HDAC6 is overexpressed in ovarian cancer and is known to be correlated with tumorigenesis. Accordingly, ACY-241, a selective HDAC6 inhibitor, is currently under clinical trial and has been tested in combination with various drugs. HDAC8, another member of the HDAC family, has recently gained attention as a novel target for cancer therapy. Here, we evaluated the synergistic anticancer effects of PCI-34051 and ACY-241 in ovarian cancer. Among various ovarian cancer cells, PCI-34051 effectively suppresses cell proliferation in wild-type p53 ovarian cancer cells compared with mutant p53 ovarian cancer cells. In ovarian cancer cells harboring wild-type p53, PCI-34051 in combination with ACY-241 synergistically represses cell proliferation, enhances apoptosis, and suppresses cell migration. The expression of pro-apoptotic proteins is synergistically upregulated, whereas the expressions of anti-apoptotic proteins and metastasis-associated proteins are significantly downregulated in combination treatment. Furthermore, the level of acetyl-p53 at K381 is synergistically upregulated upon combination treatment. Overall, co-inhibition of HDAC6 and HDAC8 through selective inhibitors synergistically suppresses cancer cell proliferation and metastasis in p53 wild-type ovarian cancer cells. These results suggest a novel approach to treating ovarian cancer patients and the therapeutic potential in developing HDAC6/8 dual inhibitors.  相似文献

Manganese oxide nanocrystals are combined with aluminum oxide nanocrystals to improve their crystallinity via calcination without a significant increase of crystal size. A nanocomposite, consisting of two metal oxides, can be synthesized by the reaction between permanganate anions and aluminum oxyhydroxide keggin cations. The as‐prepared manganese oxide–aluminum oxide nanocomposite is X‐ray amorphous whereas heat‐treatment gives rise to the crystallization of an α‐MnO2 phase at 600 °C and Mn3O4/Mn2O3 and γ‐Al2O3 phases at 800 °C. Electron microscopy and N2 adsorption‐desorption‐isotherm analysis clearly demonstrate that the as‐prepared nanocomposite is composed of a porous assembly of monodisperse primary particles with a size of ~20 nm and a surface area of >410 m2 g?1. Of particular interest is that the small particle size of the as‐prepared nanocomposite is well‐maintained up to 600 °C, a result of the prevention of the growth of manganate grains through nanoscale mixing with alumina grains. The calcined nanocomposite shows very‐high catalytic activity for the oxidation of cyclohexene with an extremely high conversion efficiency of >95% within 15 min. The present results show that the improvement of the crystallinity without significant crystal growth is very crucial for optimizing the catalytic activity of manganese oxide nanocrystals.  相似文献

Modern industries require the production of multi-functional, inorganic, micron-sized metal wires. This study suggests a novel method that could potentially offer a highly efficient dieless drawing technology for manufacturing thin stainless steel fibers. The method is based on a hot-working principle, using microwaves as the heat source and SiC as the susceptor. Experimental trials with a laboratory rig showed that the new system worked effectively for drawing the stainless steel wires and should be able to realize the diameter attenuation with a diameter reduction of up to 21%. The theoretical model describing the deformation behavior of the stainless steel wires in the working zone along with the constitutive equation of Bingham model modified with a power law and Zener–Hollomon parameter turned out to match very good with the actual results of the experiment. The coefficient of variation of the drawn wire diameter increased, as the draw ratio increased, which could be attributed to the occurrence of the narrow necking zone.  相似文献

Nanofabrication is an indispensable process in nanoscience and nanotechnology. Unconventional lithographic techniques are often used for fabrication as alternatives to photolithography because they are faster, more cost‐effective, and simpler to use. However, these techniques are limited in scalability and utility because of the collapse of preprinted structures during step‐and‐repeat processes. This study proposes a new class of temperature‐controllable polymeric molds that are coated with a metal such that any site‐specific patterning can be accomplished in a programmable manner using selective contact‐dewetting lithography. The lithography allows sub‐100 nm patterning, step‐and‐repeat processing, and hierarchical structure fabrication. The programmable feature of the lithography can be utilized for the structural coloring and shaping of objects. Large‐area programmable patterning, semiconductor device manufacturing, and the fabrication of iridescent security devices would benefit from the unique features of the proposed strategy.  相似文献

Cross-layer design breaks away from traditional network design where each layer of the protocol stack operates independently. We explore the potential synergies of exchanging information between different layers to support real-time video streaming. In this new approach information is exchanged between different layers of the protocol stack, and end-to-end performance is optimized by adapting to this information at each protocol layer. We discuss key parameters used in the cross-layer information exchange along with the associated cross-layer adaptation. Substantial performance gains through this cross-layer design are demonstrated for video streaming.  相似文献

This paper proposes and demonstrates optical 3R regeneration techniques for high-performance and scalable 10-Gb/s transmission systems. The 3R structures rely on monolithically integrated all-active semiconductor optical amplifier-based Mach-Zehnder interferometers (SOA-MZIs) for signal reshaping and optical narrowband filtering using a Fabry-Peacuterot filter (FPF) for all-optical clock recovery. The experimental results indicate very stable operation and superior cascadability of the proposed optical 3R structure, allowing error-free and low-penalty 10-Gb/s [pseudorandom bit sequence (PRBS) 223-1 ] return-to-zero (RZ) transmission through a record distance of 1 250 000 km using 10 000 optical 3R stages. Clock-enhancement techniques using a SOA-MZI are then proposed to accommodate the clock performance degradations that arise from dispersion uncompensated transmission. Leveraging such clock-enhancement techniques, we experimentally demonstrate error-free 125 000-km RZ dispersion uncompensated transmission at 10 Gb/s (PRBS 223-1) using 1000 stages of optical 3R regenerators spaced by 125-km large-effective-area fiber spans. To evaluate the proposed optical 3R structures in a relatively realistic environment and to investigate the tradeoff between the cascadability and the spacing of the optical 3R, a fiber recirculation loop is set up with 264- and 462-km deployed fiber. The field-trial experiment achieves error-free 10-Gb/s RZ transmission using PRBS 223-1 through 264 000-km deployed fiber across 1000 stages of optical 3R regenerators spaced by 264-km spans  相似文献

AlxGa1-xN (x=0.05) ultraviolet (UV) avalanche photodiodes grown on a GaN substrate are reported. The epitaxial structure was grown by metal-organic chemical vapor deposition on a free-standing bulk GaN substrate having low dislocation density. The growth conditions for AlxGa1-xN epitaxial layers on GaN substrates were optimized to achieve improved crystalline and structural quality. With UV illumination at lambda~250 nm, devices with mesa diameters of ~30 mum achieve stable maximum optical gains of ~50 at a reverse bias voltage of ~87 V.  相似文献
