A novel approach using a combination of capillary electrophoresis/mass spectrometry (CE/MS) and off-line Fourier transform ion cyclotron resonance mass spectrometry (FTICR-MS) revealed the structural details of acidic constituents of atmospheric organic aerosol. Both techniques utilized electrospray ionization (ESI), a soft ionization method, to facilitate the analysis of complex mixtures of organic compounds. CE/ESI-MS using an UltraTrol LN-precoated capillary and acidic background electrolytes at different pH values (2.5 and 4.7) was used to differentiate between weak (carboxylic) and strong (sulfonic) organic acids. On the basis of the electrophoretic mobility, m/z constraints from CE/ESI(-)-MS, and elemental composition information retrieved from off-line FTICR-MS, a variety of aliphatic and aromatic carboxylic acids (CHO-bearing molecules), nitrogen-containing carboxylic acids (CHON-bearing molecules), organosulfates (CHOS-bearing molecules), and (nitrooxy)organosulfates (CHONS-bearing molecules) were tentatively identified in the Oasis-HLB-extracted urban PM(2.5) (particulate matter with an aerodynamic diameter of <2.5 μm). The chemical known/unknown structures of detected compounds were confirmed by the semiempirical Offord model (effective mobility linearly correlated to Z/M(2/3)). Superantigens bind to MHC class II-positive cells and stimulate T lymphocytes expressing specific V beta regions of the TCR. Two distinct regions of staphylococcal enterotoxin A superantigen (SEA) have been shown to affect the binding to MHC class II molecules. Results presented here demonstrate for the first time that the SEA-DR interaction can be affected by mutations on the class II alpha-chain. Furthermore, we have precisely mapped the interaction of the SEA N-terminal domain with the alpha1 domain of HLA-DR. Scatchard analysis using DAP cells transfected with mutant class II molecules showed a role for residue DR alpha K39 in the binding of SEA. Also, complementation experiments using mutant SEA molecules revealed an interaction between SEA residue F47 and position alphaQ18 on an outer loop of HLA-DR. These interactions between SEAF47 and the DR alpha-chain are critical, as they allow the recognition by an otherwise nonreactive V beta1+ T cell hybridoma and induction of tyrosine phosphorylation through the TCR. 相似文献

The stable crack growth through three-point bend (TPB) and stiffened and unstiffened compact tension (CT) specimens of D16AT aluminium alloy has been studied both theoretically and experimentally. The specimen thickness is 8 mm. The variation of load with crack opening displacement, the extent of stable crack growth, the cumulative plastically deformed zone and crack edge profiles have been obtained experimentally. These are also predicted theoretically under the assumption of either a state of plane stress or plane strain using a finite element scheme and the COA criterion. Generally, the experimental results agree well with the predictions based on the plane stress condition. There appears to be no significant variation in size of the experimental cumulative plastic zone across the specimen thickness, thereby indicating that the constraint on the plastic zone does not develop near the mid-thickness region. 相似文献

Polyethylene (PE), polypropylene (PP) and their blends have attracted a lot of attention due to their potential industrial applications. Therefore, the current work has been carried out with the main objective of investigating the impact of the thermal aging/treatment and blend ratio (composition range) on the mechanical (tensile and hardness) and thermal characteristics (using thermogravimetric analysis in a dynamic air atmosphere) of PE, PP and PE/PP binary blends. Samples of PE/PP blends containing 100/00, 75/25, 50/50, 25/75 and 0/100 wt.% were prepared via injection moulding technique and thermally treated/aged at 100 °C for 0, 2, 4, 7, 14 days. The tensile measurements indicated that the yield strength and the modulus decrease with increasing PE content. It was also observed that PE, PP and their blends deform in ductile modes. They undergo a uniform yielding over a wide range of deformation, which is followed by strain hardening and then failure. The strain to break for pure PE is found to be much higher than that for pure PP and for their blends, intermediate values have been observed. The hardness measurements have also revealed that increasing PE content in PE/PP blends reduced the hardness value of PP, however, thermal aging at 100 °C has not affected the polymers hardness which holds also true for the tensile properties, showing a good correlation between tested mechanical properties. The thermogravimetric analysis (TGA) in a dynamic air atmosphere and derivative thermogravimetric analysis (DTA) were conducted to study the thermal degradation and stability of thermally unaged and aged PE, PP and PE/PP blends in terms of the initial (Td and Td(1%)) and final (Td(99%)) decomposition temperatures and maximum decomposition rate temperature (Tmax). All polymers start to decompose at no less than 365 °C. As for mechanical properties, the blend ratio has affected the thermal properties however, aging time has not. 相似文献

