Towards a continuous microfluidic rheometer   总被引:1,自引:1,他引:0  
In a previous paper we presented a way to measure the rheological properties of complex fluids on a microfluidic chip (Guillot et al., Langmuir 22:6438, 2006). The principle of our method is to use parallel flows between two immiscible fluids as a pressure sensor. In fact, in a such flow, both fluids flow side by side and the size occupied by each fluid stream depends only on both flow rates and on both viscosities. We use this property to measure the viscosity of one fluid knowing the viscosity of the other one, both flow rates and the relative size of both streams in a cross-section. We showed that using a less viscous fluid as a reference fluid allows to define a mean shear rate with a low standard deviation in the other fluid. This method allows us to measure the flow curve of a fluid with less than 250 μL of fluid. In this paper we implement this principle in a fully automated set up which controls the flow rate, analyzes the picture and calculates the mean shear rate and the viscosity of the studied fluid. We present results obtained for Newtonian fluids and complex fluids using this set up and we compare our data with cone and plate rheometer measurements. By adding a mixing stage in the fluidic network we show how this set up can be used to characterize in a continuous way the evolution of the rheological properties as a function of the formulation composition. We illustrate this by measuring the rheological curve of four formulations of polyethylene oxide solution with only 1.3 mL of concentrated polyethylene oxide solution. This method could be very useful in screening processes where the viscosity range and the behavior of the fluid to an applied stress must be evaluated.  相似文献

Free-living amoebae (FLA) are protozoa that can be found in water networks where they prey on bacteria within biofilms. Most bacteria are digested rapidly by phagocytosis, however some are able to survive within amoebae and some are even able to multiply, as it is the case for Legionella pneumophila. These resisting bacteria are a potential health problem as they could also resist to macrophage phagocytosis. Several publications already reported intra-amoebal bacteria but the methods of identification did not allow metagenomic analysis and are partly based on co-culture with one selected amoebal strain. The aim of our study was to conduct a rRNA-targeted metagenomic analysis on amoebae and intra-amoebal bacteria found in drinking water network, to provide the first FLA microbiome in environmental strains. Three sites of a water network were sampled during four months. Culturable FLA were isolated and total DNA was prepared, allowing purification of both amoebal and bacterial DNA. Metagenomic studies were then conducted through 18S or 16S amplicons sequencing. Hartmannella was by far the most represented genus of FLA. Regarding intra-amoebal bacteria, 54 genera were identified, among which 21 were newly described intra-amoebal bacteria, underlying the power of our approach. There were high differences in bacterial diversity between the three sites. Several genera were highly represented and/or found at least in two sites, underlying that these bacteria could be able to multiply within FLA. Our method is therefore useful to identify FLA microbiome and could be applied to other networks to have a more comprehensive view of intra-amoebal diversity.  相似文献

We compare the performance of two back reflector designs on the optoelectrical properties of microcrystalline silicon solar cells. The first one consists of a 5‐µm‐thick low‐pressure chemical vapor deposition (LPCVD)‐ZnO electrode combined with a white sheet; the second one incorporates an Ag reflector deposited on a thin LPCVD‐ZnO layer (with thickness below 200 nm). For this latter design, the optical loss in the nano‐rough Ag reflector can be strongly reduced by smoothing the surface of the thin underlying ZnO layer, by means of an Ar‐plasma treatment. Because of its superior lateral conductivity, the thin‐ZnO/Ag back reflector design provides a higher fill factor than the dielectric back reflector design. When decreasing the roughness of the front electrode with respect to our standard front LPCVD‐ZnO layer, the electrical cell performance is improved; in addition, the implementation of the thin‐ZnO/Ag back reflector leads to a significant relative gain in light trapping. Applying this newly optimized combination of front and back electrodes, the conversion efficiency is improved from 8.9% up to 9.4%, for cells with an active‐layer thickness of only 1.1 µm. Hypobetalipoproteinemia is characterized by LDL-cholesterol and apolipoprotein B (apoB) plasma levels below the fifth percentile for age and sex. Familial hypobetalipoproteinemia (FHBL) is mostly caused by premature termination codons in the APOB gene, a condition associated with fatty liver and steatohepatitis. Nevertheless, many families with a FHBL phenotype carry APOB missense variants of uncertain significance (VUS). We here aimed to develop a proof-of-principle experiment to assess the pathogenicity of VUS using the genome editing of human liver cells. We identified a novel heterozygous APOB-VUS (p.Leu351Arg), in a FHBL family. We generated APOB knock-out (KO) and APOB-p.Leu351Arg knock-in Huh7 cells using CRISPR-Cas9 technology and studied the APOB expression, synthesis and secretion by digital droplet PCR and ELISA quantification. The APOB expression was decreased by 70% in the heterozygous APOB-KO cells and almost abolished in the homozygous-KO cells, with a consistent decrease in apoB production and secretion. The APOB-p.Leu351Arg homozygous cells presented with a 40% decreased APOB expression and undetectable apoB levels in cellular extracts and supernatant. Thus, the p.Leu351Arg affected the apoB secretion, which led us to classify this new variant as likely pathogenic and to set up a hepatic follow-up in this family. Therefore, the functional assessment of APOB-missense variants, using gene-editing technologies, will lead to improvements in the molecular diagnosis of FHBL and the personalized follow-up of these patients.  相似文献

