Visual working memory (WM) is traditionally considered a robust form of visual representation that survives changes in object motion, observer's position, and other visual transients. This article presents data that are inconsistent with the traditional view. We show that memory sensitivity is dramatically influenced by small variations in the testing procedure, supporting the idea that representations in visual WM are susceptible to interference from testing. In the study, participants were shown an array of colors to remember. After a short retention interval, memory for one of the items was tested with either a same–different task or a 2-alternative-forced-choice (2AFC) task. Memory sensitivity was much lower in the 2AFC task than in the same–different task. This difference was found regardless of encoding similarity or of whether visual WM required a fine or coarse memory resolution. The 2AFC disadvantage was reduced when participants were informed shortly before testing which item would be probed. The 2AFC disadvantage diminished in perceptual tasks and was not found in tasks probing visual long-term memory. These results support memory models that acknowledge the labile nature of visual WM and have implications for the format of visual WM and its assessment. Photoreceptor disc component (PRCD) is a small protein which is exclusively localized to photoreceptor outer segments, and is involved in the formation of photoreceptor outer segment discs. Mutations in PRCD are associated with retinal degeneration in humans, mice, and dogs. The purpose of this work was to identify PRCD-binding proteins in the retina. PRCD protein-protein interactions were identified when implementing the Ras recruitment system (RRS), a cytoplasmic-based yeast two-hybrid system, on a bovine retina cDNA library. An interaction between PRCD and tubby-like protein 1 (TULP1) was identified. Co-immunoprecipitation in transfected mammalian cells confirmed that PRCD interacts with TULP1, as well as with its homolog, TUB. These interactions were mediated by TULP1 and TUB highly conserved C-terminal tubby domain. PRCD localization was altered in the retinas of TULP1- and TUB-deficient mice. These results show that TULP1 and TUB, which are involved in the vesicular trafficking of several photoreceptor proteins from the inner segment to the outer segment, are also required for PRCD exclusive localization to photoreceptor outer segment discs.  相似文献

The genetically obese Zucker rat is a widely investigated model of pathological changes associated with type 2 diabetes. To assess cognitive function, obese and lean Zucker rats were tested on a variable-interval delayed alternation test of learning and memory. There were no group differences in learning the alternation rule or at short intervals, but obese rats were impaired at longer intervals where performance is hippocampus dependent. Plasma membrane association of the insulin sensitive glucose transporter, GLUT4, was reduced in the hippocampus of obese rats in the absence of changes in total GLUT4 and insulin receptor expression. These results parallel those of human studies in pointing to the susceptibility of the hippocampus and related structures to the adverse environment of diabetes mellitus. Objective: Novel antibiotic Ramizol® is advancing to clinical trials for the treatment of gastrointestinal Clostridium difficile associated disease. Despite this, previous studies have shown a rapid plasma clearance upon intravenous administration and low oral bioavailability indicating pure drug is unsuitable for systemic infection treatment following oral dosing. The current study aims to investigate the development of poly-lactic-(co-glycolic) acid (PLGA) particles to overcome this limitation and increase the systemic half-life following subcutaneous and intramuscular dosing.

Significance: The development of new antibiotic treatments will help in combatting the rising incidence of antimicrobial resistance.

Methods: Ramizol® was encapsulated into PLGA nano and microparticles using nanoprecipitation and emulsification solvent evaporation techniques. Formulations were analyzed for particle size, loading level and encapsulation efficiency as well as in vitro drug release profiles. Final formulation was advanced to in vivo pharmacokinetic studies in Sprague–Dawley rats.

Results: Formulation technique showed major influence on particle size and loading levels with optimal loading of 9.4% and encapsulation efficiency of 92.06%, observed using emulsification solvent evaporation. Differences in formulation technique were also linked with subsequent differences in release profiles. Pharmacokinetic studies in Sprague–Dawley rats confirmed extended absorption and enhanced bioavailability following subcutaneous and intramuscular dosing with up to an 8-fold increase in Tmax and T1/2 when compared to the oral and IV routes.

