Despite the existence of an impressive body of work on human immune responses against filarial infections, the occurrence of a protective response to infection remains unclear. Here, we use a combined modelling and comparative data analysis framework to address this issue for human infections with the filarial parasite, Wuchereria bancrofti. By analogy with previous work, the analysis involves the comparison of observed field patterns of infection with epidemiological patterns predicted by a mathematical model of parasite immunity. Unlike most other human helminths, which are transmitted by ingestion or dermal penetration, exposure to infection with lymphatic filariasis can be measured explicitly in terms of vector mosquito biting rates, thereby also allowing, probably for the first time, examination of the suggested role of exposure in generating herd immunity to macroparasites. Observed field patterns in this study were derived from 19 different published studies, which gave parallel estimates of community exposure rates and the corresponding age--prevalence patterns of infection, while predictions of the epidemiological impact of herd immunity were obtained using a catalytic model framework. The results provide the first conclusive evidence to date that variations in the observed age--prevalence patterns of infection in filariasis can be effectively explained by the occurrence of an exposure-driven acquisition of herd immunity. We discuss this result in terms of implications for the new World Health Organization-led initiative for the global control of this parasitic disease.  相似文献

The reduction of nitric oxide with ammonia on an unsupported iron oxide catalyst has been studied in a continuous-flow recycle reactor using simulated flue gas. The responses of the employed reactor system to step and pulse inputs of tracer indicate that the system could be regarded as a continuous stirred tank reactor (CSTR). Preliminary tests were carried out to determine the effect of temperature and particle size on the measured reaction rates. Additional experiments were performed in order to study the influence of oxygen and water concentration on these rates. A gas chromatographic system has been developed to analyze the gas components NO, N2O, NO2, NH3, H2O, O2, CO2 and N2. In addition, the concentrations of NO and NO2 were measured with a nondisperse infrared (NDUV/NDIR) analyzer.  相似文献

Dotarizine is a new diphenylmethylpiperazine derivative with Ca2+ channel blocking properties and inhibitory effects on 5-HT2A and 5-HT2C receptors. Previous pilot studies in healthy volunteers demonstrated a good tolerability after single and multiple dosing. Dotarizine appeared to be rapidly and extensively metabolized to an active compound (FI-6020). We aimed to study the physiologic, subjective and psychomotor acute effects of oral dotarizine after single dose administration, to evaluate the tolerability and safety after multiple dosing over 2 weeks, and to study the pharmacokinetic parameters and linearity after single and multiple administration. Two different studies were carried out in 2 groups of 8 healthy male volunteers. Oral single doses of dotarizine 50, 100 and 200 mg were administered in a randomized, double-blind, crossover, placebo-controlled trial. Oral doses of 50 mg twice daily were administered in an open trial over 14 days. Drug effect assessments included vital signs, collection of adverse events, ECG and blood and urine safety evaluations, subjective effects, psychomotor performance tasks and blood sampling. Dotarizine and its metabolite were determined by gas chromatography with N-P detector. The results showed a good tolerability of dotarizine after single oral doses as well as multiple oral doses over 14 days. No clinically relevant adverse events were reported during the study. The highest single dose (200 mg) produced a slight increase in sedation-related symptoms as well as a slight impairment in psychomotor performance tasks. Dotarizine and its major metabolite proved linear kinetics at single doses. The administration of oral doses of dotarizine 50 mg b.i.d. reached the steady state after the 7th day of treatment. The pharmacokinetic parameters remained similar from day 7 to day 14. The terminal elimination half-life of dotarizine and its metabolite appeared to be between 7 and 12 h.  相似文献

We have examined the applicability of this simple and effective test to polymer systems widely used in the medical industry. First, it was found that not every polymer examined exhibited a distinct onset of autocatalytic oxidation detectable by DSC. However, for cases where distinct onsets are found, powerful insights can be obtained. These included activation energy of the degradation, thermal oxidative shelf life prediction, and correlation, with product performances. To eliminate potential "backward" priming effects, S. Glucksberg et al (see record 1986-29080-001) introduced a variant of the cross-modal lexical priming task in which subjects made lexical decisions to nonword targets that were modeled on a word related to either the contextually biased or unbiased sense of an ambiguous word. Lexical decisions to nonwords were longer than controls only when the nonword was related to the contextually biased sense of the ambiguous word, leading Glucksberg et al to conclude that context does constrain lexical access and that the multiple access pattern observed in previous studies was probably an artifact of backward priming. We did not find nonword interference when the nonword targets used by Glucksberg et al were preceded by semantically related ambiguous or unambiguous word primes. However, we did replicate their sentence context results when the ambiguous words were removed from the sentences. We conclude that the interference obtained by Glucksberg et al is due to postlexical judgments of the congruence of the sentence context and the target, not to context constraining lexical access. Based on the concept of entropy of mixing, a new method for determining the degree of mixing of the filler in a polymer matrix has been established. The measurement used in this method is based on a new variable, the effective volume fraction of the filler, ?′, which is a function of the mixing index, A, and the volume fraction of the filler, ?. The mixing index can be determined from the tensile modulus data of filled elastomers. Excellent agreement is obtained between the mixing index and the dispersion of the filler as determined by a morphological study of cryogenically fractured surfaces of filled elastomers. Using the new variable, the effective volume fraction of the filler, useful relationships for the tensile modulus and the ultimate stretch ratio of filled elastomer have been established. Experimental data confirm that these relationships can well describe the effects of concentration and degree of mixing on the two tensile properties of filled elastomers.  相似文献
