The cell wall is essential to preserve osmotic integrity of yeast cells. Some phenotypic traits of cell wall mutants suggest that, as a result of a weakening of the cell wall, hypo-osmotic stress-like conditions are created. Consequent expansion of the cell wall and stretching of the plasma membrane trigger a complex response to prevent cell lysis. In this work we examined two conditions that generate a cell wall and membrane stress: one is represented by the cell wall mutant gas1Delta and the other by a hypo-osmotic shock. We examined the actin cytoskeleton and the role of the cell wall sensors Wsc1p and Mid2p in these stress conditions. In the gas1 null mutant cells, which lack a beta(1,3)-glucanosyltransferase activity required for cell wall assembly, a constitutive marked depolarization of actin cytoskeleton was found. In a hypo-osmotic shock wild-type cells showed a transient depolarization of actin cytoskeleton. The percentage of depolarized cells was maximal at 30 min after the shift and then progressively decreased until cells reached a new steady-state condition. The maximal response was proportional to the magnitude of the difference in the external osmolarity before and after the shift within a given range of osmolarities. Loss of Wsc1p specifically delayed the repolarization of the actin cytoskeleton, whereas Wsc1p and Mid2p were essential for the maintenance of cell integrity in gas1Delta cells. The control of actin cytoskeleton is an important element in the context of the compensatory response to cell wall weakening. Wsc1p appears to be an important regulator of the actin network rearrangements in conditions of cell wall expansion and membrane stretching.  相似文献

Computational modeling of human arteries has been broadly employed to investigate the relationships between geometry, hemodynamics and vascular disease. Recent developments in modeling techniques have made it possible to perform such analyses on realistic geometries acquired noninvasively and, thus, have opened up the possibility to extend the investigation to populations of subjects. However, for this to be feasible, novel methods for the comparison of the data obtained from large numbers of realistic models in the presence of anatomic variability must be developed. In this paper, we present an automatic technique for the objective comparison of distributions of geometric and hemodynamic quantities over the surface of bifurcating vessels. The method is based on centerlines and consists of robustly decomposing the surface into its constituent branches and mapping each branch onto a template parametric plane. The application of the technique to realistic data demonstrates how similar results are obtained over similar geometries, allowing for proper model-to-model comparison. Thanks to the computational and differential geometry criteria adopted, the method does not depend on user-defined parameters or user interaction, it is flexible with respect to the bifurcation geometry and it is readily extendible to more complex configurations of interconnecting vessels.  相似文献

We discuss the shifts the frequency standard is corrected for and the procedure used for the accuracy evaluation. The optical properties of thin Sc films deposited in ultrahigh-vacuum conditions have been investigated in the 6.7-174.4-nm spectral range. We measured transmittance and multiangle reflectance in situ in the 53.6-174.4 nm spectral range and used these measurements to obtain the complex refractive index of a Sc film at every individual wavelength investigated. Transmittance measurements were made of Sc samples that were deposited over grids coated with a support C film. The transmittance and the extinction coefficient of Sc films at wavelengths shorter than 30 nm were measured ex situ. The ex situ samples were protected with an additional top C film before their removal from vacuum. To our knowledge, these are the first optical measurements of Sc films reported in the spectral ranges cited.  相似文献

The Michelson Interferometer for Passive Atmospheric Sounding (MIPAS) is a limb-scanning spectrometer that has operated onboard the Environmental Satellite since the end of March 2002. Common features of limb-scanning experiments are both high vertical resolution and poor horizontal resolution. We exploit the two-dimensional geo-fit retrieval approach [Appl. Opt. 40, 1872-1875 (2001)] to investigate the possibility of improving the horizontal resolution of MIPAS measurements. Two different strategies are considered for this purpose, one exploiting the possibility (offered by the geo-fit analysis method) for an arbitrary definition of the retrieval grid, the other based on the possibility of saving measurement time by degrading the spectral resolution of the interferometer. The performances of the two strategies are compared in terms of the trade-off between the attained horizontal resolution and the retrieval precision. We find that for ozone it is possible to improve by a factor of 2 the horizontal resolution, which in the nominal measurement plan is approximately 530 km. This improvement corresponds to a degradation of the retrieval precision, which on average varies from a factor of 1.4 to 2.5, depending on the adopted spectral resolution.  相似文献

Performances are presented of three classes of imaging slit spectrometers for extended sources with aberration-corrected gratings. A general analytical expression for minimizing off-axis grating aberrations is obtained, and it is demonstrated that these aberrations are minimized when the spectrometer is operated at a magnification higher than unity. Classical designs with toroidal uniform-line-spaced (TULS) or spherical varied-line-space (SVLS) gratings are compared with a new class of designs that utilize toroidal varied-line-space (TVLS) gratings. Although TULS and SVLS designs with two stigmatic points can be designed to operate at near-unity magnification with excellent on-axis spectral and spatial resolutions, they cannot be made to satisfy the general off-axis condition, and so their off-axis performances are not optimum. On the contrary TVLS designs with two stigmatic points can be operated at almost any magnification, thus satisfying the off-axis condition perfectly. Such designs are suitable for imaging spectrometer observations that require an extended field of view.  相似文献

This paper is an answer to the need of finding the optimal solution for the throttling system in refrigerating machines using CO2 as working fluid; such a solution must combine reliability, low installation cost and high energy efficiency. To this purpose, different expansion systems are compared by means of a simulation programme, including a new one, operating with a differential valve, a liquid receiver and a thermostatic valve. The typical compression refrigerating cycle performed by CO2 involves transcritical operations and therefore the upper pressure needs to be adjusted to the optimal value, that, unlike the traditional cycle, is not determined by heat transfer. The innovative system here proposed shows an intrinsic self-adjusting capability that leads to COP values quite close to the maximum ones when a fixed suitable value of the differential pressure is chosen, even if the temperature of the secondary fluid varies to a large extent.  相似文献

Based on the premise that learning and inertia develop and operatein organizations simultaneously, this paper examines under whatconditions adaptation or selection will take place. We haveanalyzed the process of technological scope expansion amongItalian automobile firms, and found that firms prone to expandingtheir technological competencies tend to be either specificallyorganized to benefit from variation-based learning or do soin response to pressures from competitive intensity in theirenvironment. But these effects hinge on the structure of themarket that evolves as the industry consolidates. We also examinethe failure rates of Italian auto firms and find that implementinga core change generates negative repercussions for survivalbut this effect can be countered by properties of the organizationaldesign that facilitate variation-based learning. Our resultssuggest that organization theory will benefit if ecologicaland learning theories are further integrated in future research.  相似文献

Microstructured optical fibers (MOFs) with small hole-to-hole spacing and large airholes are designed to compensate the anomalous dispersion and the dispersion slope of single-mode fibers. The geometrical parameters that characterize triangular MOFs are chosen to optimize the fiber length and the compensation over a wide wavelength range. A proper design of the photonic crystal fiber geometry allows us to achieve dispersion values of approximately -1700 ps nm(-1) km(-1) at 1550 nm and to compensate the dispersion of standard fibers within +/- 0.5 ps nm(-1) km(-1) over a 100-nm range. The MOF dispersion properties have been studied by means of a numerical simulator for modal analysis based on the finite-element method.  相似文献
