The present study deals with the short-term physicochemical reactions at the interface between bioactive glass particles [55SiO(2)-20CaO-9P(2)O(5)-12Na(2)O-4MgO. mol%] and biological fluid (Dulbecco Modified Eagle's Medium (DMEM)). The physicochemical reactions within the interface are characterized by scanning transmission electron microscopy (TEM) (STEM) associated with Energy-dispersive X-ray spectroscopy (EDXS). Microanalysis of diffusible ions such as sodium, potassium, or oxygen requires a special care. In the present investigation the cryo-technique was adopted as a suitable tool for the specimen preparation and characterization. Cryosectioning is essential for preserving the native distribution of ions so that meaningful information about the local concentrations can be obtained by elemental microanalysis. The bioglass particles immersed in biological fluid for 24 h revealed five reaction stages: (i) dealkalization of the surface by cationic exchange (Na(+), Ca(2+) with H(+) or H(3)O(+)); (ii) loss of soluble silica in the form of Si(OH)(4) to the solution resulting from the breakdown of Si--O--Si bonds (iii); repolymerization of Si(OH)(4) leading to condensation of SiO(2)); (iv) migration of Ca(2+) and PO(4) (3-) to the surface through the SiO(2)-rich layer to form CaO-P(2)O(5) film; (v) crystallization of the amorphous CaO-P(2)O(5) by incorporating OH-- or CO(3) (2-) anions with the formation of three different surface layers on the bioactive glass periphery. The thickness of each layer is approximately 300 nm and from the inner part to the periphery they consist of Si--OH, which permits the diffusion of Ca(2+) and PO(4) (3-) ions and the formation of the middle Ca--P layer, and finally the outer layer composed of Na--O, which acts as an ion exchange layer between Na(+) ions and H(+) or H(3)O(+) from the solution.  相似文献

We report on experiments performed in vacuum and at cryogenic temperatures on a tri-port nano-electro-mechanical (NEMS) device. One port is a very nonlinear capacitive actuation, while the two others implement the magnetomotive scheme with a linear input force port and a (quasi-linear) output velocity port. We present an experimental method enabling a full characterization of the nanomechanical device harmonic response: the nonlinear capacitance function C(x) is derived, and the normal parameters k and m (spring constant and mass) of the mode under study are measured through a careful definition of the motion (in meters) and of the applied forces (in Newtons). These results are obtained with a series of purely electric measurements performed without disconnecting/reconnecting the device, and rely only on known dc properties of the circuit, making use of a thermometric property of the oscillator itself: we use the Young modulus of the coating metal as a thermometer, and the resistivity for Joule heating. The setup requires only three connecting lines without any particular matching, enabling the preservation of a high impedance NEMS environment even at MHz frequencies. The experimental data are fit to a detailed electrical and thermal model of the NEMS device, demonstrating a complete understanding of its dynamics. These methods are quite general and can be adapted (as a whole, or in parts) to a large variety of electromechanical devices.  相似文献

An indenter-type high-pressure cell has been developed for electric and magnetic measurements in low-temperature and high-magnetic-field environments. The maximum pressure achieved at low temperatures is more than 4.5 GPa, which is higher than that of a conventional piston-cylinder cell. The typical sample space at maximum pressure is 1.6 mm in diameter and approximately 0.7 mm in depth, and magnetic measurements such as ac-susceptibility and nuclear magnetic resonance can be performed using a miniature coil. All the components of the indenter cell are made of nonmagnetic materials that have enough thermal conductivity for low-temperature experiments using a 3He/4He dilution refrigerator. Another indenter-type cell designed for a commercial superconducting quantum interference device magnetometer is also reported.  相似文献

Filopodia are rod-shaped cell surface protrusions composed of a parallel bundle of actin filaments. Since filopodia frequently emanate from lamellipodia, it has been proposed that they form exclusively by the convergence and elongation of actin filaments generated in lamellipodia networks. However, filopodia form without Arp2/3-complex, which is essential for lamellipodia formation, indicating that actin filaments in filopodia may be generated by other nucleators. Here we analyzed the effects of ectopic expression of GFP-tagged full length or a constitutively active variant of the human formin mDia2/Drf3. By contrast to the full-length molecule, which did not affect cell behaviour and was entirely cytosolic, active Drf3 lacking the C-terminal regulatory region (Drf3DeltaDAD) induced the formation of filopodia and accumulated at their tips. Low expression of Drf3DeltaDAD induced rod-shaped or tapered filopodia, whereas over-expression resulted in multiple, club-shaped filopodia. The clubs were filled with densely bundled actin filaments, whose number but not packing density decreased further away from the tip. Interestingly, clubs frequently increased in width after protrusion beyond the cell periphery, which correlated with increased amounts of Drf3DeltaDAD at their tips. These data suggest Drf3-induced filopodia form and extend by de novo nucleation of actin filaments instead of convergent elongation. Finally, Drf3DeltaDAD also induced the formation of unusual, lamellipodia-like structures, which contained both lamellipodial markers and the prominent filopodial protein fascin. Microarray analyses revealed highly variable Drf3 expression levels in different commonly used cell lines, reflecting the need for more detailed analyses of the functions of distinct formins in actin cytoskeleton turnover and different cell types.  相似文献

We developed a dual-probe (DP) atomic force microscopy (AFM) system that has two independently controlled probes. The deflection of each cantilever is measured by the optical beam deflection (OBD) method. In order to keep a large space over the two probes for an objective lens with a large numerical aperture, we employed the OBD sensors with obliquely incident laser beams. In this paper, we describe the details of our developed DP-AFM system, including analysis of the sensitivity of the OBD sensor for detection of the cantilever deflection. We also describe a method to eliminate the crosstalk caused by the vertical translation of the cantilever. In addition, we demonstrate simultaneous topographic imaging of a test sample by the two probes and surface potential measurement on an α-sexithiophene (α-6T) thin film by one probe while electrical charges were injected by the other probe.  相似文献

The in situ calibration method for the impurity influx monitor (divertor) is experimentally examined. The total reflectance of the optical path from the focal point of the Cassegrain telescope to the first mirror is derived using a micro retroreflector array. An optical fiber with angled physical contact (APC) connectors reduces the return edge reflection. APC fibers and a multimode coupler increase the signal-to-noise ratio by about one order compared to that of triple-branched fibers and enable measurement of the wavelength dependence of the total reflectance of the optical system even after potential deterioration of mirror surfaces reduces reflectance.  相似文献
