The Unc119 protein mediates transport of myristoylated proteins to the photoreceptor outer segment, a specialized primary cilium. This transport activity is regulated by the GTPase Arl3 as well as by Arl13b and Rp2 that control Arl3 activation/inactivation. Interestingly, Unc119 is also enriched in photoreceptor synapses and can bind to RIBEYE, the main component of synaptic ribbons. In the present study, we analyzed whether the known regulatory proteins, that control the Unc119-dependent myristoylated protein transport at the primary cilium, are also present at the photoreceptor synaptic ribbon complex by using high-resolution immunofluorescence and immunogold electron microscopy. We found Arl3 and Arl13b to be enriched at the synaptic ribbon whereas Rp2 was predominantly found on vesicles distributed within the entire terminal. These findings indicate that the synaptic ribbon could be involved in the discharge of Unc119-bound lipid-modified proteins. In agreement with this hypothesis, we found Nphp3 (Nephrocystin-3), a myristoylated, Unc119-dependent cargo protein enriched at the basal portion of the ribbon in close vicinity to the active zone. Mutations in Nphp3 are known to be associated with Senior–Løken Syndrome 3 (SLS3). Visual impairment and blindness in SLS3 might thus not only result from ciliary dysfunctions but also from malfunctions of the photoreceptor synapse.  相似文献

Coke microtexture and microstructure is a function of feed coal composition and carbonisation process. If the process remains constant, the composition plays a vital role in developing the microtexture and microstructure which also reflects the coke quality. Knowledge of the influence of coal composition and characteristics on coke quality is a must for formulating a coal blend for coke making. To better understand how the coal transforms into coke requires the ability to correlate the microtextural and microstructural features to coal properties. Petrography is an important tool for comparing the feed coal and corresponding coke. In the present study, cokes manufactured from single coals and two-component blends under similar coking conditions were studied under the microscope to provide valuable information to coke makers. Not only the rank and quantification of macerals but also the association of macerals and minerals play an important role. Also the size and type of inertinite affect the coke quality. Micro-cracking, crack lengths and fissures identify the zone of weakness, whereas microtexture of cell walls suggests the strength-controlling parameters of coke. A new beverage product was developed utilising two protein‐rich oilseed sources, namely peanut and soy. Medium‐roasted peanut flour and chocolate flavour were incorporated to offer pleasant flavour profile. The peanut–soy combination would also improve essential amino acid profile, especially that of lysine, compared with an all‐peanut product. A pilot‐plant scale beverage‐processing protocol involved filtration, homogenisation and pasteurisation as the major operating steps. Beverage formulation employed a three‐component constrained mixture design. The low‐ and high‐bound constraints were determined for peanut (30.6–58.7%), soy (28.3–43.5%) and chocolate syrup (13.0–25.9%) based on lysine content, viscosity and visual stability index values of 51‐mg g?1 protein, 36.9 mPa s and 1.00, respectively. The beverage formulation and processing protocol thus developed were the basis for further study on consumer acceptability of the new chocolate‐flavoured peanut–soy beverage.  相似文献

Retention of a firm, crisp fruit texture is a major consideration for pickled vegetables including pickles made from fermented cucumbers. It is known that cucumbers soften rapidly when fermented at low salt concentrations (<0.5 M) without added calcium. This study has shown that there is non-enzymatic softening in low salt fermentations because cucumbers soften even when heated sufficiently to inactivate pectinesterase and several glycosidases that can hydrolyse glycosidic linkages that are present in cell wall polysaccharides. Though pectinesterase activity declines and these glycosidases lose activity within the first week of fermentation there is generally greater loss of cucumber tissue firmness when enzymes are not inactivated by heat. While heating cucumbers prior to fermentation reduces softening during subsequent storage, a heat treatment after 2 weeks of fermentation does not reduce softening. This result suggested that the enzymatic reactions responsible for softening occur early in the fermentation process even though the softening does not become evident until later in the storage period. Despite the evidence of an enzymatic component of tissue softening in low salt cucumbers, softening could not be associated with specific enzymes.  相似文献

Networking cognitive radios and nodes from primary system (PS) results in a heterogeneous coexisting multi‐radio wireless network, so that significant network throughput gain can be achieved. However, by investigating cognitive radio network (CRN) architecture, the links in CRNs are unlikely to support complete security check due to link dynamics, opportunistic availability, and uni‐directional in available time window. We therefore introduce trusted cognitive radio networking (TCRN) concept to facilitate network functions such as association in dynamic spectrum access and routing. First of all, we explore the mathematical framework for trust in CRNs. We then show successful association of node to CRN based on the mathematical structure of trust from statistical decision theory. Furthermore, we modify the machine‐learning algorithm to update the trust measure for each node, and develop rules of thumbs to facilitate TCRN with learning capability, based on numerical simulations. The article describes the preparation of cast copolymer sheets of methyl methacrylate with varying mole fractions of N‐(p‐methoxyphenyl) itaconimide/N‐(2‐methoxy‐5‐chlorophenyl) itaconimide/N‐(3‐methoxy‐5‐chlorophenyl) itaconimide monomers by bulk copolymerization using azobisisobutyronitrile as an initiator. The effect of incorporation of varying mole fractions of N‐arylsubstituted itaconimides in poly(methyl methacrylate) (PMMA) backbone on the thermal, optical and physicomechanical properties of cast acrylic sheets were evaluated. The glass transition temperature and the thermal stability increased with increasing amounts of itaconimides in the polymer backbone. An increase in tensile strength, flexural strength, and storage modulus was also observed. The impact strength decreased marginally upon incorporation of imides into the polymer backbone. A slight decrease in the transparency and a significant increase (4–50%) in the haze was observed.
