In order to utilise sardinelle (Sardinellaaurita) protein by-products, which is normally discarded as industrial waste in the process of fish manufacturing, heads and viscera proteins were hydrolysed by different proteases to obtain antioxidative peptides. All hydrolysates showed different degrees of hydrolysis and varying degrees of antioxidant activities. Hydrolysate generated with crude enzyme extract from sardine (Sardinapilchardus) displayed high antioxidant activity, and the higher DPPH radical-scavenging activity (87 ± 2.1% at 2 mg/ml) was obtained with a degree of hydrolysis of 6%. This hydrolysate was fractionated by size exclusion chromatography on a Sephadex G-25 into eight major fractions (P1–P8). Fraction P4, which exhibited the highest DPPH scavenging activity, was then fractionated by reversed-phase high performance liquid chromatography (RP-HPLC). Seven antioxidant peptides were isolated. The molecular masses and amino acids sequences of the purified peptides were determined using ESI-MS and ESI-MS/MS, respectively. Their structures were identified as Leu-His-Tyr, Leu-Ala-Arg-Leu, Gly-Gly-Glu, Gly-Ala-His, Gly-Ala-Trp-Ala, Pro-His-Tyr-Leu and Gly-Ala-Leu-Ala-Ala-His. The first peptide displayed the highest DPPH radical-scavenging activity (63 ± 1.57%; at 150 μg/ml) among these peptides.  相似文献

The aim of the study was to evaluate the impact of structural and biochemical characteristics of muscle intramuscular connective tissue on beef quality. The experimental design was based on three muscles of three breeds sampled as fresh material and cooked at 55 °C (Longissimus thoracis and Semimembranosus) or at 70 °C (Semimembranosus and Biceps femoris) for quality assessment. The results showed that muscle characteristics influence beef quality differently from one muscle to another. In grilled LT, proteoglycan content contributed negatively to juiciness, and intramuscular lipids were linked positively to tenderness, flavour, residues and overall liking scores. In grilled SM, cross-link and lipid contents were involved in beef quality. In BF cooked to 70 °C, perimysial branch points were negatively linked to tenderness. In SM cooked to 70 °C, perimysial area was involved in beef quality. These results should allow a better understanding of the factors involved in background toughness, in juiciness and flavour of meat.  相似文献

The measurement of the flow through complex combined sewer overflow structures in the frame of automated monitoring remains difficult. In this paper, a methodology based on the use of computational fluid dynamics (CFD) modelling in order to improve the instrumentation of a downstream-controlled dual-overflow structure is presented. The dual-overflow structure is composed of two combined sewer overflows (CSOs) connected by a rectangular channel and controlled by a downstream gate located at the entry of the Meyzieu waste water treatment plant (close to Lyon, France). The analysis of the CFD results provides: (i) a better understanding of the interaction between the two CSOs--that means the hydraulic operation, the hydrodynamic behaviour, the backflow effect--and (ii) an ability to optimise the location of the water depth sensor. The measured water depth is used to assess the overflow rate by means of a numerical relationship. Uncertainties are also assessed.  相似文献

The major whey proteins IgG, serum albumin and alpha-lactalbumin were purified from camel milk using gel permeation and ion-exchange chromatography. Specific antisera against each of them were raised and used to quantify their heat denaturation in early or mature milk over a range of 60-90 degrees C for 10-60 min using the single radial immunodiffusion technique. The heat denaturation midpoints for the mature milk heated 30 min were 67.2, 73.0 and 77.5 degrees C for IgG, albumin and alpha-lactalbumin respectively. The early milk was more sensitive to heat treatment with coagulation at low temperature and heat denaturation midpoints of 64.8, 71.6 and 72.6 degrees C respectively. This difference was related to the high IgG content of the early milk (12.6 mg/ml v. 0.5 mg/ml for the mature milk) and stresses the importance of monitoring the IgG level of milk to assess the absence of colostrum.  相似文献

