Two gas diffusion layers based on the same carbon cloth substrate, produced by an Italian Company (SAATI), and coated with microporous layers of different hydrophobicities, were assembled in a polymer electrolyte membrane fuel cell and its performances assessed. For comparison the cell mounting the carbon cloth without microporous layer was also tested. The membrane electrode assembly was made of Nafion® 212 with Pt load 0.3/0.6 mg cm−2 (anode/cathode). The cell testing was run at 60 °C and 80 °C with fully humidified air (100%RH) and 80%RH hydrogen feedings. The assembly of gas diffusion layers and membrane with electrodes was compressed to 30% and 50% of its initial thickness. For each configuration polarization and power curves were recorded; in order to evaluate the role of different GDLs, AC impedance spectroscopy of the running cell was also performed.The higher compression ratio caused the worsening of cell performances, partially mitigated when the operating temperature was raised to 80 °C. The presence of the microporous layer onto the carbon cloth resulted extremely beneficial for the operations especially at high current density; moreover, it sensibly reduces the high frequency resistance of the overall assembly.  相似文献

Fatigue, an overwhelming lack of physical or mental energy, is a common complaint in patients affected by multiple sclerosis (MS). Although different mechanisms have been proposed to explain MS-related fatigue, injury of distinct anatomical networks seems to be relevant in fatigue etiology. Particularly, theories point to fronto-striatal network pathological changes as a possible neural basis of fatigue. To investigate the role of fronto-striatal white matter structural alterations in fatigue perception we prospectively recruited 40 relapsing remitting patients with MS and 15 healthy controls. In patients with MS, fatigue was assessed using a validated measure, the Modified Fatigue Impact Scale (MFIS; Kos et al., 2005). Brain MRI scans were acquired for each subject enrolled with diffusion tensor imaging. Diffusion tensor data were correlated with MFIS scores using voxel-wise analysis of fractional anisotropy maps and fiber tractography algorithms. A significant cluster of voxels correlating with fatigue scores located in the deep left frontal white matter was identified. Fiber tractography revealed the cluster to be included in a complex fronto-frontal, fronto-striatal, fronto-occipital, and fronto-limbic network. Structural properties of the traced white matter fiber bundles correlated with fatigue perception and patients with clinically relevant fatigue were found to present reduced white matter integrity in the aforementioned tracts compared to those with lower levels of fatigue. Our observations show a significant involvement of different frontal networks in the pathophysiology of fatigue, thus accounting for the multifaceted nature of this disabling symptom. Tumor microenvironments are often characterized by an increase in oxidative stress levels. We studied the response to oxidative stimulation in human primary (IGR39) or metastatic (IGR37) cell lines obtained from the same patient, performing patch-clamp recordings, intracellular calcium ([Ca2+]i) imaging, and RT-qPCR gene expression analysis. In IGR39 cells, chloramine-T (Chl-T) activated large K+ currents (KROS) that were partially sensitive to tetraethylammonium (TEA). A large fraction of KROS was inhibited by paxilline—a specific inhibitor of large-conductance Ca2+-activated BK channels. The TEA-insensitive component was inhibited by senicapoc—a specific inhibitor of the Ca2+-activated KCa3.1 channel. Both BK and KCa3.1 activation were mediated by an increase in [Ca2+]i induced by Chl-T. Both KROS and [Ca2+]i increase were inhibited by ACA and clotrimazole—two different inhibitors of the calcium-permeable TRPM2 channel. Surprisingly, IGR37 cells did not exhibit current increase upon the application of Chl-T. Expression analysis confirmed that the genes encoding BK, KCa3.1, and TRPM2 are much more expressed in IGR39 than in IGR37. The potassium currents and [Ca2+]i increase observed in response to the oxidizing agent strongly suggest that these three molecular entities play a major role in the progression of melanoma. Pharmacological targeting of either of these ion channels could be a new strategy to reduce the metastatic potential of melanoma cells, and could complement classical radio- or chemotherapeutic treatments.  相似文献

During must fermentation wine strains are exposed to a variety of biotic and abiotic stresses which, when prevailing over the cellular defence systems, can affect cell viability with negative consequences on the progression of the fermentative process. To investigate the ability of wine strains to survive and adapt to unfavourable conditions of fermentation, the lipid composition, membrane integrity, cell viability and fermentative activity of three strains of Saccharomyces cerevisiae were analysed during hypoxic growth in a sugar-rich medium lacking lipid nutrients. These are stressful conditions, not unusual during must fermentation, which, by affecting lipid biosynthesis may exert a negative effect on yeast viability. The results obtained showed that the three strains were able to modulate cell lipid composition during fermentation. However, only two of them, which showed highest viability and membrane integrity at the end of the fermentation process, reached a fatty acid composition which seemed to be optimal for a successful adaptation. In particular, C16/TFA and UFA/TFA ratios, more than total lipid and ergosterol contents, seem to be involved in yeast adaptation.  相似文献

