An intrinsic feature of yeast artificial chromosomes (YACs) is that the cloned DNA is generally in the same size range (i.e., approximately 200-2000 kb) as the endogenous yeast chromosomes. As a result, the isolation of YAC DNA, which typically involves separation by pulsed-field gel electrophoresis, is frequently confounded by the presence of a comigrating or closely migrating endogenous yeast chromosome(s). We have developed a strategy that reliably allows the isolation of any YAC free of endogenous yeast chromosomes. Using recombination-mediated chromosome fragmentation, a set of Saccharomyces cerevisiae host strains was systematically constructed. Each strain contains defined alterations in its electrophoretic karyotype, which provide a large-size interval devoid of endogenous chromosomes (i.e., a karyotypic "window"). All of the constructed strains contain the kar1-delta 15 mutation, thereby allowing the efficient transfer of a YAC from its original host into an appropriately selected window strain using the kar1-transfer procedure. This approach provides a robust and efficient means to obtain relatively pure YAC DNA regardless of YAC size.  相似文献

Metal halide perovskite (MHP) semiconductors have driven a revolution in optoelectronic technologies over the last decade, in particular for high-efficiency photovoltaic applications. Low-dimensional MHPs presenting electronic confinement have promising additional prospects in light emission and quantum technologies. However, the optimisation of such applications requires a comprehensive understanding of the nature of charge carriers and their transport mechanisms. This study employs a combination of ultrafast optical and terahertz spectroscopy to investigate phonon energies, charge-carrier mobilities, and exciton formation in 2D (PEA)2PbI4 and (BA)2PbI4 (where PEA is phenylethylammonium and BA is butylammonium). Temperature-dependent measurements of free charge-carrier mobilities reveal band transport in these strongly confined semiconductors, with surprisingly high in-plane mobilities. Enhanced charge-phonon coupling is shown to reduce charge-carrier mobilities in (BA)2PbI4 with respect to (PEA)2PbI4. Exciton and free charge-carrier dynamics are disentangled by simultaneous monitoring of transient absorption and THz photoconductivity. A sustained free charge-carrier population is observed, surpassing the Saha equation predictions even at low temperature. These findings provide new insights into the temperature-dependent interplay of exciton and free-carrier populations in 2D MHPs. Furthermore, such sustained free charge-carrier population and high mobilities demonstrate the potential of these semiconductors for applications such as solar cells, transistors, and electrically driven light sources.  相似文献

To improve organisational safety and enhance security efficiency, organisations seek to establish a culture of security that provides a foundation for how employees should approach security. There are several frameworks and models that provide a set of requirements for forming security cultures; however, for many organisations, the requirements of the frameworks are difficult to meet, if not impossible. In this research, we take a different perspective and focus on the core underlying competencies that high-reliability organisations (HROs) have shown to be effective in achieving levels of risk tolerance consistent with the goals of a security culture. In doing so we draw on high-reliability theory to develop a Security Culture Model that explains how a firm's supportive and practical competencies form its organisational security culture. To refine and test the model, we conducted a developmental mixed-method study using interviews and survey data with professional managers involved in the information security (InfoSec) programs within their respective HROs. Our findings emphasise the importance of an organisation's supportive and practical competencies for developing a culture of security. Our results suggest that organisations' security cultures are a product of their InfoSec practices and that organisational mindfulness, top management involvement and organisational structure are key to the development of those practices.  相似文献

Development of highly active and stable Pt‐free oxygen reduction reaction catalysts from earth‐abundant elements remains a grand challenge for highly demanded metal–air batteries. Ag‐based alloys have many advantages over platinum group catalysts due to their low cost, high stability, and acceptable oxygen reduction reaction (ORR) performance in alkaline solutions. Nevertheless, compared to commercial Pt/C‐20%, their catalytic activity still cannot meet the demand of commercialization. In this study, a kind of catalysts screening strategy on Agx Cu100?x nanoalloys is reported, containing the surface modification method, studies of activity enhancement mechanism, and applied research on zinc–air batteries. The results exhibit that the role of selective dealloying (DE) or galvanic displacement (GD) is limited by the “parting limitation”, and this “parting limitation” determines the surface topography, position of d‐band center, and ORR performance of Agx Cu100?x alloys. The GD‐Ag55Cu45 and DE‐Ag25Cu75 catalysts alloys present excellent ORR performance that is comparable to Pt/C‐20%. The relationship between electronic perturbation and specific activity demonstrates that positive shift of the d‐band center (≈0.12 eV, relative to Ag) for GD‐Ag55Cu45 is beneficial for ORR, which is contrary to Pt‐based alloys (negative shift, ≈0.1 eV). Meanwhile, extensive electrochemical and electronic structure characterization indicates that the high work function of GD‐Ag55Cu45 (4.8 eV) is the reason behind their excellent durability for zinc–air batteries.  相似文献

This paper proposes a new approach to using particle swarm optimisation (PSO) within an AdaBoost framework for object detection. Instead of using exhaustive search for finding good features to be used for constructing weak classifiers in AdaBoost, we propose two methods based on PSO. The first uses PSO to evolve and select good features only, and the weak classifiers use a simple decision stump. The second uses PSO for both selecting good features and evolving weak classifiers in parallel. These two methods are examined and compared on two challenging object detection tasks in images: detection of individual pasta pieces and detection of a face. The experimental results suggest that both approaches can successfully detect object positions and that using PSO for selecting good individual features and evolving associated weak classifiers in AdaBoost is more effective than for selecting features only. We also show that PSO can evolve and select meaningful features in the face detection task.  相似文献

Performing and composing for interactive audiovisual system presents many challenges to the performer. Working with visual, sonic and gestural components requires new skills and new ways of thinking about performance. However, there are few studies that focus on performer experience with interactive systems. We present the work Blue Space for oboe and interactive audiovisual system, highlighting the evolving process of the collaborative development of the work. We consider how musical and technical demands interact in this process, and outline the challenges of performing with interactive systems. Using the development of Blue Space as a self-reflective case study, we examine the role of gestures in interactive audiovisual works and identify new modes of performance.  相似文献

Magnetization measurements are reported for seven members of the series of compounds RE x Mo 6 Se 8 (x1; RE=La, Ce, Pr, Eu, Gd, Yb, and Lu). These measurements establish that Ce, Pr, and Gd ions are trivalent in these compounds, but that Eu and Yb are primarily divalent, in agreement with crystallographic data. Large deviations of the susceptibilities for CeMo 6 Se 8 , PrMo 6 Se 8 , and YbMo 6 Se 8 from Curie-Weiss behavior above 10 K are attributed predominantly to crystal field effects, and quantitative agreement between crystal field theory and the experimental data was obtained for CeMo 6 Se 8 above 25 K. Further, we find for Gd 1.2 Mo 6 Se 8 an unusually small spin exchange interaction between the Gd ions and conduction electrons, consistent with previous estimates of this quantity based on superconductivity data. For the Gd, Eu, and Ce compounds, the low-temperature data indicate very small Curie-Weiss temperatures of –1.7, –0.6, and +0.4 K, respectively. No evidence for magnetic ordering of the RE ions in any of the above compounds was observed, with the exception of CeMo 6 Se 8 , for which the magnetization data at 0.7–1.3 K suggest the onset of ferromagnetic order.Research sponsored by the Air Force Office of Scientific Research, Air Force Systems Command, USAF, under AFOSR Contract No. AFOSR/F-44620-72-C-0017.  相似文献
