BACKGROUND: In Tunisia, sardinelle (Sardinella aurita) catches totalled about 13 300 t in 2002. During processing, solid wastes including heads and viscera are generated, representing about 30% of the original raw material. Viscera, one of the most important by‐products of the fishing industry, are recognised as a potential source of digestive enzymes, especially proteases with high activity over a wide range of pH and temperature conditions. This paper describes the purification procedure and some biochemical characterisation of trypsin from S. aurita viscera. RESULTS: Trypsin from the viscera of sardinelle (S. aurita) was purified by fractionation with ammonium sulphate, Sephadex G‐75 gel filtration, Sepharose mono Q anion exchange chromatography, ultrafiltration and a second Sephadex G‐75 gel filtration, resulting in a 5.42‐fold increase in specific activity and 6.1% recovery. The molecular weight of the purified enzyme was estimated to be 24 kDa using size exclusion chromatography and sodium dodecyl sulfate polyacrylamide gel electrophoresis. The purified enzyme showed esterase‐specific activity on N‐α‐benzoyl‐L ‐arginine ethyl ester (BAEE) that was four times greater than its amidase‐specific activity on N‐α‐benzoyl‐DL ‐arginine‐p‐nitroanilide (BAPNA). The optimal pH and temperature for enzyme activity were pH 8 and 55 °C respectively using BAEE as a substrate. The trypsin kinetic constants Km and kcat on BAPNA were 1.67 mmol L?1 and 3.87 s?1 respectively, while the catalytic efficiency kcat/Km was 2.31 s?1 L mmol?1. CONCLUSION: Trypsin was purified from sardinelle (S. aurita) viscera. Biochemical characterisation of S. aurita trypsin showed that this enzyme can be used as a possible biotechnological tool in the fish‐processing and food industries. Inspired by successful application of evolutionary algorithms to solving difficult optimization problems, we explore in this paper, the applicability of genetic algorithms (GAs) to the cover printing problem, which consists in the grouping of book covers on offset plates in order to minimize the total production cost. We combine GAs with a linear programming solver and we propose some innovative features such as the “unfixed two-point crossover operator” and the “binary stochastic sampling with replacement” for selection. Two approaches are proposed: an adapted genetic algorithm and a multiobjective genetic algorithm using the Pareto fitness genetic algorithm. The resulting solutions are compared. Some computational experiments have also been done to analyze the effects of different genetic operators on both algorithms.  相似文献

In this article we consider the system of generalized thermoelasticity under Green–Lindsay model in one dimension with Dirichlet–Neumann boundary conditions. First, the roots of the characteristic polynomial are investigated by applying an approach based on the implicit function theorem. Then we prove the exponential decay of the associated energy and describe the optimal decay rate. The numerical calculation of the corresponding characteristic roots is done for different real materials. Then another approach based on Hurwitz criterion is applied to obtain the decay rate analytically. Finally, we present a discussion of the decay rate given by both approaches, as well as a comparison with already-existing results for Lord–Shulman and classical models of thermoelasticity.  相似文献

A nonlinear differential equation describing the pressure head grade line along nontapered laterals is derived. For horizontal pipes, the solution of the problem yields the pressure head grade line implicitly in an integral form. The outstanding outcome of this approach is the establishment of an analytical relation between the inlet discharge and the pressure heads at the inlet and at the distal ends of the pipe. The current method is deemed to be very accurate as compared to the stepwise method. It is proven that the discharge within the pipe varies quasilinearly with the distance which corroborates the assumption of constant outflow used in the drop ratio approach. The main feature of this method is that it provides the tools whereby a designer can achieve the crossover from the input to the output data and vice versa.  相似文献

Yoghurt was prepared by adding three concentrations of fennel essential oil (FEO) (2.5, 5 and 7.5 µL) in 100 mL of pasteurised milk. Fennel essential oil did not affect the viability of yoghurt starters during fermentation. Based on sensory analysis, only the 5‐µL FEO concentration was incorporated in order to investigate the physicochemical and microbiological properties of enriched yoghurt during storage. Results showed that FEO in yoghurt did not significantly affect the quality after acidification (> 0.05). However, total solid content, syneresis and lightness (L*) differed significantly (P < 0.05). Bacterial counts indicated that the control yoghurt had the highest values. Fortified yoghurt had an increased shelf life of 29 days.  相似文献

