根据拟枝孢镰刀菌等真菌中Tri101的保守序列设计简并引物,利用简并PCR和基因组步行法克隆了哈茨木霉菌Tri101基因.结果表明:Tri101基因的开放阅读框为1 314 bp,无内含子存在,与预计的Tri101序列特征一致.该基因编码蛋白质由437个氨基酸组成,预测相对分子质量约为4.75 kD,系统进化树显示其与棒曲霉(Aspergillus clavatus)NRRL 1亲缘关系最近,与NRRL 1中3-O-乙酰转移酶的的蛋白质同源性达到55%.Blast数据库分析表明其属于转移酶总科,单端孢酶烯乙酰转移化酶.以禾谷镰刀菌(Fusarium graminearum)TRI101蛋白(Parent PDB:3b2s Chain:A)的晶体为模板进行了同源建模,为进一步研究该基因的功能奠定基础. 相似文献

Over the last 20 years, the performance gap between CPU and memory has been steadily increasing. As a result, a variety of techniques has been devised to hide that performance gap, from intermediate fast memories (caches) to various prefetching and memory management techniques for manipulating the data present in these caches. In this paper we propose a new memory management technique that takes advantage of access pattern information that is available at compile time by prefetching certain data elements before explicitly being requested by the CPU, as well as maintaining certain data in the local memory over a number of iterations. In order to better take advantage of the locality of reference present in loop structures, our technique also uses a new approach to memory by partitioning it and reducing execution to each partition, so that information is reused at much smaller time intervals than if execution followed the usual pattern. These combined approaches—using a new set of memory instructions as well as partitioning the memory—lead to improvements in total execution time of approximately 25% over existing methods. 相似文献

The implementation of Asynchronous Learning Network (ALN) technology in a large on‐campus course over several years is reviewed, and recent data concerning both educational and cost effectiveness are presented. Even with higher course standards for success, student performance on examinations has improved, a larger fraction of students achieve the goals of the class, and the proportion of students who excel has increased. Female students benefit even more than their male counterparts. The level of communication and interaction among students has also increased dramatically, with mostly positive (but some negative) effects. Data concerning cost effectiveness indicate that the technology can reduce costs, but perhaps more importantly, it can increase the quality of education without increasing costs. 相似文献

The relationship between heat-treatment parameters and microstructure in titanium alloys has so far been mainly studied empirically,
using characterization techniques such as microscopy. Calculation and modeling of the kinetics of phase transformation have
not yet been widely used for these alloys. Differential scanning calorimetry (DSC) has been widely used for the study of a
variety of phase transformations. There has been much work done on the calculation and modeling of the kinetics of phase transformations
for different systems based on the results from DSC study. In the present work, the kinetics of the β ⇒ α transformation in a Ti-6Al-4V titanium alloy were studied using DSC, at continuous cooling conditions with constant cooling
rates of 5 °C, 10 °C, 20 °C, 30 °C, 40 °C, and 50 °C/min. The results from calorimetry were then used to trace and model the
transformation kinetics in continuous cooling conditions. Based on suitably interpreted DSC results, continuous cooling-transformation
(CCT) diagrams were calculated with lines of isotransformed fraction. The kinetics of transformation were modeled using the
Johnson-Mehl-Avrami (JMA) theory and by applying the “concept of additivity.” The JMA kinetic parameters were derived. Good
agreement between the calculated and experimental transformed fractions is demonstrated. Using the derived kinetic parameters,
the β ⇒ α transformation in a Ti-6Al-4V alloy can be described for any cooling path and condition. An interpretation of the results

The research presented in this paper evaluates the flexural performance of bridge deck panels reinforced with 2D fiber-reinforced polymer (FRP) grids. Two different FRP grids were investigated, one reinforced with a hybrid of glass and carbon fibers and a second grid reinforced with carbon fibers only. Laboratory measured load-deflection, load-strain (reinforcement and concrete), cracking, and failure behavior are presented in detail. Conclusions regarding failure mode, limit-state strength, serviceability, and deflection compatibility relative to AASHTO mandated criteria are reported. Test results indicate that bridge decks reinforced with FRP grids will be controlled by serviceability limit state and not limit-state ultimate strength. The low axial stiffness of FRP results in large service load flexural deflections and reduced shear strength. In as much as serviceability limits design, overreinforcement is recommended to control deflection violation. Consequently, limit-state flexural strength will be compression controlled for which reduced service stresses or ACI unified compression failure strength reduction factors are recommended. 相似文献