Conclusions: Subcutaneous and intramuscular dosing of PLGA particles successfully increased systemic half-life and bioavailability of Ramizol®. This formulation will allow further development of Ramizol® for systemic infection eradication.  相似文献

Earlier studies have suggested that higher education institutions could harness the predictive power of Learning Management System (LMS) data to develop reporting tools that identify at-risk students and allow for more timely pedagogical interventions. This paper confirms and extends this proposition by providing data from an international research project investigating which student online activities accurately predict academic achievement. Analysis of LMS tracking data from a Blackboard Vista-supported course identified 15 variables demonstrating a significant simple correlation with student final grade. Regression modelling generated a best-fit predictive model for this course which incorporates key variables such as total number of discussion messages posted, total number of mail messages sent, and total number of assessments completed and which explains more than 30% of the variation in student final grade. Logistic modelling demonstrated the predictive power of this model, which correctly identified 81% of students who achieved a failing grade. Moreover, network analysis of course discussion forums afforded insight into the development of the student learning community by identifying disconnected students, patterns of student-to-student communication, and instructor positioning within the network. This study affirms that pedagogically meaningful information can be extracted from LMS-generated student tracking data, and discusses how these findings are informing the development of a customizable dashboard-like reporting tool for educators that will extract and visualize real-time data on student engagement and likelihood of success.  相似文献

The promise of collaborative governance is to implement policies that address uncertain and multi‐scale environmental problems while also allowing local social systems to successfully adapt. The success of such efforts is contingent on the capacity of community‐based institutions such as lake associations to effect change. These organizations encapsulate the values communities hold for lakes and their catchments and can play a central role in managing the integrity of lakes’ freshwater resources. To this end, we develop a framework to delineate the pathways lake associations can take to contribute to the management of lakes and their catchments. We suggest the cornerstones of effective engagement in collaborative governance are the goals the organization sets for itself (mission), the capital that can be mobilized by the organization (capacity) and the activities the organization undertakes (actions). We propose measurable indicators for each component and suggest how the framework can be used to assess contributions of lake associations to multi‐scale governance. Finally, we suggest how this framework can be integrated with ecological monitoring frameworks as part of a long term social‐ecological research and monitoring program.  相似文献

The United States Environmental Protection Agency recommends density thresholds for the fecal indicator organism Escherichia coli in order to ensure the safety of recreational waters. A number of studies published over the past ten years indicate that E. coli is encountered frequently in sand at recreational beaches. While a majority of the sand-associated E. coli may be commensal or environmental strains, the potential for pathogenic strains of E. coli to be present exists. Therefore, the aim of this study was to assess the presence of attachment and virulence genes associated with enteropathogenic and enterohemorrhagic strains of E. coli (EPEC and EHEC) in populations of E. coli recovered from swash zone sand from seven recreational beaches along Lake Huron and Lake St. Clair in eastern Michigan, USA. Genes coding for attachment proteins in EPEC and EHEC were very prevalent in sand E. coli, but genes coding for toxin genes were uncommon. The paucity of genes associated with E. coli toxins suggests that the EPEC and EHEC pathotypes are not common in sand; however, the high prevalence of genes associated with attachment in E. coli pathotypes suggests that these genes are being retained within the beach sand E. coli population.  相似文献

We continue the study of bin packing with splittable items and cardinality constraints. In this problem, a set of n items must be packed into as few bins as possible. Items may be split, but each bin may contain at most?k (parts of) items, where k is some given parameter. Complicating the problem further is the fact that items may be larger than?1, which is the size of a bin. The problem is known to be strongly NP-hard for any fixed value of?k. We essentially close this problem by providing an efficient polynomial-time approximation scheme (EPTAS) for most of its versions. Namely, we present an efficient polynomial time approximation scheme for k=o(n). A?PTAS for k=Θ(n) does not exist unless P = NP. Additionally, we present dual approximation schemes for k=2 and for constant values of?k. Thus we show that for any ε>0, it is possible to pack the items into the optimal number of bins in polynomial time, if the algorithm may use bins of size 1+ε.  相似文献
