Electron transfer and transport at metal-molecule interfaces has been a central problem in a number of disciplines. There is renewed interest in this problem from the rapidly growing field of molecule-based electronics, where the so-called “contact problem” is usually probed in transport measurements. Here, we probe interfacial electron transfer/transport using time-resolved two-photon photo-emission spectroscopy. Such an approach allows us to quantitatively measure the energetic alignment of occupied and unoccupied molecular orbitals to the metal Fermi level, the electronic coupling strength (or spectral density) between the molecular orbital and the metal substrate, and electronic-nuclear coupling leading to dynamic localization. These three types of information are in fact key ingredients in quantitative theories for electron transfer and transport at molecule-metal surfaces. Recent examples from our laboratory are used to illustrate the point. They include femtosecond dynamics of image potential resonances on heptane-covered Au(111) and exciton relaxation dynamics in C60 epitaxial films on Au(111). On heptane/Au(111), the lifetime of the n = 1 image potential resonance increases from 52 fs at one monolayer (ML) coverage to 700 fs at 2 ML and 2.8 ps at 3 ML. On C60/Au(111), the Frenkel exciton (involving the LUMO+1 level) couples over long distance to the metal substrate, with lifetime of 350 fs at 30 ML to 80 fs at 2 ML coverage. The film-thickness-dependent decay dynamics in both cases can be attributed to interfacial electron transfer. Fitting the distance-dependent electron transfer rates to a simple exponential function gives the characteristic distance parameter of ß = 0.6 ± 0.1 and 0.023 ± 0.005 Å−1 for heptane- and C60-covered Au(111), respectively. The difference in the ß parameters illustrates the essential role of the electronic structure of molecules in mediating interfacial electron transfer/transport.  相似文献

Measurements of the spectral properties of ridge waveguide graded index separate-confinement heterostructure single-quantum-well GaAs/AlGaAs lasers are discussed. Long cavity lasers (800 μm) exhibit remarkably pure single-longitudinal mode spectra under continuous operation in spite of the short cavity mode spacing. At an output power of 5 mW, the sidemode suppression exceeds 24 dB and the linewidth is 1.5 MHz. The linewidth-power product is 6.4 MHz-mW. Measurements of the linewidth power product as a function of cavity length L gives an L-2 dependence in agreement with theory for lasers with small internal loss. The results are used to deduce the linewidth enhancement factor α at the gain peak wavelength ant its dependence on the excitation level. A decrease in α was observed for lasers operating at the second quantized state due to a recovery of the differential gain  相似文献

A set of three complementary analytical methods were developed specifically for exhaled breath as collected in evacuated stainless steel canisters using gas chromatographic-mass spectrometric detection. The first is a screening method to quantify the carbon dioxide component (generally at 4-5% concentration), the second method measures the very volatile high-level endogenous compounds [e.g. acetone and isoprene at 500-1000 parts per billion by volume (ppbv), methanol, ethanol, dimethylsulfide at 2-10 ppbv], and the third method is designed to measure trace-level environmental contaminants and other endogenous volatile organic compounds (VOCs) (sub-ppbv) in breath. The canister-based sample format allows all three methods to be applied to each individual sample for complete constituent characterization. Application of these methods is shown to be useful in the following ways: analysis of CO2 levels indicates the approximate quantity of alveolar breath collected (as opposed to whole breath) in a sample; levels of major endogenous compounds are shown to be influenced by physical activities and subsequent recovery periods; and environmental exposures to xenobiotic VOCs can be characterized by assessment of post-exposure breath elimination curves. The instrumentation and methodology are described and example chromatograms and quantitative data plots demonstrating the utility of the methods are presented.  相似文献