Knowledge of the viscosity of virgin olive oils (VOOs) is of great importance for the design of pilot plants, to determine the time required for the settling of particles at the end of the production chain and from a sensory view point. The dynamic viscosities of French VOOs from four different cultivars (‘Aglandau’, ‘Bouteillan’, ‘Salonenque’ and ‘Tanche’) were studied as a function of their fatty acid and TAG compositions and of the temperature [10–50°C]. These four VOOs had different TAG and fatty acid compositions representative of the range of compositional variations in the main French oils. Their viscosities were similar, although small but measurable differences that depended on their compositions were apparent. All the VOO samples exhibited the same dynamic viscosity pattern over temperature. For a given temperature, the viscosity difference was the greatest between Aglandau and Salonenque oils, Aglandau being the oil with the highest viscosity. The correlation between temperature and viscosity was highlighted by an Arrhenius model for this Newtonian fluid. The Arrhenius activation energy was correlated (R2 = 0.993) with the percentage of triolein, the main TAG in olive oil.  相似文献

This paper derives fundamental performance bounds for statistical estimation of parametric surfaces embedded in R3. Unlike conventional pixel-based image reconstruction approaches, our problem is reconstruction of the shape of binary or homogeneous objects. The fundamental uncertainty of such estimation problems can be represented by global confidenceregions, which facilitate geometric inference and optimization ofthe imaging system. Compared to our previous work on global confidence region analysis for curves [two-dimensional (2-D) shapes], computation of the probability that the entire surface estimate lies within the confidence region is more challenging because a surface estimate is an inhomogeneous random field continuously indexed by a 2-D variable. We derive an asymptotic lower bound to this probability by relating it to the exceedence probability of a higher dimensional Gaussian random field, which can, in turn, be evaluated using the tube formula due to Sun. Simulation results demonstrate the tightness of the resulting bound and the usefulness of the three-dimensional global confidence region approach.  相似文献

The purpose of this study was to assess the radiological findings after surgical removal of juvenile nasopharyngeal angiofibromas (JNA). The postoperative CT and MRI scans of ten patients were reviewed. The cured group included six patients. The non-controlled group included six patients with eight recurrences. Two patients belonged to both groups as they were also followed and cured after surgery for relapse. Four recurrences were asymptomatic and diagnosed by imaging. The imaging patterns were matched to the patients clinical status and endoscopic findings. In the cured group, non-enhanced residual soft tissue masses were seen in all cases. In the non-controlled group, recurrence was always demonstrated on early postoperative CT or MR as a dramatically enhanced mass. The recurrence was located in the lateral or superior aspect of the nasopharynx (n = 3), deep to the fossa of Rosenmuller (n = 4) or out of the nasopharynx (n = 1). In two cases a remaining enhanced mass disappeared spontaneously on iterated examinations. Because of numerous asymptomatic relapses, a radiological workup is recommended four months after surgery, even in patients with normal endoscopy, to rule out posterolateral or extranasopharyngeal recurrences. Spontaneous evolution of residual masses must be appreciated on iterated imaging examinations.  相似文献

Ratchetting or progressive distortion is a great concern in the field of structural analysis of nuclear components. This damage is characterized by incremental distorsion caused by cyclic loading. Generally ratchetting is the consequence of the application of primary loading (due to pressure or dead weight) and cyclic straining (like thermal straining caused by thermal transient). Usually cyclic straining is quoted by the range of secondary stress ΔQ (for instance range of thermal stress), and load controlled stress are called primary stress P.Conventional method of analysis of ratchetting are based on over simplified assumption about material behaviour. A critical review of this situation is made and it appears that real material behavior is the most important factor of analysis validity. Therefore a large experimental study is used in order to obtain sound basis for a method of ratchetting analysis. This method is fully given in this paper. It is based on the effective primary stress concept.Additional indications are given on progressive buckling and creep rupture enhanced by ratchetting.  相似文献
