Bone and cartilage generation by three-dimensional scaffolds is one of the promising techniques in tissue engineering. One approach is to generate histologically and functionally normal tissue by delivering healthy cells in biocompatible scaffolds. These scaffolds provide the necessary support for cells to proliferate and maintain their differentiated function, and their architecture defines the ultimate shape. Rapid prototyping (RP) is a technology by which a complex 3-dimensional (3D) structure can be produced indirectly from computer aided design (CAD). The present study aims at developing a 3D organic-inorganic composite scaffold with defined internal architecture by a RP method utilizing a 3D printer to produce wax molds. The composite scaffolds consisting of chitosan and hydroxyapatite were prepared using soluble wax molds. The behaviour and response of MC3T3-E1 pre-osteoblast cells on the scaffolds was studied. During a culture period of two and three weeks, cell proliferation and in-growth were observed by phase contrast light microscopy, histological staining and electron microscopy. The Giemsa and G?m?ri staining of the cells cultured on scaffolds showed that the cells proliferated not only on the surface, but also filled the micro pores of the scaffolds and produced extracellular matrix within the pores. The electron micrographs showed that the cells covering the surface of the struts were flattened and grew from the periphery into the middle region of the pores.  相似文献

The Popov invariants are used to define two matrices MAB and KAB, which describe the properties of a system x? = Ax + Bu in a basis-independent form. Also a control system with state feedback K and nonsingular input transformation M, x? = (A ? BK)x + BMw = Fx + Gw, can be uniquely specified by the corresponding matrices MFG and KFG, which may be chosen arbitrarily with the only condition, that (F, G) must have the same unordered set of Kronecker invariants as (A, B). It is shown, how K and M are computed from MAB, KAB, MFG and KFG. Implications for pole assignment, minimal polynomial assignment and observer design are shown. Also a solution to the minimal realization problem is given, which results in MAB, KAB and the output relation y = Cx.  相似文献

An antibody ELISA test and a PCR method for identifying the risk of Salmonella contamination were compared in a field study on the same lots of animals in a slaughterhouse. The results were compared to investigations carried out on two farms with different prevalences of Salmonella antibody-positive animals. Salmonella antibody ELISA testing was carried out on all 383 meat juice samples derived from the diaphragm pillar muscle of each pig. Salmonella DNA analysis was performed by PCR technique on small intestine samples with lymph nodes from all 383 pigs, and on tonsils from the last 129 pigs. The 383 animals tested came from 32 different pig farms. Furthermore, the herd antibody blood serum status against Salmonella spp. of weaners was determined on two selected pig fattening farms, one with low and one with high seroprevalence in meat juice. A total of 7.0% (ELISA cut-off OD% > or =40) of the slaughtered pigs from 6 of 32 fattening farms were seropositive. Salmonella DNA was found in 16.4% of the jejunum/lymph nodes (383 animals) and in 15.5% of the tonsils (129 animals). Salmonella DNA was found in the jejunum/lymph nodes of 41% of the seropositive pigs. However, serotitres were also positive in only 17.5% of all pigs positive in the jejunum DNA test. Two farms were selected for further investigation: farm 13 (F13), with a high prevalence of seropositive pigs, 29.0%, Category II; and F11, with 9.4%, Category I. However, categorization according to the blood serum tests of the fattening pigs after on-farm testing was very different: F13 had 5% positive animals (Category I); and F11, 23.3% (Category II). The study led to the following results and recommendations: First, ELISA tests are useful for the detection of farms that are regularly contaminated with Salmonella, but such tests cannot give information on the infectious status of a single animal (or a group) at the point of slaughter. Second, it is crucial that management measures are taken to prevent the spread of infections by trade and transport: piglets should be supplied exclusively by a single, well-known producer, and finishers should be tested serologically on farm before going to slaughter. Third, ELISA tests and the PCR method are suitable for the detection of Salmonella and are recommended as analytical tools for all pork quality control programmes. Fourth, animals from suspicious farms should always be slaughtered at the end of the slaughter day, followed by thorough cleaning and disinfection.  相似文献

The reliable determination of soluble, insoluble and total dietary fibre in baked goods and cereal flours is an important issue for research, nutritional labelling and marketing. We compared total dietary fibre (TDF) contents of selected cereal based foods determined by AOAC Method 991.43 and the new AOAC Method 2009.01. Fifteen bread and bakery products were included in the study. Our results showed that TDF values of cereal products determined by AOAC Method 2009.01 were always significantly higher than those determined by AOAC Method 991.43. This was explained by the inclusion of low molecular weight soluble fibre fractions and resistant starch fractions in the TDF measurement by AOAC 2009.01. This documents that nutritional labelling of cereal products poses the challenge how to update TDF data in nutrient databases in a reasonable time with an acceptable expenditure.  相似文献

