Targeting tumor vasculature through specific endothelial cell markers represents a promising approach for cancer treatment. Here our aim was to construct an antibiotic resistance gene-free plasmid encoding shRNAs to simultaneously target two endothelial cell markers, CD105 and CD146, and to test its functionality and therapeutic potential in vitro when delivered by gene electrotransfer (GET) and combined with irradiation (IR). Functionality of the plasmid was evaluated by determining the silencing of the targeted genes using qRT-PCR. Antiproliferative and antiangiogenic effects were determined by the cytotoxicity assay tube formation assay and wound healing assay in murine endothelial cells 2H-11. The functionality of the plasmid construct was also evaluated in malignant melanoma tumor cell line B16F10. Additionally, potential activation of immune response was measured by induction of DNA sensor STING and proinflammatory cytokines by qRT-PCR in endothelial cells 2H-11. We demonstrated that the plasmid construction was successful and can efficiently silence the expression of the two targeted genes. As a consequence of silencing, reduced migration rate and angiogenic potential was confirmed in 2H-11 endothelial cells. Furthermore, induction of DNA sensor STING and proinflammatory cytokines were determined, which could add to the therapeutic effectiveness when used in vivo. To conclude, we successfully constructed a novel plasmid DNA with two shRNAs, which holds a great promise for further in vivo testing.  相似文献

Immersion tests with different stainless steels have been performed, while the pH was stepwise decreased and then increased again. During 8.5-day exposure, the depassivation and repassivation pH values as a function of pitting resistance equivalent number were determined. There is always a gap between both pH values (depassivation and repassivation), indicating that for every steel, there are conditions where an existing passive layer can be maintained but cannot be rebuilt after depassivation. In such environments, the passive layer is thicker, consisting mainly of molybdenum and iron rich oxides, while chromium is dissolved. Usually, depending on conditions, the passive layer is more chromium-rich, especially the inner layer. This is relevant, for example, for acidizing jobs in oil and gas industry, proving that repassivation after acidizing will happen promptly, when the pH is increased again.  相似文献

The influence of orientation of a discharge channel in water and the length of a cylindrical discharge chamber on the kinematic characteristics of the maximum deflection of plates during molding into an open ring matrix have been studied experimentally. The optical method is used to measure the deflection of the plate deformed by pressure waves generated by an electric discharge in water. No significant effect of the discharge channel orientation and relative length of the cylindrical discharge chamber on the plate deflection is detected.  相似文献

In this paper, the applicability of the ultrasonic wave transmission method to estimate the initial setting time of an arbitrary cement paste is discussed. Ultrasonic pulse velocity measurements were fully automated and measured continuously. The Vicat Needle Test was used in order to determine the initial setting time of cement pastes. Different cement pastes were prepared in order to check the influence of the water/cement ratio, type of cement, curing temperature, cement fineness, and some clinker compositions, on the relationship between the initial setting time and ultrasonic pulse velocity. It was found that the initial setting time of an arbitrary cement paste can be estimated very accurately by the time the first inflection point appears on the ultrasonic pulse velocity curve. Moreover, it can be estimated quite accurately by the time the ultrasonic pulse velocity reaches a fixed value, close to the value of the ultrasonic pulse velocity in water.  相似文献

Magnetic magnesium alloys can be utilized as a load sensitive material, in which the inverse magnetostrictive effect is used in order to measure the actual loads in structural components manufactured from such lightweight sensor alloys. To achieve a material which exhibits magnetic properties, Mg is alloyed with ferromagnetic materials like cobalt or samarium-cobalt. Alloying elements commonly used with Mg are utilized to improve the mechanical properties of these alloys, which however may have a slight negative impact on the magnetic sensitivity. In this work, two separate magnetic Mg alloys are compared, each with properties matched to the opposing requirements: (a) high load sensitivity and (b) satisfactory mechanical properties, respectively. The precipitation behavior of the ferromagnetic constituent Co in Mg together with other alloying elements is shown on the basis of SEM images. In addition, the dissolving behavior of the Co powder during the casting process of a binary Mg–Co alloy is investigated. Cyclic loading tests employing harmonic analyses of eddy current signals are utilized in order to verify the alloys’ sensory properties. The mechanical properties are investigated using tensile tests.  相似文献

