Miniaturizing experimental sample volumes to the nanoliter volume range is one of the most economical ways to perform mid- and high-throughput compound screening experiments. Existing automation platforms for nanoliter fluid handling can be bulky, expensive, and require periodic calibration to provide consistent liquid dispensing. In addition, even with frequent calibration, significant instrument-to-instrument variation in low-volume dispensing can occur between different instrument platforms. Many of these issues can be addressed by the use of PocketTips. PocketTips are tips with a defined internal pocket designed to hold specific nanoliter volumes of compound dissolved in dimethylsulfoxide. Although the overall liquid-handling process with PocketTips uses the aspirate/dispense features of the specific liquid-handling device being used, the dispensed nanoliter volume is solely based on the dimensions of the pocket of the PocketTip and thus, the liquid-handling device itself need not have nanoliter dispensing capabilities. In this report, we demonstrate the performance of PocketTips on different automation platforms. In addition, we used a cell-based ?-lactamase reporter assay system to demonstrate that compound delivery by PocketTips compares favorably with a standard compound addition technique.  相似文献

Abstract— A processing technology based upon a temporary bond—debond approach has been developed that enables direct fabrication of high‐performance electronic devices on flexible substrates. This technique facilitates processing of flexible plastic and metal‐foil substrates through automated standard semiconductor and flat‐panel tool sets without tool modification. The key to processing with these tool sets is rigidifying the flexible substrates through temporary bonding to carriers that can be handled in a similar manner as silicon wafers or glass substrates in conventional electronics manufacturing. To demonstrate the power of this processing technology, amorphous‐silicon thin‐film‐transistor (a‐Si:H TFT) backplanes designed for electrophoretic displays (EPDs) were fabricated using a low‐temperature process (180°C) on bonded‐plastic and metal‐foil substrates. The electrical characteristics of the TFTs fabricated on flexible substrates are found to be consistent with those processed with identical conditions on rigid silicon wafers. These TFTs on plastic exhibit a field‐effect mobility of 0.77 cm2/V‐sec, on/off current ratio >109 at Vds = 10 V, sub‐threshold swing of 365 mV/dec, threshold voltage of 0.49 V, and leakage current lower than 2 pA/μm gate width. After full TFT‐array fabrication on the bonded substrate and subsequent debonding, the flexible substrate retains its original flexibility; this enables bending of the EPD display without loss in performance.  相似文献

The practice of implementing real-time optimization (RTO) using a rigorous steady-state model, in conjunction with model predictive control (MPC), dates back to the late 1980s. Since then, numerous projects have been implemented in refinery and chemical plants, and RTO has received significant attention in the industrial and academic literature. This history affords us the opportunity to assess the impact and success of RTO technology in the process industries. We begin with a discussion of the role RTO serves in the hierarchy of control and optimization decision making in the plant, and outline the key steps of the RTO layer and the coordination with MPC. Where appropriate, we point out the different approaches that have been used in practice and discuss the success factors that directly relate to the success of RTO within an organization. We also discuss alternative approaches that have been used to alleviate some of the challenges associated with implementing RTO and which may be appropriate for those unwilling to commit to the traditional RTO approach. Lastly, we provide suggestions for improvement to motivate further research.  相似文献

Many complex natural systems studied in the geosciences are characterized by simple local-scale interactions that result in complex emergent behavior. Simulations of these systems, often implemented in parallel using standard central processing unit (CPU) clusters, may be better suited to parallel processing environments with large numbers of simple processors. Such an environment is found in graphics processing units (GPUs) on graphics cards.This paper discusses GPU implementations of three example applications from computational fluid dynamics, seismic wave propagation, and rock magnetism. These candidate applications involve important numerical modeling techniques, widely employed in physical system simulations, that are themselves examples of distinct computing classes identified as fundamental to scientific and engineering computing. The presented numerical methods (and respective computing classes they belong to) are: (1) a lattice-Boltzmann code for geofluid dynamics (structured grid class); (2) a spectral-finite-element code for seismic wave propagation simulations (sparse linear algebra class); and (3) a least-squares minimization code for interpreting magnetic force microscopy data (dense linear algebra class). Significant performance increases (between 10× and 30× in most cases) are seen in all three applications, demonstrating the power of GPU implementations for these types of simulations and, more generally, their associated computing classes.  相似文献