Abstract Mode II stable crack extension has been examined for an aircraft grade aluminium alloy D16AT. Both theoretical and experimental results are presented. The experimental observations include load displacement diagrams, plastic wake, crack front tunnelling and scanning electron micrographs of the fracture surfaces. The crack shows a tendency for in-plane extension, and the fracture surface is very flat, smooth and free of any dimples. The crack front advances with neghgible tunnelling at all stages of extension. The span of mode II stable crack growth (SCG) is longer than in the case of mode I SCG reported earlier for the same material and there is also more extensive plastic deformation. In the presence of a slight mode I load, the crack grows out-of-plane and the fractured surface facets resemble that of a mode I or mixed-mode dimpled fracture. The theoretical study is based on a finite element analysis using small deformation theory and incremental plasticity. Some of the experimental results have been theoretically predicted using the COA criterion as the governing criterion. The theoretical results include load-displacement diagrams, crack edge displacement curves, plastic zones and the J resistance curves. There is good agreement between the load-displacement diagrams. The initiation and maximum loads differ by less than 15%. The J resistance curve has a constant slope over the whole span of stable crack growth. 相似文献

Driving blood glycaemia from hyperglycaemia to euglycaemia as fast as possible while avoiding hypoglycaemia is a major problem for decades for type‐1 diabetes and is solved in this study. A control algorithm is designed that guaranties hypoglycaemia avoidance for the first time both from the theory of positive systems point of view and from the most pragmatic clinical practice. The solution consists of a state feedback control law that computes the required hyperglycaemia correction bolus in real‐time to safely steer glycaemia to the target. A rigorous proof is given that shows that the control‐law respects the positivity of the control and of the glucose concentration error: as a result, no hypoglycaemic episode occurs. The so‐called hypo‐free strategy control is tested with all the UVA/Padova T1DM simulator patients (i.e. ten adults, ten adolescents, and ten children) during a fasting‐night scenario and in a hybrid closed‐loop scenario including three meals. The influence of antihypertensive treatment on the long-term evolution of arterial pressure and renal function was studied in a prospective controlled trial conducted in renal transplant recipients treated by cyclosporine. Within six months after transplantation, patients were randomly allocated to treatment by the angiotensin-converting enzyme inhibitor, lisinopril (ACEI, alone or associated with frusemide; N = 14), or the calcium antagonist, nifedipine (CA, alone or associated with atenolol; N = 11). Glomerular filtration rate (TcDTPA clearance) and effective renal plasma flow (hippuran clearance) as well as 24-hour urinary excretion of electrolytes and albumin were estimated at about 1 and 2.5 years of follow-up. Before initiation of antihypertensive therapy, the two groups were similar with regards to mean arterial pressure (119 +/- 2 vs. 120 +/- 4 mm Hg), effective renal plasma flow (285 +/- 26 vs. 248 +/- 33 ml/min/1.73 m2) and glomerular filtration rate (59 +/- 4 vs. 61 +/- 8 ml/min/1.73 m2 in the ACEI and CA groups, respectively). Both ACEI and CA treatments were associated with no change in renal function, a similar change in mean arterial pressure (ACEI -18 +/- 3; CA -13 +/- 5 mm Hg) and identical trough blood levels of cyclosporine. Urinary albumin excretion did not change significantly in any groups. Of interest, only in the ACEI group did filtration fraction significantly decrease (from 0.22 +/- 0.01% to 0.19 +/- 0.01% at final studies). These results indicate that in cyclosporine-treated transplant recipients, a satisfactory control of hypertension is obtained by chronic ACEI, which is as effective on arterial pressure as a combination of CA and atenolol.(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献
