Numerous studies have shown that neuronal lipids are highly susceptible to oxidative stress including in those brain areas directly involved in the neurodegenerative process of Alzheimer's disease (AD). Lipid peroxidation directly damages membranes and also generates a number of secondary biologically active products (toxic aldehydes)that are capable of easily attacking lipids, proteins, and DNA. Accumulating evidence has demonstrated regionally increased brain lipid peroxidation in patients with AD; however, extensive studies on specific targets of lipid peroxidation‐induced damage are still missing. The present study represents a further step in understanding the relationship between oxidative modification of protein and neuronal death associated with AD. We used a proteomics approach to determine specific targets of lipid peroxidation in AD brain, both in hippocampus and inferior parietal lobule, by coupling immunochemical detection of 4‐hydroxynonenal‐bound proteins with 2‐D polyacrylamide gel electrophoresis and MS analysis. We identified 4‐hydroxynonenal‐bound proteins in the hippocampus and inferior parietal lobule brain regions of subjects with AD. The identified proteins play different biological functions including energy metabolism, antioxidant system, and structural proteins, thus impairing multiple molecular pathways. Our results provide further evidence for the role of lipid peroxidation in the pathogenesis of AD. 相似文献

Silicon oxycarbide (SiOC) glasses in the form of thin, dense, and crack-free samples were fabricated according to the polymer pyrolysis route starting from cross-linked polysiloxane. The amount of free carbon in the final SiOC materials was varied in the range 18-60 vol%. The mechanical properties of the SiOC glasses were measured by nanoindentaion technique and revealed that both the Young's modulus and the hardness decrease with increase in the free carbon content and follow a simple rule of mixtures model. 相似文献

Silicon-based polymers evolve into refractory ceramics when heated gradually up to ~1000°C. The conversion is accompanied by the loss of gaseous species, and by a two-fold increase in density. The shrinkage can produce microcracks if the heating rate is too high, or if the specimen is too thick. This communication builds on earlier work whereby the measurement of gas evolution, and its relationship with viscous flow, are related to the onset of fracture in disk-shaped green (polymer) samples. The onset is determined as a function of the thickness of the disks, and of the heating rate. The results are presented in the form of a processing map. The overlay with gas evolution, and strain-rate measurements, suggest that fracture initiates with the release of hydrogen and methane, starting at temperatures near 750°C. 相似文献

Mimicking the properties of the extracellular matrix is crucial for developing in vitro models of the physiological microenvironment of living cells. Among other techniques, 3D direct laser writing (DLW) has emerged as a promising technology for realizing tailored 3D scaffolds for cell biology studies. Here, results based on DLW addressing basic biological issues, e.g., cell‐force measurements and selective 3D cell spreading on functionalized structures are reviewed. Continuous future progress in DLW materials engineering and innovative approaches for scaffold fabrication will enable further applications of DLW in applied biomedical research and tissue engineering. 相似文献

Motivated by nano‐scale experimental evidence on the dispersion characteristics of materials with a lattice structure, a new multi‐scale gradient elasticity model is developed. In the framework of gradient elasticity, the simultaneous presence of acceleration and strain gradients has been denoted as dynamic consistency. This model represents an extension of an earlier dynamically consistent model with an additional micro‐inertia contribution to improve the dispersion behaviour. The model can therefore be seen as an enhanced dynamic extension of the Aifantis' 1992 strain‐gradient theory for statics obtained by including two acceleration gradients in addition to the strain gradient. Compared with the previous dynamically consistent model, the additional micro‐inertia term is found to improve the prediction of wave dispersion significantly and, more importantly, requires no extra computational cost. The fourth‐order equations are rewritten in two sets of symmetric second‐order equations so that ‐continuity is sufficient in the finite element implementation. Two sets of unknowns are identified as the microstructural and macrostructural displacements, thus highlighting the multi‐scale nature of the present formulation. The associated energy functionals and variationally consistent boundary conditions are presented, after which the finite element equations are derived. The genetic diversity among 15 fig accessions (Ficus carica L), belonging to 9 renowned cultivars of the Calabrian fig collection, and 24 unidentified genotypes of figs also located in the Calabrian region of Southern Italy, was investigated by random amplified polymorphic DNA (RAPD) analysis. The genetic similarity values were calculated for the 39 samples, and a dendrogram was elaborated by cluster analysis according to the UPGMA algorithm. The generated DNA fragments grouped the samples into two main clusters of RAPD profiles. Most of the 24 unknown samples, coming from the Luzzi area, formed a unique cluster with high degree of genetic similarity. This indicates that it is possible to distinguish, at DNA level, the fig trees with an already well‐known potential to produce figs suitable for the drying process and possibly to specify cultivars with suitable features for industrial transformation. A numerical study is performed to simulate the vertical deformation of a depth interval representing a marker spacing (10.5 m) located in a deep heterogeneous sedimentary reservoir. Realistic lithostratigraphic sequences typical of the Northern Adriatic basin are used. A number of scenarios are addressed consistent with the available data. In particular two basic geologic scenarios at the marker scale are simulated, one where sands prevail within the marker spacing (SD), and another where thin sandy and clayey layers alternate (CL). The sensitivity of the marker response is investigated in relation to clay and sand permeability and compressibility contrast, Biot’s coefficient, and respective position of monitoring and fluid pumping wells. The modeling results show that rock may indeed expand above and below depleted layers. Depending on the marker position the expansion may partially offset the compaction, especially in the CL scenario with a very low permeable clay. To obtain a representative field compaction the markers should span a depth interval made mostly by sand and entirely depleted, and should be installed in a test hole far from producing wells. Compressibility contrast and the Biot coefficient play a secondary role. Very critical measurements are provided by two markers which incorporate a thin (≈1 m) depleted level overlain and underlain by almost impermeable clay layers. 相似文献

In this article, we propose an agent‐based approach for managing e‐commerce activities. In our approach, an agent is present in each e‐commerce site, managing the information stored there. In addition, another agent is associated with each customer, handling his/her profile. The proposed approach is based on the use of a particular conceptual model called the Behaviour‐Semantic Distance and Relevance (B‐SDR) network, which is capable of uniformly representing and handling information stored in e‐commerce sites and customer profiles. The capabilities of the B‐SDR network model are exploited to let customer and site agents cooperate in such a way in order to support a customer in identifying, whenever he/she accesses an e‐commerce site, those products and services present in the site itself and for better matching his/her interests.
