In microbiological research, it is important to understand the time course of each step in a pathogen’s lifecycle and changes in the host cell environment induced by infection. This study is the first to develop a real-time monitoring system that kinetically detects luminescence reporter activity over time without sampling cells or culture supernatants for analyzing the virus replication. Subgenomic replicon experiments with hepatitis C virus (HCV) showed that transient translation and genome replication can be detected separately, with the first peak of translation observed at 3–4 h and replication beginning around 20 h after viral RNA introduction into cells. From the bioluminescence data set measured every 30 min (48 measurements per day), the initial rates of translation and replication were calculated, and their capacity levels were expressed as the sums of the measured signals in each process, which correspond to the areas on the kinetics graphs. The comparison of various HuH-7-derived cell lines showed that the bioluminescence profile differs among cell lines, suggesting that both translation and replication capacities potentially influence differences in HCV susceptibility. The effects of RNA mutations within the 5′ UTR of the replicon on viral translation and replication were further analyzed in the system developed, confirming that mutations to the miR-122 binding sites primarily reduce replication activity rather than translation. The newly developed real-time monitoring system should be applied to the studies of various viruses and contribute to the analysis of transitions and progression of each process of their life cycle. 相似文献

For the past few years, hybrid generation systems including solar panel and gas cogeneration have been used for residential houses. Solar panels can generate electronic power at daytime but not at night. But the power consumption of residential houses usually peaks in the evening. The gas engine cogeneration system can generate electronic power without such a restriction, and it also can generate heat power to warm up a house or to produce hot water. In this paper we propose a solar panel and gas engine cogeneration hybrid system with an energy storage device, combined by a DC bus. If a blackout occurs, the system still can supply electronic power for special house loads. We propose a control scheme for the system related to the charging level of the energy storage device and the voltage of the utility grid, which can be applied to both grid‐connected and standalone operation. The accelerator-driven transmutation system has been studied at the Japan Atomic Energy Research Institute. This system is a hydrid system which consists of a high intensity accelerator, a spallation target and a subcritical core region. In the conceptual design study, two types of system concepts, sodium cooled and lead-bismuth cooled system, are being studied. In this study, we fucus on our lead-bismuth cooled accelerator-driven transmutation system to investigate basic characteristics. Numerical results of three-dimensional separated flow and heat transfer in a rectangular channel with a suddenexpansion are presented in this paper.Numerical simulations of Navier-Stokes and energy equations are carriedout using the finite difference method.The results of three-dimensional calculations are compared with thetwo-dimensional ones,and effects of the aspect ratio of channel upon the flow are shown.The transition fromsymmetric to asymmetric flow appears at lower Reynolds number as increasing the aspect ratio.The details oflocal heat transfer characteristics in two different separated flow regions on two downstream walls are clarified.Two-dimensionality of the flow and heat transfer almost disappears for the aspect ratio considered. 相似文献

Osteoporosis is a common bone disease, particularly in menopausal women. Herein, we screened four Kampo medicines (Unkeito (UKT), Kamishoyosan (KSS), Kamikihito (KKT), and Ninjinyoeito (NYT)), frequently used to treat menopausal syndromes, for their effects on receptor activator of nuclear factor-kappaB ligand (RANKL)-induced osteoclast differentiation in RAW 264 cells. Considering that UKT exhibited the most potent effect, we examined its effect on RANKL-induced osteoclastogenesis, the induction of osteoclast apoptosis, and the mechanisms underlying its effects. UKT inhibits RANKL-induced osteoclast differentiation in the early stage and decreases osteoclast-related genes, including tartrate-resistant acid phosphatase (Trap), dendritic cell-specific transmembrane protein (Dcstamp), matrix metalloproteinase-9 (Mmp9), and cathepsin K (Ctsk). Specifically, UKT inhibits the nuclear factor of activated T cells 1 (NFATc1), which is essential for osteoclastogenesis. UKT increases Bcl6, which antagonizes NFATc1 and Dc-stamp, thereby blocking the progression of osteoclasts to maturation. UKT also decreased nuclear translocation by downregulating the activity of p65/NF-κB. In addition, UKT enhances mononuclear osteoclast apoptosis via activation of caspase-3. Herein, we demonstrate that UKT suppresses RANKL-mediated osteoclastogenesis via the Blimp1–Bcl6 and NF-κB signaling pathways and enhances mononuclear osteoclast apoptosis. Furthermore, UKT prevents bone loss in OVX mice. Thus, UKT might be a potential therapeutic agent for postmenopausal osteoporosis. 相似文献

