This study presents a three‐dimensional numerical analysis of the effect of boundary layer thickness on vortex structures and heat transfer behind a hill mounted in a laminar boundary layer. When the thickness of the velocity boundary layer is comparable to the hill height, a hairpin vortex is formed symmetrically to the center of the spanwise direction in the wake. A secondary vortex is formed between the legs, and horn‐shaped secondary vortices appear under the concave parts of the hairpin vortex. When the boundary layer thickness increases, the legs and horn‐shaped secondary vortices move toward the center of the spanwise direction, and thus heat transport and heat transfer increase there. At this time, high‐turbulence areas generated locally move toward the center of the spanwise direction with an increase in the boundary layer thickness. With a further increase in the boundary layer thickness, steady streamwise vortices are formed downstream of the hill, but the heat transfer decreases. © 2009 Wiley Periodicals, Inc. By 2012, Japan must cut down on its annual emissions of greenhouse gases by 6% from 1990 levels. However, greenhouse emissions increased by 6.4% in 2006. More effective responses to reduce greenhouse gases are required. Attention is currently focused on increasing the use of renewable energy, and wind energy has received a lot of attention. The national target for wind power capacity in the year 2010 is 3000 MW; however, there are many barriers to the development of wind energy. Japan's climate differs from that of the European Union countries. It often experiences typhoons and lightning strikes because of its meteorological characteristics. Wind has a stronger turbulence level due to the complex terrain. Furthermore, power fluctuation of wind causes power system problems because of issues related to the grid connection. Many endeavours have been made to find the best solutions for these problems. The degradation in performance of proton-exchange membrane fuel cells (PEMFCs) under open circuit conditions was investigated. The oxygen reduction reaction (ORR) kinetic current density at 0.9 V was found to decrease from 36 to 4 mA cm−2 (geometric) without significant crossover increase or loss in the electrochemically active surface area. Cyclic voltammograms for the electrodes show characteristic changes, e.g. appearance of peaks at ∼0.2 V and shift of the onset of platinum oxide formation to higher potentials. It was identified that the large ORR kinetic decay has its origins in the reduction of available Pt sites due to adsorption of anions, which are postulated to be membrane decomposition products such as sulfate ions. Procedures carried out to condense water in the fuel cell led to the expulsion of anions out of the membrane electrode assembly (MEA) resulting in the partial recovery of ORR kinetic current density to 15 mA cm−2. In order to attain complete performance recovery of the catalyst, a more effective and practical method to flush out the anions is desirable.  相似文献

The enzyme that catalyzes N-acyl linkage between myristic acid and the NH(2)-terminal glycine residue of the octapeptide Gly-Asn-Ala-Ala-Ala-Ala-Arg-Arg-NH(2) in aqueous solution without ATP and coenzyme A was found in Pseudomonas aeruginosa. The enzyme was purified from cell-free crude extract using DEAE-Cellulose, Sephadex G-200, CM-Sephadex C-50, and hydroxyapatite column chromatographies, and then purified approximately 1900-fold with about 1.5% recovery of enzyme activity from the crude extract. Finally, the purified enzyme showed a main band on SDS polyacrylamide gel electrophoresis after staining with Coomassie Brilliant Blue. The band corresponded to a molecular mass of approximately 60 kDa. The K(m)s of the purified enzyme for the substrate myristic acid and the octapeptide were 0.36 and 2.6 mM, respectively. When myristoyl-CoA instead of myristic acid was used as the substrate for the enzyme reaction, myristoyl octapeptide could be synthesized as observed in the case of myristic acid. The K(m) of myristoyl-CoA was 0.17 mM.  相似文献

A mesophilic, mixotrophic iron-oxidizing bacterium strain OKM-9 uses ferrous iron as a sole source of energy and L-glutamate as a sole source of cellular carbon. Uptake of L-glutamate into OKM-9 cells is absolutely dependent on ferrous iron oxidation. Thus, the Fe(2+)-dependent L-glutamate uptake system of strain OKM-9 is crucial for the bacterium to grow mixotrophically in iron medium with L-glutamate. The relationship between iron oxidation and L-glutamate transport activities was studied. Iron oxidase containing cytochrome a was purified 9-fold from the plasma membrane of OKM-9. A purified iron oxidase showed one rust-colored band following disc gel electrophoresis after incubation with Fe(2+). The Fe(2+)-dependent L-glutamate transport system was also purified 14.5-fold from the plasma membrane using the same purification steps as for iron oxidase. Fe(2+)-dependent L-glutamate and L-cysteine uptake activities of OKM-9 were 0.36 and 0.24 nmol/mg/min, respectively, when a concentration of 18 mM of these amino acids was used as a substrate. Both uptake activities were completely inhibited by potassium cyanide (KCN), suggesting that cytochrome a in the iron oxidase is involved in the transport process. The iron-oxidizing activity of strain OKM-9 was activated 1.7-fold by 80 mM L-glutamate. In contrast, the activity was noncompetitively inhibited by L-cysteine. The Michaelis constant of iron oxidase for Fe(2+) was 12.6 mM and the inhibition constant for L-cysteine was 41.6 mM. A marked inhibition of iron oxidase by 50 mM L-cysteine was completely reversed by the addition of 60 mM L-glutamate. The results suggest the possibility that iron oxidase has a binding site for L-cysteine and the cysteine first bound to the iron oxidase was replaced by the added L-glutamate.  相似文献