Monitoring rumination behavior serves multiple purposes in feeding and herd management of dairy cows. The process of calving is a major event for cows, and a detailed understanding of alterations in behavioral patterns of animals in the time around calving is important in calving detection. The objective of this study was to describe the short-term changes in rumination patterns in dairy cows immediately before and after parturition. In total, 17 cows were fitted with rumination sensors that were able to monitor rumination time, number of rumination boli, and number of rumination jaw movements. Rumination time was decreased in the last 4 h antepartum and in the first 8 h postpartum. Cows stopped ruminating 123 ± 58 min (mean ± standard deviation) before calving and resumed ruminating 355 ± 194 min after calving. The number of rumination jaw movements and boli per day were decreased in the 24-h period postpartum. Rumination rate, the number of rumination jaw movements per rumination minute, and the number of boli per rumination minute changed little around calving. The calving event primarily influenced the duration and frequency of various rumination characteristics but not rumination intensity. Among detected characteristics, rumination time showed the greatest potential for monitoring of calving events.  相似文献

Visualization by high-speed videography and infrared surface thermography was used to compare the spatial and temporal maldistribution of flow, manifesting itself in pulsation and hot spot formation, respectively, in water evaporators consisting of either a single metallic foil with an array of mechanically micromachined microchannels or of several such foils assembled into an electrically powered micro heat exchanger. In the single layer devices examined by high-speed videography, pulsation in the frequency range below 20 Hz was found to be dominated by the generation of large bubbles in the inlet plenum. A redesign of the inlet with microchannels instead of a large plenum eliminated the pulsation at sub-audio frequencies, at the expense of a significantly increased pressure drop across the device. Infrared thermography of an electrically powered micro heat exchanger operated as an evaporator showed the formation of metastable hot spots as the result of maldistribution among different microchannel array layers. The formation of these hot spots could be eliminated by operating the device under heater cartridge temperature control conditions instead of constant power conditions.  相似文献

The objective of this study was to determine the impact of different slaughter procedures on animal welfare and meat quality. Before slaughter in a dip-lift, one-gondola system, 460 pigs were exposed to an atmosphere containing either 80% or 90% CO(2) for 70 or 100s, and at the longer exposure time with stun-to-stick intervals of either 25-35 or 40-50s. Clinical parameters (reflexes, catecholamines and lactate) showed deficiencies in animal welfare after stunning with 80% CO(2) for 70 and 100s, with an interval of 40-50s, and with 90% CO(2) for 70s. Stunning with 80% CO(2) for 70 or 100s always induced stress, as indicated by higher lactate levels, and reduced meat quality as indicated by low pH(24) values (5.4 in Musculus longissimus and 5.5 in Musculus semimembranosus) and low impedance (Py(24)) values, especially in combination with the longer stun-to-stick interval (40-50s). Stunning with 80% or 90% CO(2) in a dip-lift system was found to be acceptable for animal welfare (percentage of clinical reflexes) only in combination with the longer exposure time of 100s and the shorter stun-to-stick times of 25-35s. When 90% CO(2) was used, the longer stun-to-stick interval of 40-50s also gave results acceptable with regard to animal welfare. Generally, in comparison to 80% CO(2) stunning meat quality was superior (higher pH(24) and Py(24) values) after stunning with 90% CO(2).  相似文献

Bioresorbable implantable medical devices show a great potential for applications requiring medical care over well‐defined periods of time. Once their function is fulfilled, such implants naturally degrade and resorb in the body, which eliminates adverse long‐term effects or the need for a secondary surgery to extract the implanted device. Since biodegradable materials are water‐soluble, the fabrication of such transient electronic circuits and devices requires special care and needs to rely solely on dry processing steps without exposure to aqueous solutions. A further challenge is the in vivo powering of medical implants that are only constituted of biodegradable materials. This paper describes the design, fabrication, and testing of radio‐frequency biodegradable magnesium microresonators. To this end, an innovative microfabrication process with minimal exposure to aqueous media is developed to fabricate magnesium‐based, water‐soluble electronic components. It consists of a novel sequence of only three steps: one physical vapor deposition, one photolithography, and one ion beam etching step. The frequency‐selective wireless heating of different resonators is demonstrated. This represents a significant step toward their use as power receivers and microheaters in biodegradable implantable medical devices, for applications such as triggered drug release.  相似文献