Although ceramics are considered linear elastic materials, we have observed a non‐linear pseudo‐elastic behavior in porous cellular microcracked ceramics such as β‐eucryptite. This is attributed to the evolution of microstructure in these materials. This behavior is particularly different from that of non‐microcracked ceramics such as silicon carbide. It is shown that in microcracked materials two processes, namely stiffening and softening, always compete when a compressive external load is applied. The first regime is attributed to microcrack closure, and the second to microcracks opening, i.e. to a damage introduced by the applied stress. On the other hand rather a continuous damage is observed in the non‐microcracked case. A comparison has been done between the microscopic (as measured by neutron diffraction) and the macroscopic stress‐strain response. Also, it has been found that at constant load a significant strain relaxation occurs, which has two timescales, possibly driven by the two phenomena quoted above. Indeed, no such relaxation is observed for non‐microcracked SiC. Implications of these findings are discussed.  相似文献

When do haptic estimates of discordant visual-haptic size capture vision? Observers looked at a square through a minifying lens (50%) whilst they simultaneously touched the square from below through a hand-concealing cloth. Their subsequent match of the square’s size, rendered by touching a set of comparison squares, was haptically biased when they practised estimating the square’s size (Experiment 1, N = 72), when they actively explored rather than passively touched the square (Experiment 2, N = 24), but not when they were uninformed before inspecting the square that they would estimate its size (Experiment 3, N = 36). Evidently, the haptic exploratory strategies occasioned by the practise influenced the integration of the felt size and the seen size by weighing the haptic input more than the visual input, and this weight shifting manifested itself by strengthening haptic capture. Carotenoids are well known for their nutritional properties and health promoting effects representing attractive ingredients to develop innovative functional foods, nutraceutical and pharmaceutical preparations. Pumpkin (Cucurbita spp.) flesh has an intense yellow/orange color owing to the high level of carotenoids, mainly α-carotene, β-carotene, β-cryptoxanthin, lutein and zeaxanthin. There is considerable interest in extracting carotenoids and other bioactives from pumpkin flesh. Extraction procedures able to preserve nutritional and pharmacological properties of carotenoids are essential. Conventional extraction methods, such as organic solvent extraction (CSE), have been used to extract carotenoids from plant material for a long time. In recent years, supercritical carbon dioxide (SC-CO2) extraction has received a great deal of attention because it is a green technology suitable for the extraction of lipophylic molecules and is able to give extracts of high quality and totally free from potentially toxic chemical solvents. Here, we review the results obtained so far on SC-CO2 extraction efficiency and quali-quantitative composition of carotenoids from pumpkin flesh. In particular, we consider the effects of (1) dehydration pre-treatments; (2) extraction parameters (temperature and pressure); the use of water, ethanol and olive oil singularly or in combination as entrainers or pumpkin seeds as co-matrix.  相似文献

In this article, shear stress between an aluminum tool and a carbon fiber‐epoxy prepreg is characterized during cure using polymeric release agent and release film at the tool‐part interface. The effects of surface roughness, release materials, pull‐out speed, temperature, and normal force (autoclave pressure) on the shear stress are investigated using a customized friction rig. Results show that the interfacial shear stress decreases as the temperature increases and it increases as the normal force increases when using either the release film or the release agent. Additionally, changes in surface roughness from 1.35 to 0.18 μm decrease the shear stress 10–27% while the use of release agent shows a decrease between 23% and 51% in the shear stress. Furthermore, strong adhesion between the tool and the part is observed when using release agent and pull‐out speeds of 0.05 mm/min (static/dynamic friction ratio of 5.29 ± 0.19). Using the experimental data, a mathematical approach based on the Coulomb's friction model is proposed to predict the friction force at the tool‐part interface. © 2013 Wiley Periodicals, Inc. J. Yttria partially stabilized zirconia (YSZ) thick thermal barrier coatings were fabricated by Atmospheric Plasma Spraying (APS) and isothermally annealed at 1315 °C for different durations. The phase composition of as-sprayed and heat-treated free-standing coatings was investigated by X-ray Diffraction (XRD) and the Rietveld method was employed for quantitative phase analysis. High-temperature exposure of YSZ coatings produced the partial decomposition of metastable t′ zirconia phase and the corresponding increase in the amount of stable tetragonal t, cubic c and monoclinic m phases with increasing the aging time.  相似文献
