RT-qPCR is the gold standard and the most commonly used method for measuring gene expression. Selection of appropriate reference gene(s) for normalization is a crucial part of RT-qPCR experimental design, which allows accurate quantification and reliability of the results. Because there is no universal reference gene and even commonly used housekeeping genes’ expression can vary under certain conditions, careful selection of an appropriate internal control must be performed for each cell type or tissue and experimental design. The aim of this study was to identify the most stable reference genes during osteogenic differentiation of the human osteosarcoma cell lines MG-63, HOS, and SaOS-2 using the geNorm, NormFinder, and BestKeeper statistical algorithms. Our results show that TBP, PPIA, YWHAZ, and EF1A1 are the most stably expressed genes, while ACTB, and 18S rRNA expressions are most variable. These data provide a basis for future RT-qPCR normalizations when studying gene expression during osteogenic differentiation, for example, in studies of osteoporosis and other bone diseases.  相似文献

Climbing plants must reach supports and navigate gaps to colonize trees. This requires a structural organization ensuring the rigidity of so-called ‘searcher’ stems. Cacti have succulent stems adapted for water storage in dry habitats. We investigate how a climbing cactus Selenicereus setaceus develops its stem structure and succulent tissues for climbing. We applied a ‘wide scale’ approach combining field-based bending, tensile and swellability tests with fine-scale rheological, compression and anatomical analyses in laboratory conditions. Gap-spanning ‘searcher’ stems rely significantly on the soft cortex and outer skin of the stem for rigidity in bending (60–94%). A woody core contributes significantly to axial and radial compressive strength (80%). Rheological tests indicated that storage moduli were consistently higher than loss moduli indicating that the mucilaginous cortical tissue behaved like a viscoelastic solid with properties similar to physical or chemical hydrogels. Rheological and compression properties of the soft tissue changed from young to old stages. The hydrogel–skin composite is a multi-functional structure contributing to rigidity in searcher stems but also imparting compliance and benign failure in environmental situations when stems must fail. Soft tissue composites changing in function via changes in development and turgescence have a great potential for exploring candidate materials for technical applications.  相似文献

In this paper we present a general strategy for finding efficient permutation routes in parallel networks. Among the popular parallel networks to which the strategy applies are mesh networks, hypercube networks, hypercube-derivative networks, ring networks, and star networks. The routes produced are generally congestion-free and take a number of routing steps that is within a small constant factor of the diameter of the network. Our basic strategy is derived from an algorithm that finds (in polynomial time) efficient permutation routes for aproduct network, G×H, given efficient permutation routes forG andH. We investigate the use of this algorithm for routingmultiple permutations and extend its applicability to a wide class of graphs, including several families ofCayley graphs. Abstract: Export of cocoa beans is of great economic importance in Ghana and several other tropical countries. Raw cocoa has an astringent, unpleasant taste, and flavor, and has to be fermented, dried, and roasted to obtain the characteristic cocoa flavor and taste. In an attempt to obtain a deeper understanding of the changes in the cocoa beans during fermentation and investigate the possibility of future development of objective methods for assessing the degree of fermentation, a novel combination of methods including cut test, colorimetry, fluorescence spectroscopy, NIR spectroscopy, and GC-MS evaluated by chemometric methods was used to examine cocoa beans sampled at different durations of fermentation and samples representing fully fermented and dried beans from all cocoa growing regions of Ghana. Using colorimetry it was found that samples moved towards higher a* and b* values as fermentation progressed. Furthermore, the degree of fermentation could, in general, be well described by the spectroscopic methods used. In addition, it was possible to link analysis of volatile compounds with predictions of fermentation time. Fermented and dried cocoa beans from the Volta and the Western regions clustered separately in the score plots based on colorimetric, fluorescence, NIR, and GC-MS indicating regional differences in the composition of Ghanaian cocoa beans. The study demonstrates the potential of colorimetry and spectroscopic methods as valuable tools for determining the fermentation degree of cocoa beans. Using GC-MS it was possible to demonstrate the formation of several important aroma compounds such 2-phenylethyl acetate, propionic acid, and acetoin and the breakdown of others like diacetyl during fermentation. Practical Application: The present study demonstrates the potential of using colorimetry and spectroscopic methods as objective methods for determining cocoa bean quality along the processing chain. Development of objective methods for determining cocoa bean quality will be of great importance for quality insurance within the fields of cocoa processing and raw material control in chocolate producing companies.  相似文献

Extensive reviews of research are available on the use of ascorbic acid, and its consequent degradation pathways, in physiological conditions or food matrices. However, very little information can be found for wine-related systems. This review highlights the relevant chemistry and reactivity of ascorbic acid with a focus on its behavior and potential behavior in a wine environment. The review describes the use of ascorbic acid as a complementary antioxidant preservative to sulfur dioxide along with the metal-catalyzed and radical-dependent manner by which it achieves this role. The relevant degradation products of ascorbic acid in aerobic and anaerobic conditions are presented as well as the interaction of these degradation products with sulfur dioxide and other wine-relevant sulfur compounds. Limitations in existing knowledge, especially regarding the crossover between the antioxidant and pro-oxidant roles of ascorbic acid, are identified.  相似文献