Aberrant glycosylation of IgA1 is involved in the development of IgA nephropathy (IgAN). There are many reports of IgAN markers focusing on the glycoform of IgA1. None have been clinically applied as a routine test. In this study, we established an automated sandwich immunoassay system for detecting aberrant glycosylated IgA1, using Wisteria floribunda agglutinin (WFA) and anti-IgA1 monoclonal antibody. The diagnostic performance as an IgAN marker was evaluated. The usefulness of WFA for immunoassays was investigated by lectin microarray. A reliable standard for quantitative immunoassay measurements was designed by modifying a purified IgA1 substrate. A validation study using multiple serum specimens was performed using the established WFA-antibody sandwich automated immunoassay. Lectin microarray results showed that WFA specifically recognized N-glycans of agglutinated IgA1 in IgAN patients. The constructed IgA1 standard exhibited a wide dynamic range and high reactivity. In the validation study, serum WFA-reactive IgA1 (WFA+-IgA1) differed significantly between healthy control subjects and IgAN patients. The findings indicate that WFA is a suitable lectin that specifically targets abnormal agglutinated IgA1 in serum. ABSTRACTThis paper proposes a novel, simultaneous bipedal locomotion method using haptics for remote operation of biped robots. In general, traditional biped walking methods require very high computational power and advanced controllers to perform the required task. However, in this proposed method, a master exoskeleton attached to the human’s lower body is used to obtain the trajectory and haptic information to generate the trajectory of the slave biped robot in real time. Lateral motion of the center of mass of the biped is constrained in this experiment. Also, it is considered that no communication delay is presented in between the two systems in this experiment, and they are not discussed in this paper. Since a direct motion transmission is used in the proposed method, this method is quite straight forward and a simultaneous walking can be realized at the same time with high performance. Also, it does not require an exact dynamic model of the biped or specific method to plan the trajectory. The gait pattern of the biped is directly determined by that of the human. Also, the operator can feel the remote environment through the exoskeleton robot. Results obtained from the experiments validate the proposed method. 相似文献

Blends of polylactide (PLA) and recycled polypropylene (rPP) were prepared by melt-processing using a corotating twin-screw extruder and subsequent pelletizing of the extrudates for injection molding. The PLA/rPP blends were characterized by Thermogravimetric Analysis (TGA), Differential Scanning Calorimetry (DSC), Dynamic Mechanical Analysis (DMA), rheometer (MCR-102), scanning electron microscopy(SEM), tensile tests, and impact measurements. The results indicate that the PLA/rPP blend is immiscible and has a two-phase structure. TGA revealed enhancement of the thermal stability of the blends upon addition of rPP. The storage modulus, loss modulus, and complex viscosity of the blends increased with rPP concentration. Mechanical studies showed that introduction of rPP results in a decrease in tensile strength and modulus and enhancement of the impact strength of PLA in the blends. The effects of a silane coupling agent on the morphology and on the tensile and impact properties of the rPP blends of silane-modified PLA were also examined. SEM studies suggest that silane is an effective interfacial modifier. Thus, better interfacial adhesion was observed with silane-modified blends as compared with unmodified blends. Silane also improved the mechanical properties of the modified blends. The blends reached maximum tensile strength at 1.5 wt.% silane (relative to modified PLA content), and impact strength increased with increasing silane concentration.
