Bis-(monoacylglyceryl) phosphate and acyl phosphatidylglycerol were isolated from the liver of two patients with lipidosis induced by 4,4′-diethylaminoethoxyhexesterol. Identification was based upon the results of alkaline hydrolysis, acetolysis, IR spectrometry, and upon the determination of molar ratio of phosphorus-glycerol-ester. The contents of the bis-(monoacylglyceryl) phosphate were 10 and 16% total phospholipid phosphorus in them. The bis-(monoacylglyceryl) phosphate contained mainly docosahexaenoic (42%), oleic (29%), and linoleic acid (14%) and had the hemolytic activity of ca. one-eighth lysolecithin from egg yolk. Acidic lipids from the liver also were found to contain a lipid which is less polar than bis-(monoacylglyceryl) phosphate. The results of lipid analysis showed that the lipid possessed the structure of an acyl phosphatidylglycerol, and its content was ca. 2% total phospholipid phosphorus. Accumulation of 4,4′-diethylaminoethoxyhexesterol and its derivatives was found in clinical cases by thin layer chromatography and IR spectrometry. This fact suggested that human liver has an ability to metabolize the drug.  相似文献

Specimens of two kinds of isotropic nuclear graphite, IG-110U and ETP-10, were neutron-irradiated at fluence of 1.92 × 1024 n/m2 (E > 1.0 MeV) at 473 K. The recoveries of the macroscopic lengths of these specimens during isothermal and isochronal annealing at temperatures of up to 1673 K were investigated in a step-wise manner by using a precision dilatometer. The macroscopic lengths after isochronal annealing for 6 h at each temperature decreased gradually as the temperature was increased to 1673 K. The recovery trends of the c-axis and a-axis lattice parameters differed from one another, and from the macroscopic length recovery trends. For the IG-110U specimen, the activation energies (Ea) of macroscopic volume recovery corresponding to annealing at 523–773, 773–923, 923–1073, and 1073–1173 K were found to be 0.15, 0.34, 0.73, and 2.59 eV, respectively. For the ETP-10 specimen, the Ea corresponding to 523–923, 923–1223, and 1223–1373 K were determined to be 0.15, 0.46, and 2.19 eV, respectively. These results indicate that both graphite specimens underwent three or four stages of macroscopic length recovery between 523 K and the annealing temperatures at which their initial lengths were recovered. It is suggested that during the first stage recovery proceeded via the migration of single interstitials along the basal plane and the resulting V-I recombination. In the middle stages, recovery occurred due to the migration of interstitial groups such as C2 along the basal plane, while in the last stage, it proceeded via through-layer migration of interstitials or migration of single vacancies.  相似文献

Advanced glycation end-products (AGE) play a role in the pathogenesis of several diseases, including diabetic complications and atherosclerosis. In atherosclerotic lesions of human aortas, AGE are localized in the extracellular matrix and intracellularly in foam cells. Two interpretations are possible for AGE accumulation inside macrophages, one is endocytic uptake of extracellular AGE-proteins by scavenger receptors; the other is intracellular AGE formation inside the macrophages. In the present study, we determined the pathways involved in AGE accumulation inside macrophages. RAW 264.7 cells, a murine macrophage cell line, incubated with BSA and 1600 mM glucose for 40 weeks, recognized heavily modified AGE- BSA. In contrast, the cells showed no ligand activity for mildly modified AGE-BSA, prepared by incubating BSA with 50 mM glucose for 24 weeks. Nepsilon-(carboxymethyl)lysine (CML)-modified proteins of about 65 kDa were detected in human monocyte-derived macrophages incubated for 7 days with 30 mM glucose and phorbol myristate acetate. Furthermore, CML was generated when glycated protein was incubated with hypochloric acid. Taken together, our results indicate that AGE detected inside foam cells in atherosclerotic lesions are generated intracellularly rather than representing endocytic uptake of extracellular AGE-proteins by scavenger receptors.  相似文献

The velocity and absorption coefficient of longitudinal ultrasonic waves with 5 and 10 MHz in molten and glassy silicates and borates have been measured by means of the pulse-echo method in 300 to 1600 K. The silicate and borate were Na2O−SiO2, K2O−SiO2, PbO−SiO2, Na2O−B2O3, and PbO−B2O3. The velocity of sound decreased with increasing temperature and decreased rapidly near the transition temperature of glasses. The product of the one-third power of velocity of sound and the molar volume of the oxide melt was constant and independent of temperature. The absorption coefficient of ultrasonic waves in the oxide melts increased with an increase in viscosity. The absorption mechanisms of ultrasonic waves and the mean free path of phonons in the oxide melts were discussed.  相似文献