Three commercial phage sensitive Lactobacillus delbrueckii strains (identified as Ab(1), YSD V and Ib(3)), and four spontaneous phage-resistant mutants isolated from them were tested for their capacity to activate the gut mucosal immune response in mice, as indicated by the numbers of IgA-producing cells. Random Amplified Polymorphic DNA (RAPD) analysis revealed a strong genetic homology between the sensitive strains and their respective derivatives. The phage-resistant mutants exhibited high levels of phage resistance, elevated stability of this phenotype and technological properties comparable to those of their respective parent strains. The tolerance to acidic conditions, bile salts and lysozyme was strain dependent and total cell viability losses as a result of exposure to all three stresses ranged from 2.0 to 3.7 log units. All the strains were highly resistant to a simulated gastric solution of pH 3, while significant additional losses in cell viability were observed when acid treated cells were exposed to bile salts and lysozyme. BALB/c mice received pure cultures of Lb. delbrueckii sensitive and phage-resistant strains for 2, 5 or 7 consecutive days. The ability of the parent strains to activate the small intestine immune response was preserved or enhanced in phage-resistant mutants. The maximal proliferation of IgA(+) cells was observed at day 5 or 7, depending on the strain. Mutants isolated in this study using natural selection strategies had improved phage resistance, adequate technological properties and satisfactory gut mucosal immunostimulation ability, and so would be good candidates for industrial applications in functional foods.  相似文献

Hen egg yolk is an ideal example of natural supramolecular assemblies of lipids and proteins with different organization levels. These assemblies are mainly due to interactions between proteins and phospholipids, and these interactions are essential in understanding and controlling the production of food made with yolk, and particularly emulsions. Furthermore, these assemblies can be modulated by external constraints among which thermo‐mechanical and high‐pressure treatments. This review focuses on multi‐scale structures present in egg yolk, and their modulation by processes, in relation with their emulsifying properties. Egg yolk is mainly composed of two fractions—plasma and granules—which are natural nano‐ and micro‐assemblies. These two fractions possess different composition, structures and functionalities and exhibit specific behaviour under treatments such as high pressure and temperature. Plasma contains a large quantity of lipids structured as lipoproteins (low‐density lipoproteins), whereas granules are mainly composed of proteins aggregated in micrometric assemblies. If plasma is responsible for the important emulsifying properties of yolk, granules bring interesting emulsifying properties when assemblies are in the form of micelles in presence of salts. High‐pressure or thermal treatments, applied before or after emulsion fabrication, alter their functionalities and could be used to commercially exploit these fractions. © 2013 Society of Chemical Industry  相似文献

Virtualized datacenters and clouds are being increasingly considered for traditional High-Performance Computing (HPC) workloads that have typically targeted Grids and conventional HPC platforms. However, maximizing energy efficiency and utilization of datacenter resources, and minimizing undesired thermal behavior while ensuring application performance and other Quality of Service (QoS) guarantees for HPC applications requires careful consideration of important and extremely challenging tradeoffs. Virtual Machine (VM) migration is one of the most common techniques used to alleviate thermal anomalies (i.e., hotspots) in cloud datacenter servers as it reduces load and, hence, the server utilization. In this article, the benefits of using other techniques such as voltage scaling and pinning (traditionally used for reducing energy consumption) for thermal management over VM migrations are studied in detail. As no single technique is the most efficient to meet temperature/performance optimization goals in all situations, an autonomic approach that performs energy-efficient thermal management while ensuring the QoS delivered to the users is proposed. To address the problem of VM allocation that arises during VM migrations, an innovative application-centric energy-aware strategy for Virtual Machine (VM) allocation is proposed. The proposed strategy ensures high resource utilization and energy efficiency through VM consolidation while satisfying application QoS by exploiting knowledge obtained through application profiling along multiple dimensions (CPU, memory, and network bandwidth utilization). To support our arguments, we present the results obtained from an experimental evaluation on real hardware using HPC workloads under different scenarios.  相似文献

由于用传统的刚体匹配方法难以解决待匹配图象之间的结构差异 ,因此需要引入变形模型来进行图象的非刚体匹配 .为此提出了一种利用混合弹性模型 (HEM)来解决图象变形匹配问题的新方法 .该方法不需要预先提取图象的特征 ,而是直接利用匹配图象之间的灰度信息来实现图象之间的匹配 .匹配时 ,首先通过基于主轴的方法来实现两幅图象之间的全局仿射匹配 ;然后利用线性弹簧网模型来求取两幅图象之间的相关性 ,并进一步利用薄板样条模型来实现两幅图象的变形匹配 .该方法在匹配过程中还采取了多分辨率匹配策略 ,合成图象和医学脑图象的实验结果表明 ,该方法是有效的  相似文献
