BACKGROUND: The release of grape components during wine making might be related to the mechanical properties of the skin, in particular its hardness. Samples from three varieties collected during the 2005 vintage season in Piedmont, Italy, were tested for their skin hardness using a texture analyser. The goal was to understand the statistical interactions between three factors—variety, cluster position and puncture point—and their influence on the grape skin hardness. A discussion on the relationship between the size of the sample used and the confidence level is also provided. RESULTS: Results of the ANOVA test showed that there is an interaction between the variety and the puncture point when measuring the skin hardness with the break energy. The position of the berry on the cluster does not affect the berry skin break energy. We also show that a sample size depend on the variety tested. CONCLUSION: The break energy is more useful in understanding the effect of the three factors on the skin hardness. Other factors that might affect the puncture test applied to grapes need to be studied in the future and the usefulness of the test in winemaking will need to be further developed. In this paper we describe the development of a quantitative PCR (qPCR) technique to detect, quantify and determine the vitality of Listeria monocytogenes in foods. The method was based on the amplification of the intergenic region spacer (IGS) between the 16S and 23S rRNA genes. A panel of more than 100 strains of Listeria spp. and non-Listeria was used in order to verify the specificity of the primers and Taqman probe and amplification signals were obtained only when L. monocytogenes DNA and RNA were loaded in the qPCR mix. Standard curves were constructed in several food matrices (milk, meat, soft cheese, fermented sausage, cured ham and ready-to-eat salad). The quantification limit was of 10(3)-10(4) cfu/g or ml, while for the determination of vitality it was 10(4)-10(5) cfu/g or ml. After an overnight enrichment in BHI at 37 degrees C also 10 cfu/g or ml could be detected in all the matrices used in this study. When we applied the protocol to food samples collected from the market or from small food processing plants, on a total number of 66 samples, 4 fresh cheeses from raw milk gave positive results prior to the overnight incubation, while 9 samples, of which only one represented by fresh meat and the others by cheeses from raw milk, were positive after the enrichment. Out of the 4 positive samples, only one could be quantified and it was determined to contain 4x10(3) cfu/g.  相似文献

The concentration of polychlorinated biphenyls (PCBs) in the air and vegetation was measured periodically in two alpine forests, during the growing season. Foliage samples from nine plant species typical of the temperate and boreal environment were collected and analyzed. Leaf concentrations of tri- and tetra-CBs showed fast response times with changing temperature and gas-phase concentrations, suggesting that a partitioning equilibrium is approached relatively rapidly (few days) in the field. Heavier compounds showed kinetically limited accumulation trends, not reaching equilibrium during the growing season. Results were used to estimate the bioconcentration factors or equilibrium plant/air partition coefficient (KPA) for each species. Values of log KPA (calculated on a mass/volume basis) ranged between 0.78 and 1.96 and were correlated to the log KOA. Uptake trends of the higher chlorinated compounds showed intraspecific differences which were partially explained by the specific leaf area (SLA).  相似文献

In this study, the microbial ecology of the blood sausage morcilla de Burgos, subjected to high hydrostatic pressure treatment (HPP), was studied by culture-dependent and -independent methods. Morcilla de Burgos is the most traditional and famous blood sausage in Spain. The producers are interested in extending its shelf-life in order to expand their market and to reduce losses attributed to spoilage. Sausage batter prior to stuffing and blood sausages HPP treated or not (control) were analyzed at 0, 9, 14, 21, 28 and 35 days of storage at 4 degrees C. Lactic acid bacteria, Pseudomonas spp. and aerobic mesophilic bacteria were investigated by traditional plating. PCR-denaturing gradient gel electrophoresis (DGGE) was used to analyze the DNA and the RNA extracted directly from the blood sausages, as well as bulk cells of LAB and Pseudomonas spp. The results showed that HPP improved the shelf life of morcilla de Burgos to 28 days in comparison with control samples. The populations responsible for spoilage, namely LAB, remained lower in HPP treated samples when compared with the control samples. Only at 35 days of storage they reached values of 10(8) cfu/g, leading to the spoilage of the product. Although, HPP affected the LAB population, they were able to recover the injury provoked by the treatment. Lastly, HPP seemed to affect differently LAB species detected. While Leuconostoc mesenteroides was completely inactivated by HPP, Weissella viridescens was able to recover and carry out the typical spoilage of the product. Pseudomonas spp. remained under detection level (<10(2) CFU/g) after the HPP treatment.  相似文献

Storage hydropower plants, which are an important component of energy production in Switzerland, can lead to hydro- and thermopeaking, affecting river habitats and organisms. In this study, we developed an approach for integrating water temperature simulations into a habitat model to assess the impact of both hydro- and thermopeaking on the availability of suitable fish habitats. We focused on the habitat requirements of juvenile brown trout (Salmo trutta) in a semi-natural braided floodplain along the Moesa River (Southern Switzerland) in early summer. First, we defined different scenarios (with and without hydropeaking) based on the local hydrological and meteorological conditions. Second, we used a two-dimensional depth-averaged hydro- and thermodynamic model to simulate the spatial distributions of water depth, flow velocity, and water temperature. Third, we applied generalized preference curves for juvenile brown trout to identify hydraulically suitable habitats, and developed a new index to assess the availability of thermally suitable habitats. Finally, we quantified the extent to which hydraulically and thermally suitable habitats overlap in space and time. During both base and peak flow phases, most of the hydraulically and thermally suitable habitats are located in the side channels. High flow conditions combined with strong cold-thermopeaking lead to a higher thermal heterogeneity. However, disconnected habitats originate in the dewatering zone, increasing the risk of stranding as well as thermal stress. By helping to better understand the effects of thermopeaking on the availability of fish habitats, our approach could contribute to the design and evaluation of ecological restoration in hydropeaking rivers.  相似文献

