Treatments of 1.25% or 2.5% ascorbic acid (AA); 0.1% or 0.2% rosemary; or a combination of 0.15% Origanox™ + 0.3% AA on beef lumbar vertebrae (n = 72) were evaluated for their effectiveness in preventing bone discoloration in retail packages. Vertebrae were held either 6 or 14 d postmortem; cut into 2.54-cm sections; and packaged into either polyvinyl chloride film overwrap (PVC), high-oxygen (80% O2/20% CO2) modified-atmosphere packages (MAP), or ultra-low-oxygen (70% N2/30% CO2) MAP. Controls and vertebrae treated with 0.1% or 0.2% rosemary discolored significantly, whereas 0.15% Origanox™ + 0.3% AA was effective through d 2 of display in PVC and high-oxygen MAP. In both high- and ultra-low-oxygen MAP, 1.25% AA was as effective as 2.5% AA in preventing bone discoloration. Vertebrae treated with AA or packaged in ultra-low-oxygen MAP had the largest a* values over display time. Bone discoloration was effectively prevented with AA, especially at the 2.5% concentration.  相似文献

During the North Atlantic marine boundary layer experiment (NAMBLEX) sampling campaign at Mace Head, Ireland, both continental and maritime air masses were sampled. Aerosol was characterized both with a TSI 3800 time-of-flight mass spectrometer (ATOFMS) and a MOUDI microorifice impactor, and particle number counts were measured independently with an aerodynamic particle sizer. The data have been analyzed in order to elucidate factors determining the particle detection efficiencies of the ATOFMS. These are broken down according to the efficiency of the inlet system, the hit efficiency on particles which enter the sensing zone of the instrument and the sensitivity of the measured ion signal to the chemical species. A substantial matrix effect depending on the chemical composition of the aerosol sampled at the time was found, which is reflected in variations in the hit efficiency of particles entering the sensing zone of the instrument with the main desorption-ionization laser. This is in addition to the strong inverse power-law dependence of inlet transmission efficiency on particle diameter. The variation in hit efficiency with particle type is likely attributable to differences in the energetics of laser energy absorption, ablation, and ion formation. However, once variations in both inlet transmission and hit efficiencies are taken into account, no additional matrix dependence of ATOFMS response is required to obtain a linear relationship between the ion signal and the concentration of a particular chemical species. The observations show that a constant mass of material is ionized from each particle, irrespective of size. Consequently the integrated ion signal for a given chemical component and particle size class needs to be increased by a factor related to the cube of particle diameter in order to correlate with the airborne mass of that component.  相似文献

In an effort to identify a more suitable antibiotic for utilization in mycological media, 12 food borne fungal species from various genera including Alternaria alternata, Aspergillus flavus, A. niger, Eurotium chevalieri, Fusarium moniliforme, Penicillium sp., Rhizopus stolonifer, Saccharomyces cerevisiae, Candida tropicalis, Geotrichum candidum, Rhodotorula glutinis and Kluyveromyces thermotolerans along with 21 chloramphenicol-resistant bacterial isolates from fresh produce and ATCC cultures of Pseudomonas aeruginosa, P. fluorescens, E. coli, Pectobacterium carotovorum, Bacillus cereus and Staphylococcus spp. were tested for their abilities to grow on dichloran rose bengal agar containing various levels of gentamicin, chlortetracycline or chloramphenicol. Results indicated that all fungal isolates except for Rh. glutinis and R. stolonifer grew well on all media tested. Rh. glutinis did not grow on media containing gentamicin whereas R. stolonifer produced very restricted or no growth on these media. All bacterial isolates from fresh produce, P. aeruginosa (ATCC 27853) and P. fluorescens (ATCC BAA-477) grew well at 100, 125 and 150 mg chloramphenicol/liter medium, but they did not grow on media containing chlortetracycline (100, 125, or 150 mg/L) or gentamicin (15, 25, or 35 mg/L). P. aeruginosa (ATCC 10145) grew well on media containing chloramphenicol or gentamicin, but not in the presence of chlortetracycline. P. carotovorum, E. coli, B. cereus and Staphylococcus spp. did not grow on any of the selective media tested.  相似文献