This in vivo study examines the ability of 5'-amino-5'-deoxythymidine (5'-AdThd) to modulate 5-iododeoxyuridine (IdUrd) cellular metabolism in two human colon cancer xenografts (HT 29 and HCT-116), two actively proliferating normal mouse tissues (bone marrow and intestine), and a quiescent normal mouse tissue (liver). 5'-AdThd is a thymidine analogue that at low concentrations (<30 micrometer) can increase thymidine kinase activity, which is the rate-limiting enzyme for activation of IdUrd. We reported recently that the in vitro incubation of HT 29 and HCT-116 cells in 5'-AdThd + IdUrd resulted in an enhancement of 5-iodo-2'-dUTP pools, IdUrd DNA incorporation, and subsequent radiosensitization compared with incubation with IdUrd alone (Clin. Cancer Res., 1: 407-416, 1995). These in vitro effects were more significant in the radioresistant cell line HT 29. Using a 6-day continuous infusion of IdUrd (50 or 100 mg/kg/day) and/or 5'-AdThd (200 mg/kg/day), no increase in systemic toxicity (percentage of body weight loss) was observed in athymic nude mice with 5'-AdThd alone or when combined with IdUrd. There was significant dose-dependent, systemic toxicity with IdUrd, which was reversible within 3 days of completing the lower-dose IdUrd infusion. However, a comparison of plasma levels during the 6-day continuous infusion of IdUrd +/- 5'-AdThd showed a significant interaction of IdUrd and 5'-AdThd, resulting in higher plasma levels by day 6 of both compounds and the principal metabolites, iodouracil and deoxyuridine, which is consistent with nonlinear saturating effects on dihydrouracil dehydrogenase. Coadministration of IdUrd and 5'-AdThd resulted in an increase in the percentage of IdUrd DNA incorporation in the two proliferating normal tissues, which was significant only with the lower IdUrd dose. No effect on IdUrd DNA incorporation was found in normal liver at either IdUrd dose +/- 5'-AdThd. Similar to our in vitro data, the continuous infusion of IdUrd and 5'-AdThd showed a significant effect by increasing the percentage of IdUrd DNA incorporation in HT-29 xenografts at both IdUrd doses, whereas coadministration of 5'-AdThd had no such effect in HCT-116 xenografts.  相似文献

Water-soluble models of ligand-gated ion channels would be advantageous for structural studies. We investigated the suitability of three versions of the N-terminal extracellular domain (ECD) of the alpha7 subunit of the nicotinic acetylcholine receptor (AChR) family for this purpose by examining their ligand-binding and assembly properties. Two versions included the first transmembrane domain and were solubilized with detergent after expression in Xenopus oocytes. The third was truncated before the first transmembrane domain and was soluble without detergent. For all three, their equilibrium binding affinities for alpha-bungarotoxin, nicotine, and acetylcholine, combined with their velocity sedimentation profiles, were consistent with the formation of native-like AChRs. These characteristics imply that the alpha7 ECD can form a water-soluble AChR that is a model of the ECD of the full-length alpha7 AChR.  相似文献

A mathematical model simulating the blood transport and tissue residue distributions of the highly toxic and highly lipid soluble pesticide dieldrin in mammals is presented. This model is a significant improvement over our previously published preliminary model for dieldrin distribution in mammals. The assumptions and working hypotheses of the model are presented and used in generating a set of differential equations based upon mass balance principles. Two simulation cases are examined. The first simply demonstrates the gross features of: 1) Transport limiting conditions; 2) equal transport-equal membrane transfer conditions, and 3) membrane transfer limit conditions. The second studies a single tissue (the blood-brain barrier case) example of the above mentioned conditions. All simulations made were conducted for a hypothetical mature male rate of the average Wistar type eating food ad lib.  相似文献

Evaluated the ability of 2 screening instruments for geriatric psychiatric patients: one by A. Simon et al and the other by R. D. Savage and P. G. Britton. The overall correctness in discriminating was 66% for the Simon et al scale and 74% for the Savage and Britton scale, although the former misidentified normals as impaired, and the latter misidentified impaired Ss as normal.
