An effective strategy is highly desirable for preventing acute kidney injury (AKI) to chronic kidney disease (CKD) transition. Thioredoxin-1 (Trx), a redox-active protein that has anti-oxidative and anti-inflammatory properties, would be a candidate for this but its short half-life limits its clinical application. In this study, we examined the renoprotective effect of long-acting Trx that is comprised of human albumin and Trx (HSA-Trx) against AKI to CKD transition. AKI to CKD mice were created by renal ischemia-reperfusion (IR). From day 1 to day 14 after renal IR, the recovery of renal function was accelerated by HSA-Trx administration. On day 14, HSA-Trx reduced renal fibrosis compared with PBS treatment. At the early phase of fibrogenesis (day 7), HSA-Trx treatment suppressed renal oxidative stress, pro-inflammatory cytokine production and macrophage infiltration, thus ameliorating tubular injury and fibrosis. In addition, HSA-Trx treatment inhibited G2/M cell cycle arrest and apoptosis in renal tubular cells. While renal Trx protein levels were decreased after renal IR, the levels were recovered by HSA-Trx treatment. Together, HSA-Trx has potential for use in the treatment of AKI to CKD transition via its effects of modulating oxidative stress and inflammation.  相似文献

Despite advances in medicine, cardiac disease remains an increasing health problem associated with a high mortality rate. Maladaptive cardiac remodeling, such as cardiac hypertrophy and fibrosis, is a risk factor for heart failure; therefore, it is critical to identify new therapeutic targets. Failing heart is reported to be associated with hyper-ubiquitylation and impairment of the ubiquitin–proteasome system, indicating an importance of ubiquitylation in the development of cardiac disease. Ubiquitylation is a post-translational modification that plays a pivotal role in protein function and degradation. In 1995, homologous to E6AP C-terminus (HECT) type E3 ligases were discovered. E3 ligases are key enzymes in ubiquitylation and are classified into three families: really interesting new genes (RING), HECT, and RING-between-RINGs (RBRs). Moreover, 28 HECT-type E3 ligases have been identified in human beings. It is well conserved in evolution and is characterized by the direct attachment of ubiquitin to substrates. HECT-type E3 ligase is reported to be involved in a wide range of human diseases and health. The role of HECT-type E3 ligases in the development of cardiac diseases has been uncovered in the last decade. There are only a few review articles summarizing recent advancements regarding HECT-type E3 ligase in the field of cardiac disease. This study focused on cardiac remodeling and described the role of HECT-type E3 ligases in the development of cardiac disease. Moreover, this study revealed that the current knowledge could be exploited for the development of new clinical therapies.  相似文献

PV output of multi-junction cells is strongly influenced by spectrum change. The influence of Sun height was quantitatively analyzed, considering seasonally and daily changes of spectrum. The new model also considered the presence of clouds. The influence of daily random fluctuation of spectrum was shown averaged out in the integration of yearly PV output fluctuation. It was suggested that the overall mismatch loss by the change of Sun height and Sun orbital would be <4% for III–V multi-junction cells for concentration application, when the bandgap of each junction was well balanced. The sensitivity of spectrum fluctuation was shown enlarged with the discrepancy of current matching conditions.  相似文献

Better radiation resistance of InP-based solar cells such as InGaP and InGaAsP cells, and minority-carrier-injection-enhanced annealing phenomena of radiation-induced defects in those materials previously found by the authors has been analyzed by considering introduction rates and annealing behavior of radiation-induced defects in InP-related materials. Radiation resistance of InP-related materials is found to be explained by lower damage coefficients and band-gap energy effects on solar cell degradation compared to other materials.  相似文献

A low-cost and simple on-site technique for genotyping single nucleotide polymorphisms (SNPs) was developed. The technique is based on allele-specific primer PCR and the recently developed bead arrays in a single tip technique. Microwave heating, because of its advantages of direct and rapid heating of materials, has the potential to be employed as a novel regeneration method of desiccant rotors in humidity conditioners. We proposed a combined regeneration process, which combines microwave heating and conventional hot-air heating. The system is expected to achieve high heating rate during an initial regeneration period by assisting water desorption using the additional energy of the microwave. In this study, the regeneration characteristics of a desiccant rotor were experimentally investigated under conditions of microwave heating, hot-air heating, and combined heating at various microwave powers and hot-air temperatures. The effectiveness of the combined regeneration was evaluated in terms of the regeneration ratio, the initial regeneration rate, the temperature distribution in the rotor, and finally in terms of the energy consumption. It was demonstrated that combined heating was effective at leveling non-uniform temperature distribution in the rotor. Combined heating achieved higher ratios and initial rates in regeneration compared to just microwave and hot-air heating. This result was obviously attributed to the additional input of microwave energy, resulting that average rotor temperature increased by microwave absorption of rotor. Moreover, it was also effective for enhancement of regeneration to level the temperature distribution in the rotor by combination of two heating methods with different heating mechanisms. Both the initial regeneration rate and the equilibrium regeneration ratio for combined heating were found to increase as the microwave power increased. A linear relationship was observed with respect to microwave power. From the viewpoint of energy consumption, it may be possible to apply combined and microwave heating to humidity control systems that switch between adsorption and regeneration in short cycle times, if the conversion and absorption efficiencies of the microwave are significantly improved.  相似文献

