Two glial cell types surround olfactory axons and glomeruli in the olfactory bulb (OB) and may influence synapse development and regeneration. OB astrocytes resemble type-1 astrocytes, and OB ensheathing cells resemble non-myelinating Schwann cells. We have produced clonal OB astrocyte and ensheathing cell lines from rat neonatal and adult OB cultures by SV40 large T antigen transduction. These cell lines have been characterized by morphology, growth characteristics, immunophenotype, and ability to promote neurite outgrowth in vitro. Neonatal and adult ensheathing cell lines were found to support higher neurite outgrowth than OB astrocyte lines. Neonatal OB astrocyte lines were of two types, high and low outgrowth support. The low support astrocyte lines express J1 and a chondroitin sulfate-containing proteoglycan as do astrocytes encircling the neonatal glomeruli in vivo. The adult OB astrocyte cell lines supported lower levels of outgrowth than adult ensheathing cell lines. These results are consistent with a positive role for ensheathing cells in OB synapse regeneration, in vivo. Further, based on our results, we hypothesize that ensheathing cells and high-outgrowth astrocytes facilitate axon growth in vivo, while low outgrowth astrocytes inhibit axon growth and may facilitate glomerulus formation.  相似文献

We have studied the inhibitory action of long- and short-chain fatty acids on hepatic glucose utilization in hepatocytes isolated from fasted rats. The rates of hepatic glucose phosphorylation and glycolysis were determined from the tritiated products of [2-3H] and [6-3H]glucose metabolism, respectively. The difference between these was taken as an estimate of the 'cycling' between glucose and glucose-6-phosphate. In the presence of 40 mM glucose this cycling was estimated at 0.68 mumol/min/g wet wt. Glucose phosphorylation was unaffected during palmitate and hexanoate oxidation to ketone bodies but glycolysis was inhibited. The rate of glucose cycling was increased during this phase to 1.25 mumol/min/g. Following the complete metabolism of the fatty acids, glycolysis was reinstated and cycling rates returned to control levels. Hepatic glucose cycling appears to be an important component of the glucose/fatty acid cycle.  相似文献

It has previously been shown that the Copenhagen (COP) rat contains several genetic loci that contribute to its mammary tumor-resistant phenotype after 7,12-dimethylbenz(a)anthracene (DMBA) administration. One of these loci, mammary carcinoma susceptibility 1 (Mcs1), is located on the centromeric end of chromosome 2 and appears to act in a semidominant fashion. To confirm the existence and independent action of this locus and also aid in the identification of the physical location of the Mcs1 gene, congenic lines were generated by transferring the Mcs1 COP allele onto a Wistar Furth (WF) genetic background. Male carriers were genotyped using microsatellite markers spanning 20-30 cM of the Mcs1 locus. One of the congenic lines minimally retained the COP allele at D2Mit29 on the centromeric end of chromosome 2 and extended distally to D2Rat201. Heterozygous Mcs1 carrier rats were interbred, and the female offspring were treated with DMBA. The female rats from the Mcs1 congenic line that carried one or two COP alleles of the Mcs1 region had a significantly reduced (65 and 85%, respectively) tumor development (P < 0.001) compared with rats carrying zero COP alleles at this locus. A WF.COP-D2Mit29/D2Rat201 homozygous congenic strain derived at the N10 generation was treated with DMBA, and the COP homozygous rats developed 1.5 +/- 0.3 carcinomas/rat versus 6.3 +/- 0.5 in WF control rats (P < 0.0001). Fine mapping of this congenic interval using several recombinant lines identified three genetic loci within the Mcs1 congenic region that independently supported a tumor resistance phenotype. These genetic loci have been termed Mcs1a, Mcs1b, and Mcs1c. In rats for which each locus was homozygous for the COP allele, tumor development was reduced by approximately 60% compared with littermate controls. The identification of these independent loci within the Mcs1 COP allele provide a model of the genetic complexity of cancer.  相似文献

We have previously shown that the tumor suppressor gene for hepatocellular carcinoma (HCC) without cirrhosis may be located on chromosome 5q35-qter. In this study, we analyzed nine cases of primary HCC without cirrhosis using probes from the MCC and APC genes, which are in the region 5q21-22. None of the informative cases had allele loss detected by these probes, whereas the probe lambda MS8 for the region 5q35-qter showed allele loss in six out of six informative cases. The results confirm that the putative tumor suppressor gene for HCC without cirrhosis on chromosome 5q is distinct from the MCC and APC genes.  相似文献