Modern businesses are facing the challenge of effectively coordinating their supply chains from upstream to downstream services. It is a complex problem to search, schedule, and coordinate a set of services from a large number of service resources under various constraints and uncertainties. Existing approaches to this problem have relied on complete information regarding service requirements and resources, without adequately addressing the dynamics and uncertainties of the environments. The real-world situations are complicated as a result of ambiguity in the requirements of the services, the uncertainty of solutions from service providers, and the interdependencies among the services to be composed. This paper investigates the complexity of supply chain formation and proposes an agent-mediated coordination approach. Each agent works as a broker for each service type, dedicated to selecting solutions for each service as well as interacting with other agents in refining the decision making to achieve compatibility among the solutions. The coordination among agents concerns decision making at strategic, tactical, and operational level. At the strategic level, agents communicate and negotiate for supply chain formation; at the tactical level, argumentation is used by agents to communicate and understand the preferences and constraints of each other; at the operational level, different strategies are used for selecting the preferences. Based on this approach, a prototype has been implemented with simulated experiments highlighting the effectiveness of the approach.  相似文献

In this paper, we investigate the camera network placement problem for target coverage in manufacturing workplaces. The problem is formulated to find the minimum number of cameras of different types and their best configurations to maximise the coverage of the monitored workplace such that the given set of target points of interest are each k-covered with a predefined minimum spatial resolution. Since the problem is NP-complete, and even NP-hard to approximate, a novel method based on Simulated Annealing is presented to solve the optimisation problem. A new neighbourhood generation function is proposed to handle the discrete nature of the problem. The visual coverage is modelled using realistic and coherent assumptions of camera intrinsic and extrinsic parameters making it suitable for many real world camera based applications. Task-specific quality of coverage measure is proposed to assist selecting the best among the set of camera network placements with equal coverage. A 3D CAD of the monitored space is used to examine physical occlusions of target points. The results show the accuracy, efficiency and scalability of the presented solution method; which can be applied effectively in the design of practical camera networks.  相似文献

The use of information technology to enhance teaching and learning processes has been practiced for a number of years now. However, the rapid growth in the use of the Internet has led to a new dimension in interactive and collaborative learning anytime and anyplace dynamically. With the explosion of “virtual education initiatives”, the question of the feasibility and success criteria for such projects quickly arises. To address the question in an organized way, we propose a project assessment based on critical success factors. Hence, in this article, we draw on a widely recognized critical success factor framework. We (slightly) adjust the framework to fit the special characteristics of virtual education initiatives, and apply to one case study, namely the virtual education initiative at the Faculty of Business at the City University of Hong Kong. The results suggest that the past success of the case is due to the adherence to the large majority of critical success factors. However, it also outlines some areas of concern. The paper concludes with a discussion on the strength and limitations of virtual learning environment as well as future directions. The mechanical properties of one of the new high strength polymer modified hydraulic cements have been investigated. An important parameter for the material is the amount of polymer present, and the properties are found to be dependent upon the degree of drying. For example, in the wet state, polymer content has little effect upon ultimate flexural strength, but does cause nonlinearity in the stress-strain behaviour. Although increasing polymer content causes a reduction in the initial tangent modulus, it is shown that retardation of hydration may account for this. In the dry state, increasing polymer content leads to increasing flexural strength, fracture toughness and failure strain, but leaves initial elastic modulus relatively unchanged. It is concluded that removal of pores is not the principal strengthening mechanism since strength increases are consequent upon water removal from the microstructure in the presence of the polymer. A fibrillar pull-out model is proposed to explain the observed behaviour of both wet and dry material and ordinary Portland cement paste, which shows good correlation with experimental results. The principal effect of the polymer is to act as an adhesive at the interface between interacting CSH fibrils.  相似文献