This work discusses the conflict between two of the main objectives of the EU Water Framework Directive: cost recovery and economic efficiency in the use of irrigation water. In the Mediterranean region, this conflict is commonly embedded in a state of under-utilization of irrigation networks managed by Water User Associations (WUAs). This under-utilization arises from factors independent of farmers’ choice, such as water shortage, crises in the sector, and changes to the Common Agricultural Policy. This prevents the facilities from operating at the minimum average cost. This paper argues that farmers should not suffer this inefficiency, which results in them paying higher water prices to cover the costs of water supply. Indeed, the application of the Water Directive should be rethought, taking into account the specific problems related to irrigation in the Mediterranean region. Based on an econometric analysis of the costs of water distribution in a WUA in Sardinia (Italy), we propose a payment system based on two components. Primarily there is a fee related to the number of hectares under irrigation. In addition there is a fee that considers the intensity of irrigation: this component is to encourage farmers to save water. The results show that the proposed approach has some desirable effects, including higher rates of cost recovery and possibly a reduction in the use of groundwater. Finally, the proposed system is not overly expensive for the rest of the community, who must bear the costs of inefficiencies in the use of irrigation water that do not depend on choices of farmers.  相似文献

Climate change is likely to have a profound effect on many agricultural variables, although the extent of its influence will vary over the course of the annual farm management cycle. Consequently, the effect of different and interconnected physical, technical and economic factors must be modeled in order to estimate the effects of climate change on agricultural productivity. Such modeling commonly makes use of indicators that summarize the among environmental factors that are considered when farmers plan their activities. This study uses net evapotranspiration (ETN), estimated using EPIC, as a proxy index for the physical factors considered by farmers when managing irrigation. Recent trends suggest that the probability distribution function of ETN may continue to change in the near future due to changes in the irrigation needs of crops. Also, water availability may continue to vary due to changes in the rainfall regime. The impacts of the uncertainties related to these changes on costs are evaluated using a Discrete Stochastic Programming model representing an irrigable Mediterranean area where limited water is supplied from a reservoir. In this context, adaptation to climate change can be best supported by improvements to the collective irrigation systems, rather than by measures aimed at individual farms such as those contained within the rural development policy.  相似文献

The article reports the results of heat transfer experimental tests on water-based TiO2 (9 wt%) and SiC (3, 6, 9 wt%) nanofluids. Measurements were performed in a two-loop test rig for immediate comparison of the thermal performances of the nanofluid with the base fluid. The convective heat transfer is evaluated in a circular pipe heated with uniform heat flux (from 20 to 240 kW/m2) and flow regimes from laminar to turbulent. Tests have been performed to compare the heat transfer of nanofluids and water at the same velocity (from 0.7 to 1.6 m/s) or Reynolds number (from 300 to 6000), and they have also been compared with values calculated from some of the most widely used correlations. The analysis of the experimental data shows a strong dependence on the parameter used, while both the nanofluid and water data have the same agreement with the calculated values. Nanofluids were manufactured through a two-step procedure: laser synthesis of nanoparticles followed by dispersion in water.  相似文献

Modified 13X zeolites and active carbons were studied for HCl adsorption from biogas employed as fuel of MCFCs. 13X zeolite was modified by addition of Cu or Zn by ion exchange or impregnation: commercial active carbon was modified by impregnation with KOH, NaOH or Na2CO3 solutions. The materials were tested for HCl adsorption in a laboratory apparatus designed to the scope. The tests were carried out under flow conditions with a 100 ppm HCl/N2 mixture at T = 40 °C and atmospheric pressure. Breakthrough curves were obtained from potentiometric analysis of HCl in the stream effluent from the adsorption cell. The zeolites modified by ion exchange were more effective for HCl adsorption than those modified by impregnation. The active carbon modified with Na2CO3 solution showed the highest adsorption capacity of 3.3 × 10−4 mol HCl g−1 at breakthrough concentration of 1 ppm.  相似文献

Micron-sized metal powders carried by a nitrogen flow were fed along the axis of a cylindrical hydrogen/oxygen diffusion flame. The particles ignited and burned in the water vapor at approximately 2500 K. Experiments were performed at atmospheric pressure. The environment in which particles burned was characterized in detail using computational fluid dynamics. The computations confirmed that the metal powders burned in water while the effect of oxygen and other oxidizing species could be neglected. Combustion was characterized experimentally for micron-sized powders of both aluminum and magnesium. Particle size distributions were measured using low-angle laser light scattering. Optical emission of the burning particles was recorded using filtered photomultiplier tubes. Measured durations of individual particle emission pulses were assumed to represent their burn times; these data were classified into logarithmically spaced time bins. The distribution of the particle burn times was correlated with their size distributions assuming that larger size particles burned longer. It was observed that correlation between the burn times, t, and particle diameters, D, can be approximately described as t ∼ D0.64 and t ∼ D0.68 for aluminum and magnesium powders, respectively. The results were compared to previous reports and possible reasons for discrepancies between the present and earlier results were discussed.  相似文献
