An exercise design approach to understanding assessment center dimension and exercise constructs.

Arobust finding in assessment center research is that ratings cluster according to exercises and not dimensions. Seeking further understanding of exercise effects, the authors proposed 2 exercise-based factors (exercise form and exercise content) as sources of variance in assessment center ratings. Exercise designs were manipulated so that 2 levels of form (leaderless group discussions and role-play exercises) were crossed with 2 levels of content (cooperative and competitive task designs). Analysis of variance (ANOVA) and confirmatory factor analysis procedures applied to multitrait–multimethod ratings of 89 high school student assessees revealed that most of the variance in the ratings was explained by exercises and not dimensions. Exercise form accounted for 16% of method variance, whereas exercise content had near zero effect. The form effect was primarily due to higher correlations of dimension ratings across role-play exercises. Practical implications and future directions for exercise design research are discussed. Effects of a quick-release form of bromocriptine (Ergoset) on fasting and postprandial plasma glucose, insulin, lipid, and lipoprotein concentrations in obese nondiabetic hyperinsulinemic women

OBJECTIVE: To assess the effect on various aspects of carbohydrate and lipid metabolism of administering a quick-release formulation of bromocriptine (Ergoset) to obese, nondiabetic, hyperinsulinemic women. RESEARCH DESIGN AND METHODS: Hourly concentrations of prolactin, glucose, insulin, free fatty acid (FFA), and triglyceride were measured for 24 h before and after approximately 8 weeks of treatment with Ergoset. In addition, fasting lipid and lipoprotein concentrations and the steady-state plasma glucose (SSPG) concentration in response to a continuous infusion of somatostatin, insulin, and glucose were determined before and after Ergoset administration. RESULTS: Circulating prolactin concentrations were dramatically decreased (P < 0.001) following treatment, associated with a significant fall (P < 0.05) in 24-h-long plasma glucose, FFA, and triglyceride concentrations. Neither circulating plasma insulin concentrations nor the ability of insulin to mediate glucose disposal changed with treatment. Finally, fasting total cholesterol fell (P < 0.05) and the ratio of total to HDL cholesterol decreased (P = 0.06) in association with Ergoset treatment. CONCLUSIONS: The fact that significant metabolic improvement was seen in the obese nondiabetic hyperinsulinemic women studied suggests that Ergoset could be of therapeutic benefit in clinical conditions of hyperglycemia and/or dyslipidemia.     

The effects of induction conditions on production of a soluble antitumor sFv in Escherichia coli

CC49 is a second generation monoclonal antibody(mAb) with highaffinity to a pancarcinoma antigen, TAG-72. A single-chain Fv(sFv)ofCC49 may have a role in managing human carcinomas. Most reportedsFvs have been expressed as insoluble products that must besolubilized and renatured. Soluble sFv expression is advantageousas activity can be assayed directly from the periplasmic fraction.Also, gene-level immunoconjugates may not be amenable to refoldingprotocols. Using a vector that carries the tac promoter andomp A signal, we have examined the effects of four variableson the expression and accumulation of soluble CC49 sFv: (i)linker sequence joining VL and VH, (ii) isopropylthio-ß-D-galactosideconcentration for induction, (iii) temprature, and (iv) theaddition of nonmetabolizable sugars to the medium. We have beenable to demonstrate, using rapidly prepared periplasmic extracts,that the yield of soluble sFv improves by the addition of 0.4Msucrose to the medium and by inducing expression with a verylow concentration of IPTG (0.02–0.03 mM). Under theseinduction conditions periplasmic extracts demonstrate increasedexpression of the sFv, as shown by the larger amount of a 27kDa band on SDS-polyacrylamide gel, and an increased abilityto inhibit binding of the mAb CC49 to immobilized tumor extracts.     

The large deflection dynamic plastic response of rectangular plates

By assuming a kinematically admissible, time-dependent velocity field and considering the global equilibrium of all the forces acting on each rigid segment during large deflection of a plate, a complete theoretical investigation is undertaken herein to trace the large deflection dynamic plastic response (including the transient phase) of simply-supported or fully-clamped rectangular plates. This procedure deduces two modifying factors ƒ₁ and ƒ₂ which reflect the effect of the membrane forces and are employed to formulate the governing equations in the case of large deflections. The present prediction of the final plate deflection coincides excellently with the corresponding experimental results for deflections up to 5–10 times the plate thickness. This theory greatly improves the estimates based on the bending-only theory and provides a new way to trace the transient phase of dynamically loaded plates when the effect of membrane forces is significant.

All-optical photonic switches using integrated inverted asymmetricFabry-Perot modulators and heterojunction phototransistors

InGaAs/GaAs(100) multiple-quantum-well-based inverted cavity asymmetric Fabry-Perot modulators are vertically integrated with GaAs/AlGaAs heterojunction phototransistors to yield all-optical photonic switches. The photonic switches using `normally on' modulator pixels exhibit an output on-off ratio of 12:1 with internal optical gain of 4 dB. The photonic switches using `normally off' modulator pixels yield similar contrast and gain, but exhibit intrinsic bistable behavior. The inverted cavity modulators employed permit utilizing the transparency of the GaAs substrate at the operating wavelength and offer advantages for fabricating large arrays for optical signal processing