Effect of lameness on ovarian activity in postpartum holstein cows   总被引:1,自引:0,他引:1  
A longitudinal study was conducted to examine the relationship between lameness and delayed ovarian cyclicity during the first 60 d postpartum and days to first luteal activity during the first 300 d postpartum in Holstein cows. Two hundred thirty-eight cows from a 600-cow dairy that calved during a 12-mo period were used. Cows were classified into 1 of 6 categories of lameness during the first 35 d postpartum using a locomotion scoring system. Cows were blood-sampled weekly for detection of plasma progesterone concentrations during the first 300 d postpartum. Cows with delayed resumption of ovarian cyclicity were defined as those with progesterone concentrations consistently <1 ng/mL during the first 60 d postpartum. The null hypothesis that risk of delayed cyclicity is the same in cows classified as nonlame, moderately lame, or lame (after adjusting for potential modifying or confounding effects of loss of body condition and other variables related with delayed cyclicity) was tested using logistic regression. Analysis of results of the study reported here support the hypothesis that lameness is associated with delayed ovarian activity in Holstein cows during the early postpartum period. Cows classified as lame had 3.5 times greater odds of delayed cyclicity, compared with cows classified as nonlame. Attributable proportion analysis indicated that delayed ovarian cyclicity in lame cows would be reduced by 71%, if lameness had been prevented.  相似文献

The objective of this study was to improve the accumulation of ginsenosides by the adventitious root cultures of ginseng, which are important secondary metabolites with pharmaceutical applications. The adventitious roots were cultured in bioreactors for 50 d using 1.5-strength Murashige and Skoog (MS) medium supplemented with 10 mg/l indole acetic acid and 75 g/l sucrose. Kinetic studies of the nutrient composition of the spent medium revealed the gradual depletion of various inorganic nutrients and sugars. Cultures were supplied with fresh nutrient medium (medium exchange or replenishment with 0.75- and 1.0-strength MS medium) after 10 and 20 d of culture initiation to fulfill the nutritional requirements of adventitious roots. Medium replenishment strategy (with 1.0-strength MS medium after 20 d of culture) significantly improved the growth of adventitious roots and the biosynthesis of ginsenosides by the adventitious roots. This work is useful for the large-scale cultivation of adventitious roots for the production of ginsenosides.  相似文献

ABSTRACT:  This study was undertaken to evaluate the applicability of bacterial alkaline phosphatase (ALP) activity for rapid monitoring of total mesophilic bacteria counts in spinach. A set of fresh and decayed spinach mixtures were tested to rapidly (10 min) monitor spinach bacterial counts. To assay ALP activity, Lumigen APS-5 was used as a substrate. Bovine ALP was reduced after heat treatment at 75 °C for 1 min; in contrast, bacterial ALP activity increased. To differentiate bacterial ALP from other ALP, heat treatment (75 °C, 1 min) was applied before measurement. As a result, a regression equation was established between the actual mesophilic aerobic bacteria count and ALP activity of spinach mixtures ( r = 0.90). The predicted total mesophilic aerobic bacterial count calculated from the fitted regression line (predicted log10 CFU/g = 0.00056 × ALP values + 1.4002) showed a high correlation with the actual observed total bacterial count ( r = 0.93). The ALP assay is a simple and rapid method to utilize for estimation of existing or contaminating microorganism levels on spinach.  相似文献

In this study, the reactivity of lead (Pb(II)) on naturally occurring Mn(III,IV) (oxyhydr)oxide minerals was evaluated using kinetic, thermodynamic, and spectroscopic investigations. Aqueous Pb(II) was more strongly adsorbed to birnessite (delta-MnO1.7) than to manganite (gamma-MnOOH) under all experimental conditions. The isoteric heat of Pb adsorption (delta HT) or birnessite was 94 kJ mol-1 at a surface loading of 1.1 mmol g-1, and decreased with increasing adsorption density. This indicated that adsorption was an endothermic process and that birnessite possessed heterogeneous sites of reactivity for Pb. X-ray absorption fine structure (XAFS) spectra revealed that Pb was adsorbed as inner-sphere complexes on both birnessite and manganite with no evidence to suggest oxidation as an operative sorption mechanism. Lead appeared to coordinate to vacancy sites in the birnessite layer structure with concurrent release of Mn to solution, which resulted in a greater number of second shell Mn scatterers in Pb-birnessite when compared to Pb-manganite samples. The difference in Pb coordination apparently explained the contrasting desorption behavior between the two Mn minerals. These results have significant implications for Pb partitioning in soil environments containing solid-phase Mn(III,IV) (oxyhydr)oxides.  相似文献