The angiotensin I-converting enzyme (ACE) inhibitory activities of protein hydrolysates prepared from muscle of cuttlefish (Sepia officinalis) by treatment with various digestive proteases were investigated. The most active hydrolysate was obtained with the crude protease extract from the hepatopancreas of cuttlefish (64.47 ± 1.0% at 2 mg of dry weight/ml) with a degree of hydrolysis of 8%. By gel filtration on Sephadex G-25 and RP-HPLC on C18 column, three novel peptides with high ACE-inhibitory activity were purified and their molecular masses and amino acid sequences were determined. The three peptides Val-Tyr-Ala-Pro, Val-Ile-Ile-Phe and Met-Ala-Trp with IC50 values of 6.1, 8.7 and 16.32 μM, respectively, were novel ACE-inhibitory peptides. Lineweaver–Burk plots suggest that the three purified peptides act as non-competitive inhibitors against ACE. These results suggest that some peptides from cuttlefish could be a beneficial ingredient for nutraceuticals against hypertension.  相似文献

BACKGROUND: Veratric acid (VA, 3,4‐dimethoxy‐benzoic acid) is representative of the polyphenolic type compounds present in olive mill wastewater (OMW). Given the bactericide factor, the inhibitor character and the anti bacteriological activity of this compound, traditional biological digestion cannot be applied and therefore new technologies, such as electrochemical oxidation using a boron‐doped diamond (BDD) anode have to be considered to avoid its accumulation in the environment. RESULTS: The electrochemical oxidation of aqueous solutions containing 1 mmol L?1 VA has been investigated using a filter‐press reactor with a BDD anode during galvanostatic electrolysis. The influence of several operating parameters, such as applied current density, temperature, flow‐rate and supporting electrolyte concentration and type has been investigated. The experimental results showed that under the experimental conditions used the oxidation of VA was under mass‐transfer control and VA was completely degraded by the reaction with hydroxyl radicals electrogenerated at the BDD surface. The chemical oxygen demand (COD) decay kinetic followed a pseudo‐first‐order reaction and the apparent rate constant increased with flow rate and temperature. Under optimal experimental conditions of flow‐rate (300 L h?1), temperature (35 °C) and current density (10 mA cm?2), 99.5% of COD was removed during 2 h electrolysis, with 16.4 kWh m?3 energy consumption. CONCLUSIONS: This study suggests that anodic oxidation with a BDD electrode is an excellent method for the treatment of effluents contaminated with VA and related polyphenols. This paper refines and validates the predictions of a simplified analysis method for evaluating the potential of daylighting to save energy use associated with electrical lighting. Specifically, impacts on daylighting performance are investigated for several combinations of building geometry, window size, and glazing type for several US and international locations. The impact of both dimming and stepped daylighting controls and their settings are also investigated. Predictions from the simplified method are validated using measurements obtained from field-testing of a daylighting control system utilized to operate lighting fixture illuminating an office space.  相似文献

Trypsin was purified from the viscera of barbel by precipitation using ammonium sulphate (0-80%), Sephadex G-100, and Mono Q-Sepharose ion exchange chromatography. The trypsin was purified 27-fold, with 79 U/mg specific activity and 31% recovery. The enzyme had a molecular weight of 24 kDa; purified trypsin appeared as a single band on native-PAGE. The optimum pH and temperature for enzyme activity were pH 10.0 and 55 °C with BAPNA used as a substrate. The N-terminal amino acid sequence of the first 12 amino acids of the purified trypsin was IVGGYECTPYSQ. The Michaelis-Menten constant (Km) and catalytic constant (kcat) values of the enzyme were 0.018 mM and 1.21 s−1, respectively. The study also investigated the effects of purified trypsin on the recovery of carotenoproteins from shrimp (Parapenaeus longirostris) shells through hydrolysis using 1.0 U barbel trypsin/g shrimp shells for 1 h at 30 °C. The freeze-dried carotenoproteins recovered contained 71.09% protein, 16.47% lipid, 7.78% ash, and 1.79% chitin.  相似文献

The present study describes the chemical composition, and antimicrobial, antioxidant and angiotensin I-converting enzyme (ACE) inhibitory activities of essential oil from Periploca laevigata root barks (PLRB), an aromatic plant widely distributed in Tunisia and used as a traditional medicinal plant. Gas chromatography/mass spectrometry was used to determine the composition of the PLRB oil. Forty-three components were identified in the essential oil and the main compounds were benzaldehyde (56%), methyl 4-methoxysalicylate (6.55%) and carvacrol (4.75%). The PLRB essential oil exhibited a dose-dependent manner of inhibitory activity toward ACE. The highest ACE inhibitory activity (54%) was observed at a concentration of 30 μg/ml. The PLRB oil was also found to possess antioxidant activities, as evaluated by the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical method, β-carotene bleaching and reducing power assays. The antimicrobial activity of the essential oil was also investigated on several microorganisms. The inhibition zones and minimal inhibitory concentration (MIC) values of bacterial strains were in the range of 12–46 mm and 50–300 μg/ml, respectively. The inhibitory activity of the PLRB essential oil against Gram-positive bacteria was significantly higher than against Gram-negative.