The present study was carried out to examine if the positional distribution of medium-chain fatty acid (MCF) in dietary synthetic fat influences lymphatic transport of dietary fat and the chemical composition of chylomicrons in rats with permanent cannulation of thoracic duct. Four types of synthetic triacylglycerol were prepared: (i) sn-1(3) MCF-sn 2 linoleic acid, (ii) interesterified sn-1(3) MCF-sn 2 linoleic acid, (iii) sn-2 MCF-sn-1(3) linoleic acid, and (iv) interesterified sn-2 MCF-sn-1(3) linoleic acid. A purified diet composed of equal amounts of the synthetic fat and cocoa butter was given to rats with permanent lymph duct cannulation. The positional distribution of MCF in the dietary fat had no significant effect on the lymph flow, triacylglycerol output, phospholipid output, lipid composition of chylomicrons, or the particle size. The positional distribution of MCF in the synthetic triacylglycerol was maintained in the chylomicron triacylglycerol. These results showed that MCF in the dietary triacylglycerol is transported into lymphatics and the positional distribution is well preserved in chylomicron triacylglycerol.  相似文献

The thermal behavior of nitrocellulose (NC) containing diphenylamine (DPA), 2‐nitrodiphenylamine (2‐NO2‐DPA), N‐nitrosodiphenylamine (N‐NO‐DPA), ethyl centralite (EC), akardite II (AKII), 1,1,3‐tri‐(2‐methyl‐4‐hydroxy‐5‐tert‐butyl‐phenyl)‐butane (BPless), 3‐(3,5‐di‐tert‐butyl‐4‐hydroxy‐phenyl)‐propionic acid octadecyl ester (BPhin), and 3‐(3‐tert‐butyl‐4‐hydroxy‐5‐methyl‐phenyl)‐propionic acid 2‐(9‐{2‐[3‐(3‐tert‐butyl‐4‐hydroxy‐5‐methyl‐phenyl)‐propionyloxy]‐1,1‐dimethyl‐ethyl}‐2,4,8,10‐tetraoxa‐spiro [5.5] undec‐3‐yl)‐2‐methyl‐propyl ester (BPsemi) was observed during isothermal storage at 393 K using a C80 microcalorimeter. The results indicate that each stabilizer decreased NC's maximum heat release rate and increased the induction period of heat release. We also observed that the maximum heat release rate and the induction period were dependent on the amount of stabilizer. DPA decreased the maximum heat release rate to the greatest extent, with the other stabilizers having similar effects on the decrease of maximum heat release rate. AKII prolonged the induction period of heat release most. The order of prolongation of the induction period was AKII>2‐NO2‐DPA≈DPA>N‐NO‐DPA≈EC>BPless≈BPsemi≈BPhin.  相似文献

Recently, we have reported that anthocyanins show strong antioxidative activity, but no attention has been paid to anthocyanins from the viewpoint of the reaction mechanism of alkylperoxyl radicals; therefore, we investigated the reaction products of antioxidative anthocyanins (cyanidin 3-O-β-d-glucoside). Cyanidin 3-O-β-d-glucoside was reacted with 2,2′-azobis(2,4-dimet hylvaleronitrile) to generate the alkylperoxyl radicals, and the reaction products were isolated by high-performance liquid chromatography. The products were identified as 4,6-dihydroxy-2-O-β-d-glucosyl-3-oxo-2,3-dihydrobenzofuran and protocatechuic acid. Based on reaction products, the antioxidative mechanism of cyanidin 3-O-β-d-glucoside may be different from that of α-tocopherol; cyanidin 3-O-β-d-glucoside would produce another radical scavenger, as it would break down the structure and scavenge the radicals.  相似文献

This paper discusses the theoretical foundation of Stone’s BSS (Stone in Neural Comput 13:1559–1574, 2001; Stone in Independent Component Analysis: A Tutorial Introduction, A Bradford Book, London, 2004), and it proposes a novel BSS approach based on second-order statistics of the responses of two different linear filters to source signals. The proposed approach which includes Stone’s BSS as a special case helps us to understand how generalized eigenvalue decomposition (GEVD) concludes separating vectors in Stone’s BSS. It obtains the separating vectors by simultaneous diagonalization of covariance matrices of two different linear filters responses to the mixtures. The two employed linear filters are selected dependent on source signals structures under the assumption that they have different responses to source signals. Here, two FIR filters with coefficients selected in an opposite probabilistic way have been suggested for the proposed BSS. The proposed BSS method has been compared with Stone’s BSS, SOBI and AMUSE over speech and image mixtures in different noise levels.  相似文献
