Criteria for detection of chromosome aberrations by Comparative Genomic Hybridization (CGH) are not standardized and improvement of this part of the analysis is of paramount importance to the applicability of the technique. The aim of this work was to suggest CGH detection criteria that increase the specificity and sensitivity and at the same time include chromosome regions previously excluded from CGH analysis. We analyzed 33 hybridizations with normal DNA and modified our CGH software in order to use a selection of these normal analyses as a model for interpretation of analyses of unknown samples. This approach was successfully tested on 14 samples with known aberrations.  相似文献

Excessive brain iron has been found in several neurodegenerative diseases. However, little information is available about mechanism of iron uptake by different types of brain cells including neurons. In this study, transferrin-bound iron (Tf-Fe) accumulation in the cultured cerebellar granule cell was investigated in vitro. After 5 days of culture, the cells were incubated with 1 microM of double-labelled transferrin (1251-Tf-59Fe) at 37 degrees C for 60 min. The cellular Tf-Fe and transferrin (Tf) uptake was analysed. The result showed (1) Tf uptake by the cells increased rapidly at the first 5 min, reaching its maximum after about 20 min of incubation; (2) Tf-Fe uptake kept increasing in a linear manner during the whole period of incubation; (3) the addition of either NH4Cl or CH3NH2, the blockers of Tf-Fe uptake via inhibiting iron release from Tf within endosomes, decreased the cellular Tf-Fe uptake but had no significant effect on Tf uptake; (4) trypsin and unlabelled Tf-Fe inhibited the uptake rate of Tf-Fe as well as Tf. The results suggested that Tf-Fe transport across the membrane of this type of neuron, much like other mammalian cells, was mediated by Tf-TfR endocytosis. Dysfunction of Tf or TfR would possibly lead to iron irregulation in the brain and consequently cause damage to neuronal functions.  相似文献

We have grown Ge x Si1-x (0 <x < 0.20,1000–3000Å thick) on small growth areas etched in the Si substrate. Layers were grown using both molecular beam epitaxy (MBE) at 550° C and rapid thermal chemical vapor deposition (RTCVD) at 900° C. Electron beam induced current images (EBIC) (as well as defect etches and transmission electron microscopy) show that 2800Å-thick, MBE Ge0.19Si0.81 on 70-μm-wide mesas have zerothreading and nearly zero misfit dislocations. The Ge0.19Si{0.81} grown on unpatterned, large areas is heavily dislocated. It is also evident from the images that heterogeneous nucleation of misfit dislocations is dominant in this composition range. 1000Å-thick, RTCVD Ge0.14Si0.86 films deposited on 70 μm-wide mesas are also nearly dislocation-free as shown by EBIC, whereas unpatterned areas are more heavily dislocated. Thus, despite the high growth temperatures, only heterogeneous nucleation of misfit dislocations occurs and patterning is still effective. Photoluminescence spectra from arrays of GeSi on Si mesas show that even when the interface dislocation density on the mesas is high, growth on small areas results in a lower dislocation density than growth on large areas.  相似文献

The neutron spectra transmitted across a fission-suppressed hybrid blanket and its components, driven by a low intensity 14 MeV Haefely neutron generator, were measured with a 2×2 NE213 detector at LOTUS facility. These experiments have been analyzed with 2D and 3D codes DOT3.5 and MCNP, respectively. The spectral integrals between 15 to 1 MeV show good agreement among the 2D, the 3D, and the NE213 for 15 cm lead, 18 cm beryllium, and 25 cm graphite slabs. However, there are large discrepancies for 6.2 cm stainless steel and 15 cm lithium carbonate slabs. The assemblies involving two or more of these slabs reflect these tendencies. We observe also considerable disagreement over pointwise spectra for a number of assemblies.Work done while on visiting assignment to Institut de Génie Atomique, E.P.F.L.  相似文献

This paper investigates matrix debonding from filler particles as a mechanism for weakening of a filled polymer. We analyze several simplified models of debonding. Our main concern is its effect on the overall elastic instants; for each debonding model, we calculate the change ΔW in overall strain energy density due to debonding. The resulting formulas for ΔW are all special cases of one general expression. Accordingly, we propose that this expression describes a broad class of debonding models, including some for which a detailed stress and strain analysis is not possible.  相似文献

In this paper, a new solution method for the modified eigenvalue problem with specific application to structural dynamic reanalysis is presented. The method, which is based on the block Lanczos algorithm, is developed for multiple low rank modifications to a system and calculates a few selected eigenpairs. The caterpillars of Gloveria sp. mark trails with a pheromone they deposit by dragging the ventral surface of the tip of the abdomen along branch pathways as they move between their communal nest and distance feeding sites. The threshold sensitivity of the caterpillar for an extract prepared from the secretory site was approximately 0.5 × 10–3 caterpillar equivalents/cm of trail. Bioassays show that Gloveria follows neither authentic trails of Malacosoma americanum nor artificial trails prepared from 5-cholestane-3-one, a chemical previously reported to elicit trail following from other social caterpillars. Although our observations show that fed caterpillars mark heavily as they return to their nest, we found no evidence that individual caterpillars are able to recruit hungry nestmates to new food finds. In this species, recruitment to food occurs only after many caterpillars have reinforced a trail to a newly discovered food source. In contrast, hungry caterpillars of the confamilial species M. americanum, tested under identical conditions, responded strongly to the postprandial trails of individual caterpillars and rapidly abandoned depleted sites in favor of new food finds. We postulate that the difference in the efficiency with which these two species recruit nestmates to food evolved in response to differences in the spatial distribution of their food supplies.  相似文献

In this research we examined the prediction of children’s body mass index (BMI), body weight, academic performance, social self-esteem and overall self-esteem from their IT (information technology) use, specifically, their Internet use, cell phone use and videogame playing, after controlling for the effects of gender, race, age and household income on these measures. Participants were 482 children whose average age was 12 years old. One-third was African American and two-thirds were Caucasian American. Results indicated that IT use did not predict BMI or body weight, contrary to one previous survey and the widespread belief that screen time is responsible for the obesity epidemic among our nation’s children. Instead, BMI and body weight were higher for African Americans, older children and children from lower income households. The sole and strong positive predictor of visual-spatial skills was videogame playing, which also predicted lower grade point averages (GPAs). Gender and Internet use predicted standardized test scores in reading skills. Females and children who used the Internet more had better reading skills than did males and children who used the Internet less, respectively. Implications of these findings for future research on the benefits and liabilities of IT use are discussed.  相似文献
