenitor cells crossing the interstitial interface. Fixation with GA containing ruthenium red or tannic acid illustrates strands of extracellular matrix that originate from the basal lamina of epithelial stem/progenitor cells and line through the interstitial interface. Thus, for the first time, improved contrasting techniques make it possible to analyze in detail a microheterogeneous composition of the interstitial interface within the renal stem/progenitor cell niche.  相似文献

A novel organic field-effect transistor (OFET)-sensor concept is presented based on the application of an ion-conducting organic dielectric material, which is chemically adapted to change its electronic properties upon contact with an analyte, thereby generating an electrically detectable response. By employing pH-sensitive, ring-opening metathesis polymerized materials as gate dielectrics in bottom-contact OFETs with a meander-shaped top-gate structure, the concept was successfully realized and evaluated with ammonia (NH3) as gaseous analyte, easily providing distinct sensor response at concentration levels as low as 100 ppm. In addition to current–voltage OFET-analysis, optical spectroscopy and capacitance measurements were used to rationalize the underlying sensor mechanism, which is mainly attributed to a deprotonation of the pH-sensitive groups of the active-sensing dielectrics by NH3 and a resulting generation of mobile ions, leading to an increase of the charge carrier density within the OFET channel. The proposed concept provides several advantages over existing OFET-sensor detection principles, including the separation of the sensing mechanism from the charge-transport functionality of the semiconductor, inherent protection of the latter against air exposure and increased selectivity by the application of specific dielectric materials. It therefore offers a great deal of promise in contributing to the development of cheap, integrated, smart and flexible (bio)sensor systems.  相似文献

BACKGROUND: Chemical flower thinning with ammonium thiosulfate (ATS) was applied on branches of sweet cherry (Prunus avium L.) cultivars ‘Blaze Star’, ‘Samba’, ‘Techlovan’ and ‘Merchant’ (all grafted on GiSelA 5) in a two‐year field study in 2006 and 2007. Different concentrations of ATS (8.82 and 17.64 g L?1) and different application times were studied. Fruit quality parameters analysed included crop load, fruit size, fruit weight, flesh/stone ratio, content of soluble solids, pH value and content of titratable acids. RESULTS: Crop load was significantly affected (P < 0.01) by application, cultivar and year. Fruit size, fruit weight and content of titratable acids were not improved by any of the application strategies. An influence of cultivar and application on pH value was observed (P < 0.05). Flesh/stone ratio was significantly enhanced (P < 0.05) in ‘Blaze Star’ only, while content of soluble solids was significantly affected in ‘Techlovan’ only in 2007. CONCLUSION: No general improvement in quality parameters was achieved by chemical flower thinning in the P. avium cultivars studied over a two‐year field experiment. In organic bulk heterojunction solar cells (oBHJ) the blend morphology in combination with the charge transport properties of the individual components controls the extracted photocurrent. The organic field‐effect transistor (OFET) has been proved as a powerful instrument to evaluate the unipolar carrier transport properties in a wide range of cases. In our work we extend the OFET concept to the evaluation of the bipolar transport properties in polymer‐fullerenes blends and propose a method to improve the accuracy of the evaluation. The method is based on capacitance–voltage (C–V) measurements on MOS structures prepared on the same blends and delivers complementary information on the bulk heterojunction to the one obtained with FETs. The relevance for photovoltaic applications is investigated through the correlation between the current–voltage behavior of solar cells and the bipolar mobility for composites with varying polymer molecular weight and processed from different solvents. In particular the transport features of solar cells produced from o‐Xylene (oX), a non chlorinated solvent more suitable to production requirements, have been compared to the one of devices cast from Chlorobenzene (CB) solution. For the P3HT‐PCBM blend a consistent correlation between the mobility and the electrical fill factor and power performance was found. A significant asymmetry in the bipolar carrier mobility, together with low electron mobility dependent on the Mw value, affects the performances of thick o‐Xylene cast devices. In the case of devices processed from Chlorobenzene the slower carrier has higher mobility and the small electrical losses detected are eventually more related to the formation of space‐charge and eventually to surface recombination. This results in an efficient charge collection that is almost thickness independent. We report a dependence of the slow‐carrier type (electrons or holes) and their mobility on the specific combination of molecular weight and solvent. The mobility data and the solar cell performance coherently fit to the prediction of a device model only based on the drift of carriers under the built‐in electric field originated in the donor‐acceptor oBHJ.  相似文献

The aim of the study was the analysis of the greater omentum morphology and vascularization and the presentation of new, own classification of omentum by the type of vascularization and to the criterion of the possibility for simpler preparation and lengthening of omental flap. Prospective clinical study included the first 100 operated in whom omental graft was formed for omentomyelopexy. In 10% of all cases, the omenta deviated from the common pattern and size (they were extremely small in 6%). Omenta were classified by the vascularization type into: omental type I (had either complete gastroepiploic arcade or/and Haller-Barcow's arcade or/and a. omentalis propria) and there were 54%; type II (had none of those arteries) was presented in 42%; omental type III (unordinary, bizarre vascularization forms) was presented in 4%. Every omental type was divided into subtypes. In 41% of cases, the omental flap needed to be lengthen to achieve the level of spinal lesion, and the need of omental flap lengthening depended significantly on the omental type. This classification provides fast orientation and expeditious forming of omental flap in all its modalities.  相似文献

The application of Adams methods for the numerical solution of stochastic differential equations is considered. Especially we discuss the path-wise (strong) solutions of stochastic differential equations with additive noise and their numerical computation. The special structure of these problems suggests the application of Adams methods, which are used for deterministic differential equations very successfully. Most excitatory synaptic connections occur on dendritic spines. Calcium imaging experiments have suggested that spines constitute individual calcium compartments, but recent results have challenged this idea. Using two-photon microscopy to image fluorescence with high resolution in strongly scattering tissue, we measured calcium dynamics in spines from CA1 pyramidal neurons in slices of rat hippocampus. Subthreshold synaptic stimulation and spontaneous synaptic events produced calcium accumulations that were localized to isolated spines, showed stochastic failure, and were abolished by postsynaptic blockers. Single somatic spikes induced fast-peaking calcium accumulation in spines throughout the cell. Pairing of spikes with synaptic stimulation was frequently cooperative, that is, it resulted in supralinear calcium accumulations. We conclude: (1) calcium channels exist in spine heads; (2) action potentials invade the spines; (3) spines are individual calcium compartments; and (4) spines can individually detect the temporal coincidence of pre- and postsynaptic activity, and thus serve as basic functional units of neuronal integration.  相似文献