The biodegradation characteristics of palm oil mill effluent (POME) and the related microbial community were studied in both actual sequential anaerobic ponds in Malaysia and enrichment cultures. The significant degradation of the POME was observed in the second pond, in which the temperature was 35-37 °C. In this pond, biodegradation of major long chain fatty acids (LCFA), such as palmitic acid (C16:0) and oleic acid (C18:1), was also confirmed. The enrichment culture experiment was conducted with different feeding substrates, i.e. POME, C16:0 and C18:1, at 35 °C. Good recovery of methane indicated biodegradation of feeds in the POME and C16:0 enrichments. The methane production rate of the C18:1 enrichment was slower than other substrates and inhibition of methanogenesis was frequently observed. Denaturing gradient gel electrophoresis (DGGE) analyses indicated the existence of LCFA-degrading bacteria, such as the genus Syntrophus and Syntorophomonas, in all enrichment cultures operated at 35 °C. Anaerobic degradation of the POME under mesophilic conditions was stably processed as compared with thermophilic conditions.  相似文献

The glucosyl transfer reaction of kojibiose phosphorylase (KP; EC 2.4.1.230) was examined using glycerol or myo-inositol as an acceptor. In the case of glycerol, KP produced two main transfer products: saccharides A and B. The structure of saccharide A was O-alpha-D-glucopyranosyl-(1-->1)-glycerol and that of saccharide B was O-alpha-D-glucopyranosyl-(1-->2)-O-alpha-D-glucopyranosyl-(1-->1)-glycerol. These results show that KP transferred a glucose residue to the hydroxyl group at position 1 of glycerol. On the other hand, when myo-inositol was used as an acceptor, KP produced four transfer products: saccharides 1-4. The structures of saccharides 1 and 2 were O-alpha-D-glucopyranosyl-(1-->1)- and O-alpha-D-glucopyranosyl-(1-->5)-myo-inositol, respectively; those of saccharides 3 and 4 were O-alpha-D-glucopyranosyl-(1-->2)-O-alpha-D-glucopyranosyl-(1-->1)- and O-alpha-D-glucopyranosyl-(1-->2)-O-alpha-D-glucopyranosyl-(1-->5)-myo-inositol, respectively. KP transferred a glucose residue to the hydroxyl group at position 1 or 5 of myo-inositol. On the basis of the structures of their glucosyl transfer products, glycerol and myo-inositol were found to have a common structure with three hydroxyl groups corresponding to the hydroxyl group of the glucose molecule at positions 2, 3 and 4. The conformation of these three hydroxyl groups in the structure is equatorial. This structure is the substrate recognition site of KP. It has been suggested that KP strictly recognizes the structures of glycerol and myo-inositol, and catalyzes the transfer reaction of a glucose residue to the hydroxyl group at position 1 in glycerol, and at position 1 or 5 in myo-inositol, corresponding to position 2 in glucose.  相似文献

We fabricated two types of polyacene capacitor with extremely stable polyacenic semiconductor (PAS) as the positive and negative electrodes. The first one is a coin-type PAS capacitor (six different sizes), which possesses large capacity with high reliability. Its capacity is much larger than that of the conventional electric double-layer capacitor which uses activated carbon as electrode. PAS capacitor can maintain more than 70% of the initial capacity even after 100 000 cycles. Moreover, this capacitor can be charged and discharged in a few minutes as well as at low rate. The second one is a cylinder-type PAS capacitor (diameter: 18 mm, height: 65 mm) which shows high capacity of 100 F and can discharge at the extremely high rate of 80 C. The coin-type PAS capacitor is currently used for memory back-up of electrical and communication equipment, and the cylinder-type is considered to be useful as power back-up for starting drive parts of electric equipment which needs high power density.  相似文献

With the application of an energy rebound circuit for the pulsewidth modulation (PWM)-controlled rectifier, an improved current source gate turn-off thyristor (GTO) inverter system capable of producing sinusoidal inputs and outputs is presented. The energy rebound circuit in the inverter is used to turn off the thyristors in the rectifier for applying PWM control techniques and also to clamp the spike voltage produced in the inverter circuit. The principles and circuit operations of this system are described in detail, and requirements for PWM control of the rectifier are investigated. The conditions for satisfying the requirements of the energy rebound circuit are analyzed by experimental and theoretical methods. The PWM-controlled current source GTO inverter system is used to drive an eight-pole 400 Hz 5.5 kW induction motor. Analytical and experimental results, as well as the characteristics of the motor drives, are given  相似文献