The paper analyzes the two major components of the Bush Administration’s climate policy, namely an emission intensity target and a technology strategy. The question is whether those components will generate net emission reductions that will contribute to the stabilization of the greenhouse gas concentration at a safe level in the long run. It comes out that the Bush Administration climate policy does not guarantee any meaningful contribution to climate protection. The lenient emission intensity target set by the Administration will most likely allow near term emissions to grow. In the long run, the Bush Administration places a big bet on future climate-friendly technological breakthroughs to cost-effectively compensate for the current and near term net emission increases. But the outcomes of those technological developments are uncertain in terms of emission reduction potential, cost, and timing. The way towards enhanced climate protection will most likely not come from the policies of the current Administration, but rather from the growing concern about the climate issue in Congress and at the state, corporate and civil society levels. These combined forces may raise the playing field at the federal level in the near future.  相似文献

This paper examines climate change impacts on the water resources system of the Manicouagan River (Québec, Canada). The objective is to evaluate the performance of existing infrastructures under future climate projections and the associated uncertainties. The main purpose of the water resources system is hydropower production. A reservoir optimization algorithm, Sampling Stochastic Dynamic Programming (SSDP), was used to derive weekly operating decisions for the existing system subject to reservoir inflows reflecting future climate, for optimum hydropower production. These projections are simulations from the SWAT hydrologic model for climate change scenarios for the period from 2010 to 2099. Results show that the climate change will alter the hydrological regime of the study area: earlier timing of the spring flood, reduced spring peak flow, and increased annual inflows volume in the future compared to the historical climate. The SSDP optimization algorithm adapted the operating policy to the future hydrological regime by adjusting water reservoir levels in the winter and spring, and increasing the release through turbines, which in the end increased power generation. However, there could be more unproductive spills for some power plants, which would decrease the overall efficiency of the existing water resources system.  相似文献

A novel thermoplastic programmable preforming process, TP-P4, has been used to manufacture preforms for non-isothermal compression molding. Commingled glass and polypropylene yarns are deposited by robot onto a vacuum screen, followed by a heat-setting operation to stabilize the as-placed yarns for subsequent handling. After an optional additional preconsolidation stage, the preforms are molded by preheating and subsequent press forming in a shear edge tool. The in- and out-of-plane flow capabilities of the material were investigated, and compared to those of 40 wt% Glass Mat Thermoplastics (GMTs). Although the TP-P4 material has a fiber fraction of 60 wt%, the material could be processed to fill 77 mm deep ribs with a thickness of 3 mm, indicative of complex part production. The pressure requirements for out-of-plane flow were shown to depend on the fiber length and fiber alignment. Segregation phenomena were found to be less severe with TP-P4 than with GMT material.  相似文献

Many uses of protein hydrolysates have been developed and applied to areas such as nutritional therapy, culture media, and the isolation of biologically active peptides. All these applications need carefully controlled and characterized hydrolysates. In order to produce such a type of hydrolysate, it is possible to use haemoglobin which is a very well defined and constant protein source. Enzymic hydrolysis of haemoglobin by pepsin was carried out at pilot-plant scale in an ultrafiltration reactor with mineral membranes. The object was to obtain a reproducible, decolorized, salt-free enzymic hydrolysate. Two types of membranes were tested having 10000 dalton (M5 type) and 20000 dalton (M4 type) cut-offs. Little significant difference was observed in the final products when both types of membranes were used. Reproducibility of hydrolysates was verified by amino acid analysis and gel filtration chromatography. The haemoglobin hydrolysates produced contained more than 90% protein and are especially suitable for fine applications.  相似文献

An integral equation and method of moments (MM) solution are presented for the two-dimensional (2-D) problem of transverse magnetic (TM) scattering by an impedance-sheet extension of a perfectly conducting parabolic cylinder. An integral equation is formulated for a dielectric cylinder of general cross section in the presence of a perfectly conducting parabolic cylinder. It is then shown that the solution for a general dielectric cylinder considerably simplifies for the special case of TM scattering by a thin multilayered dielectric strip that can be represented as an impedance sheet. The solution is termed an MM/Green's function solution, where the unknowns in the integral equation are the electric surface currents flowing in the impedance sheet; the presence of the parabolic cylinder is accounted for by including its Green's function in the kernel of the integral equation. The MM solution is briefly reviewed, and expressions for the elements in the matrix equation and the scattered fields are given. Sample numerical results are provided  相似文献
