Qualitative and quantitative PCR assays were developed for detection of commercialised Bt cotton events, i.e. MON531, MON15985 and other Bt crops, which are under different stages of field trials in India, i.e. Bt brinjal, Bt rice, Bt cauliflower, Bt potato and Bt okra. Multiplex PCR assays simultaneously detecting specific cry1Ac, cry1Ab, cry2Ab genes, Cauliflower Mosaic Virus 35S promoter, nptII marker gene along with species- or taxon-specific endogenous gene in these Bt crops have been developed. The quantitative real-time PCR assays were also reported for cry1Ac gene using designed primers and TaqMan probe. The sensitivity of developed assays for detection of specific transgene was established up to 0.01%. The analytical methods developed in the present report will be of immense use for qualitative screening and detection of Bt crops along with the quantitative analysis of inserted cry1Ac gene to meet the threshold level for regulatory compliance.  相似文献

DNA repair proteins participate in extensive protein−protein interactions that promote the formation of DNA repair complexes. To understand how complex formation affects protein function during base excision repair, we used SpyCatcher/SpyTag ligation to produce a covalent complex between human uracil DNA glycosylase (UNG2) and replication protein A (RPA). Our covalent “RPA−Spy−UNG2” complex could identify and excise uracil bases in duplex areas next to ssDNA−dsDNA junctions slightly faster than the wild-type proteins, but this was highly dependent on DNA structure, as the turnover of the RPA−Spy−UNG2 complex slowed at DNA junctions where RPA tightly engaged long ssDNA sections. Conversely, the enzymes preferred uracil sites in ssDNA where RPA strongly enhanced uracil excision by UNG2 regardless of ssDNA length. Finally, RPA was found to promote UNG2 excision of two uracil sites positioned across a ssDNA−dsDNA junction, and dissociation of UNG2 from RPA enhanced this process. Our approach of ligating together RPA and UNG2 to reveal how complex formation affects enzyme function could be applied to examine other assemblies of DNA repair proteins.  相似文献

The understanding of the flow characteristics and effect of gas-solid interactions in pneumatic risers is fundamental to investigate to ensure effective design cost-effective operation. Thus, to understand the effect of gas-solid interactions on the hydrodynamics of newly proposed conversing risers, this study mainly focused on predicting pressure drop in the dilute phase pneumatic conveying system. The experiments were conducted in a converging riser having a convergence angle of 0.2693°. Various solid particles such as sago, black mustard, and alumina have been considered to study the effect of particle sizes and density on the pressure drop. The experimental outcomes indicate that the total pressure drop increases with an increase in the solid density and gas mass flow rate. Moreover, smaller particle sizes are also increased the pressure drop. An empirical correlation is developed for the prediction of total pressure drop ΔPT in converging pneumatic riser via dimensional analysis. All dependent variables such as particle and air density, drag force, acceleration due to gravity, the mass flow rate of air and particle, the diameter of particle and converging riser, the height of converging riser were considered to develop the empirical correlation. The established relationship is tested, and experimental data have been fitted for its validation. The estimated relative error of less than 0.05 proved the significance of the developed correlation. Hence, it can be stated that the established relationship is useful in studying the effects of various parameters on the pressure drop across the length of the conversing riser.  相似文献

The aim of this investigation was to design iron oxide containing nanocomposites which could display superparamagnetic behavior and thus find application in biomedical and allied fields. To achieve the proposed objectives methyl methacrylate was polymerized by a redox system comprising of metabisulphite and persulphate in the immediate presence of a crosslinker, N,N′‐methylene bis acrylamide and a preformed polymer, i.e., polyvinyl alcohol. Into the prepared polymer matrix nanosized magnetite (Fe3O4) particles were evenly dispersed by in situ precipitation of Fe2+/Fe3+ ions. The nanocomposite materials were characterized by techniques like FTIR, SEM, TEM, XRD, and DSC. The magnetic behavior of nanocomposites and bulk magnetite particles was studied under varying applied magnetic fields and their superparamagnetic property was examined. A viral infection involves entry and replication of viral nucleic acid in a host organism, subsequently leading to biochemical and structural alterations in the host cell. In the case of SARS-CoV-2 viral infection, over-activation of the host immune system may lead to lung damage. Albeit the regeneration and fibrotic repair processes being the two protective host responses, prolonged injury may lead to excessive fibrosis, a pathological state that can result in lung collapse. In this review, we discuss regeneration and fibrosis processes in response to SARS-CoV-2 and provide our viewpoint on the triggering of alveolar regeneration in coronavirus disease 2019 (COVID-19) patients.  相似文献

Galectins are a class of carbohydrate‐binding proteins named for their galactose‐binding preference and are involved in a host of processes ranging from homeostasis of organisms to immune responses. As a first step towards correlating the carbohydrate‐binding preferences of the different galectins with their biological functions, we determined carbohydrate recognition fine‐specificities of galectins with the aid of carbohydrate microarrays. A focused set of oligosaccharides considered relevant to galectins was prepared by chemical synthesis. Structure–activity relationships for galectin–sugar interactions were determined, and these helped in the establishment of redundant and specific galectin actions by comparison of binding preferences. Distinct glycosylations on the basic lactosyl motifs proved to be key to galectin binding regulation—and therefore galectin action—as either high‐affinity ligands are produced or binding is blocked. High‐affinity ligands such as the blood group antigens that presumably mediate particular functions were identified.  相似文献