A planar concentration analysis (PCA) system is used for observing the transport and mixing of a tracer mass in a shallow turbulent free-surface wake flow of a large cylindrical obstacle. The nonintrusive, fieldwise PCA measuring technique is applied to evaluate depth-averaged mass concentrations by making use of light attenuation due to absorption and scattering processes related to a dissolved tracer mass. The scalar fields are decomposed into a low-frequency quasiperiodic part, the coherent flow, and a randomly fluctuating part. From accompanying near-surface velocity measurements, large-scale coherent structures are identified and related to the coherent mass fields. This allows one to assess the role of the large-scale vortices for advection and diffusion in shallow wake flows. The time–mean wake flow displays a self-similar spanwise distribution both for mass and velocity. The longitudinal development of shallow wakes initially shows the growth of unbounded wakes; in the wake far field an attenuated behavior applies.  相似文献

The aim of this work was to investigate the compressibility behavior of alpha-methyldopa and phenobarbitone using a Korsch EK0 instrumented eccentric tablet machine, with force-time and force-displacement curves constructed and applied to calculate different compressional values to study the compressional behavior. The results of this work revealed a difference in compressibility behavior between the two drugs during the compressional process. alpha-Methyldopa gave an abnormal compressional curve with high friction in the pre- and postcompressional phases. A residual force could be seen on the lower punch. Furthermore, capping and sticking were observed visually during tablet pressing, indicating poor compressibility behavior. In the case of phenobarbitone, no friction was observed in the precompressional phase, but there was higher friction in the postcompressional phase, especially in the ejection phase. The compressibility of the drugs was improved by the addition of Avicel PH-301 and magnesium stearate.  相似文献

GRA4, a dense granule protein of Toxoplasma gondii elicits both mucosal and systemic immune responses following oral infection of mice with cysts. We studied the antigenicity and immunogenicity of truncated and soluble forms of GRA4 expressed as glutathione S-transferase fusion proteins in Escherichia coli. Protein C (amino-acids 297-345) was particularly well recognized by serum IgG antibodies, milk IgA antibodies and intestinal IgA antibodies from T. gondii infected mice and by serum IgG antibodies from T. gondii infected humans and T. gondii infected sheep. One major B epitope was localized within the last 11 C-terminal residues of GRA4. A second epitope, recognized with lower frequency, was mapped within the region 318-334. In contrast, the N domain of GRA4 (amino acids 25-276) was poorly recognized. Oral immunization of C57BL/6 mice with N, C or NC (amino acids 25-276 fused to 297-345) in association with cholera toxin induced a significant production of serum anti-GRA4 IgG antibodies but a weak and inconsistent intestinal anti-GRA4 IgG antibody response and afforded partial resistance to oral infection with T. gondii. These results provide new molecular and immunological understanding of GRA4 and indicate that it is a potential candidate for oral vaccination against T. gondii.  相似文献

It has been reported that blood trombopoietin (TPO) levels can discriminate between thrombocytopenia due to increased platelet destruction and decreased platelet production. With our TPO ELISA and a glycocalicin ELISA we analysed a large group of patients in detail and could confirm and amplify the above notion in detail. TPO levels were determined in plasma from 178 clinically and serologically well-defined thrombocytopenic patients: 72 patients with idiopathic autoimmune thrombocytopenia (AITP), 29 patients with secondary AITP, 5 patients with amegakaryocytic thrombocytopenia and 72 patients who suffered from various diseases (46 in whom megakaryocyte deficiency was not and 26 in whom it was expected). In addition, we measured the level of glycocalicin as a marker of total body mass of platelets. In all patients with primary AITP and secondary AITP, TPO levels were within the normal range or in some (n = 7) cases only slightly increased. The level of glycocalicin was not significantly different from that of the controls (n = 95). The patients with amegakaryocytic thrombocytopenia had strongly elevated TPO levels and significantly decreased glycocalicin levels. Similarly, among the 72 thrombocytopenic patients with various disorders, elevated TPO levels were only found in patients in whom platelet production was depressed. The mean level of glycocalicin in these patients was decreased compared to that in controls and patients with AITP, but was not as low as in patients with amegakaryocytic thrombocytopenia. In conclusion, all patients with depressed platelet production had elevated levels of circulating TPO, whereas the TPO levels in patients with an immune-mediated thrombocytopenia were mostly within the normal range. Therefore, measurement of plasma TPO levels provides valuable diagnostic information for the analysis of thrombocytopenia in general. Moreover, treatment with TPO may be an option in AITP.  相似文献