To examine dioxin contamination in commercial baby foods in Japan, congener analyses of polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs) and coplanar polychlorinated biphenyls (Co-PCBs) were performed on 102 varieties of baby foods obtained from supermarkets in 2001-2002. The toxic equivalent quantity (TEQ) levels for dioxins in samples ranged from < 0.001 to 0.135 pg-TEQ/g wet weight when undetected or trace levels of congeners were taken as zero. Among 102 samples tested, 26 samples exceeded 0.010 pg-TEQ/g. The highest TEQ value was for "sardine, vegetables" (0.135 pg-TEQ/g), followed by "Japanese radish (daikon), sardine" (0.080 pg-TEQ/g). Thus, dioxins were detected at low levels in baby foods containing animal products such as fishes and/or dairy products.  相似文献

A laser-guided BTA tool has been developed to prevent axial hole deviations. Four kinds of experiments were conducted to examine the performance of this tool in detail and to determine its practical application using a duralumin (A2017-T4) workpiece with a prebored 108-mm diameter hole. The first experiment was performed to examine the conditions of the hole deviation in the case of normal deep-hole boring. The second was to examine whether the tool corrects for the stated hole deviation. The third was to examine whether the tool can go straight through to the target without shifting toward a thin wall (t = 0.7 mm) on one side of the workpiece. The fourth was to examine whether the tool can be guided toward the target without affecting a prebored hole, which was inclined to radial direction at 100 μm for a hole length of 100 mm. The experiments were performed with a rotating tool-stationary workpiece system. Rotational speed was 270 rpm and feed ws 0.125 mm/rev. Tool diameter was 110 mm. The results show that the tool can be guided to go straight through toward the target despite the disturbances.  相似文献

The feasibility of applying a polyethylene (PE) sponge as a biomass carrier in an anaerobic ammonium oxidation (anammox) reactor and its nitrogen removal performance were also investigated. Experiments were carried out in an up-flow column reactor with synthetic inorganic wastewater. Experimental results indicate that reactor containing PE sponge biomass carriers showed a high nitrogen removal capability and exhibited stable performance. In addition, the reactor with 8 strips PE sponge as biomass carrier exhibited greater adaptation capacity compared to that with 6 strips and could achieve a high TN removal rate within a very short period. The ratio of NO2-N removal and NO3-N production to NH4-N removal for the reactor was 1.26:0.21. Furthermore, to investigate the bacterial composition of the mature community, 16S rRNA sequences were amplified by PCR and analyses were conducted using DNA databases. Results showed that a new kind of anammox microorganism (Kumadai-1) was the dominant species in the reactor when using PE sponge as a biomass carrier.  相似文献

A simple capillary electrophoretic method was developed for the determination of glucosamine using in-capillary derivatisation. Glucosamine in commercial products was extracted with purified water. The CE separation was achieved on an uncoated fused-silica capillary using a 20 mM borate buffer (pH 9.2) containing 5 mM o-phthalaldehyde (OPA) and 5 mM 3-mercaptopropionic acid (MPA) at 25 kV, followed by UV detection at 340 nm. The detector response was linear (r2 > 0.999) in the concentration range 10–1000 μg/mL. The limit of detection (LOD) was 1.3 mg/g. Spiked glucosamine recoveries at 50 and 100 mg/g level were 95.1% and 104.3%, respectively. The method was applied to 16 commercial products. The concentrations of glucosamine were 109–705 mg/g, and the ratios of detected glucosamine content to the labelled value were 88.8–124%. No significant bias was observed (r2 = 0.989, p < 0.01), between results obtained by the proposed CE method and an official colorimetric method (Japanese Health Food & Nutrition Food Association).  相似文献