For environmental considerations, the substitution of the conventionally used oil-based grinding fluids has nowadays become strongly recommended. Although several alternatives have been proposed, cryogenic cooling by liquid nitrogen is the non-polluting coolant that has been given relatively more attention because of its very low temperature. In this investigation, in order to contribute to developing this promising cooling mode, its beneficial effects on the ground surface integrity of the AISI 304 stainless steel and their consequences on the fatigue lifetime are explored. Results of this investigation show that grinding under cryogenic cooling mode generates surfaces with lower roughness, less defects, higher work hardening and less tensile residual stresses than those obtained on surfaces ground under oil-based grinding fluid. These surface enhancements result into substantial improvements in the fatigue behaviour of components ground under this cooling mode. An increasing rate of almost 15% of the endurance limit at 2 × 106 cycles could be realized. SEM analyses of the fatigue fracture surfaces have shown that the fatigue cracks observed on the specimens ground under cryogenic cooling are shorter (i.e., 30-50 μm) than those generated under oil-based cooling mode (i.e., 150-200 μm). The realized improvements in the surface integrity and in the fatigue behaviour are thought to be related to the reduction of the grinding zone temperature observed under cryogenic cooling, as no significant differences between the grinding force components for both cooling modes have been observed.  相似文献

Erosion is one of the most serious problems in various gas and liquid flow passages such as flow in pipes, pumps, turbines, compressors and many other devices. Sand presence causes loss of pipe wall thickness that can lead to pipe erosion, frequent failures and loss of expensive production time. The importance of this problem is mainly due to many related engineering applications, viz. heat exchangers. In order to reduce the frequency of such pipe erosions, caps in the form of replaceable pipes are protruded in the sudden contraction regions which are exposed to most of the serious erosion rates. In the present work, numerical investigation of the erosion of a pipe protruded in a sudden contraction is presented. The turbulent, steady, 2-D axi-symmetric flow inside an axi-symmetric abrupt contraction pipe with a pipe protrusion embedded in it was solved by steady-state time averaged conservation equations of mass and momentum along with two equation model for turbulence. Particles are tracked using Lagrangian particle tracking. An erosion model was employed to investigate the erosion phenomena for the given geometry. The influence of the different parameters such as the inlet flow velocity (3–10 m/s), the particle diameter (10–400 μm), the protruded pipe geometry (thickness T=1–5 mm and depth H=2–5 mm) and the pipe contraction ratio (Cr=0.25–0.5) on the erosion of pipe protrusion was investigated. Correlations for the influence of inlet flow velocity, depth and thickness of the protruded pipe on the erosion rate are presented.  相似文献

BACKGROUND: Many species of bats have two sets of submandibular glands, principal and accessory. The accessory gland may resemble the principal one but more often shows wide morphological divergence. The free-tailed bat, Tadarida thersites, is very unusual in that it has two sets of parotid glands rather than binary submandibular glands. We studied the ultrastructure of the principal parotid gland to establish a baseline for comparison with the accessory parotid. METHODS: Two specimens of adult free-tailed bats, one male and one female, were live-trapped in western Kenya. Parotid glands were fixed for electron microscopy using a protocol expressly designed for field fixation and then embedded by conventional means. RESULTS: Histologically, the principal parotid is a typical serous gland. The secretory granules of the endpiece cells have an unusual substructure in that they contain variable numbers of lucent halos and one or several spherules. Intercalated duct cells contain a significant number of dense, serous-like granules. Striated ducts have the usual basal configuration of mitochondria and folded plasma membranes, but the supranuclear cytoplasm contains many small, dense granules, so that these ducts resemble the granular convoluted tubules found in the submandibular glands of many families of rodents. The apices of the duct cells have a peculiar contour--the luminal surfaces obliquely invaginate into the apical cytoplasm, so that in thin section the luminal membranes appear to be underlaid by a layer of vacuoles. CONCLUSION: Although the principal parotid gland of the free-tailed bat shows some distinctive, species-specific ultrastructural features, it basically is similar to the parotid gland in two other molossid bats, Tadarida brasiliensis and Molossus molossus. The distinctive features in the principal parotid gland of T. thersites might relate to its feeding on hard-bodied insects and perhaps to the production of lysozyme.  相似文献
