As part of a North-Rhine-Westphalian state project the district of Unna developed a vaccination programme for continuation schools in 1996. The reason for this programme realized for the first time in a vocational school, was a considerable lack of protection by vaccination in ninth year classes. The present results from the first vocational school show a significant lack of standard vaccination as well as boosters. To improve the current situation it will be necessary to authorize and enable the Public Health Services to carry out even more standard and indication vaccinations at secondary and vocational schools as well as at institutions for the handicapped.  相似文献

In the policy literature, crises are often presented as opportunities for reform initiation. Little is known, however, about the effects that crises may have on the implementation of such reform initiatives. This article explores how a crisis may affect the administrative relations between central authorities and field administrations. The crisis impact on central-field relations appears to be twofold. First, a crisis may facilitate, if not induce, the centralization of decision-making processes. Secondly, a crisis may affect the degree of consensus between central authorities and field administrations with regard to problem definitions and preferred solutions. This paper uses a case study of the Dutch prison system to explore the crisis impact on these two dimensions of central-field relations. In this particular case, crisis circumstances did not facilitate a successful implementation of a major policy overhaul. We suggest that crises may offer opportunities for reform implementation, but only under highly restricted conditions. When these conditions are not met, crises may have perverse effects on central-field relations which limit the prospects for the effective implementation of reform.  相似文献

The thermal conductivity and the particle-to-fluid heat-transfer coefficient for peanut-shaped polyester particles in a packed bed were determined simultaneously using the pseudo-random binary noise sequence (PRBNS) response technique. The modified Dispersion-Concentric (D-C) model was applied to beds packed with the non-spherical particles. A time domain method, using the convolution integral, was developed for removing the effects of the bed ends and the transducer time lag on the experimental responses. Parameter estimation was performed in the time domain, using a simplex method to fit the thermal response calculated from the parametric D-C model to the experimental response curve. The particle-to-fluid heat transfer coefficient in the bed was determined to be 106 W/m2. K at an interstitial fluid velocity of 2.0 m/s. The thermal conductivity of polyester was 0.22 W/m. Recently a number of crystal structures were determined using electron diffraction data with an almost parallel electron beam. In many cases no energy filtering was applied. On the other hand, the contrast in convergent beam electron diffraction patterns is greatly improved by energy filtering of the electrons. To investigate whether energy filtering will improve the accuracy of the structure analysis from diffraction data recorded under an almost parallel beam condition, we recorded diffraction patterns of the [100] zone of YBa(2)Cu(3)O(7) using unfiltered electrons, zero-loss electrons and plasmon-loss electrons, respectively. Subsequently, the structure is refined based on these different electron diffraction datasets, using the program MSLS (Acta Crystallogr. A 54 (1998) 91). The results show that the obtained atomic positions are not significantly different for the chosen filter conditions. Even with amorphous carbon deposited on the specimen, which will cause a significant increase (>40 times) of energy-loss electrons, the structure refinement led to the same atomic positions.  相似文献

Flexible and stretchable electronic skins capable of replicating the human sense of touch are a subject of active research. One of the most popular materials for force sensors in skins is carbon black (CB)/polydimethylsiloxane composite. To aid in skin design, a characterization of this composite is presented here. The sensitivity of composite resistance to uniaxial tension, compression, and shear for each CB concentration is measured and found to be similar for tension and compression, but smaller for shear, with resistance monotonically increasing with strain. In addition, under tension and compression the resistance of the material is measured both in line with and perpendicular to the axis of applied strain, and the response is found to be approximately equal in both cases. The electrical and mechanical relaxation time of the material is also measured and modeled for tension, compression, and shear. The mechanical relaxation time is found to be shorter than the electrical, with both increasing with CB concentration. However, the shortest mechanical relaxation time, 200 s, precludes a sensor with human‐like response times without an active modeling and compensation system. The present study investigated the possibility of forming a multidimensional scale for the measurement of nicotine dependence among adolescents, based on the modified Fagerstr?m Tolerance Questionnaire (mFTQ) and the Hooked on Nicotine Checklist (HONC). A survey was conducted among 33 Dutch secondary schools, resulting in 2,041 smokers who completed the questionnaire. Motivation to quit and number of quit attempts were assessed and used as convergent construct variables for the construct of nicotine dependence. The findings show that combining the items of the mFTQ and the HONC results in three distinct dimensions: behavioral aspects of nicotine dependence, craving, and nervousness during abstinence. We examined this new multidimensional model in a second sample using confirmatory factor analysis. The new multidimensional measure fitted the data satisfactorily and showed good psychometric properties. Results of this study support the notion that nicotine dependence among adolescents is multidimensional.  相似文献

A simplified procedure for cell‐free biocatalytic reductions of conjugated CC double bonds using old yellow enzymes (OYEs) is reported. Instead of indirectly regenerating YqjM (an OYE homologue from B. subtilis) or NemA (N‐ethylmaleimide reductase from E. coli) via regeneration of reduced nicotinamide cofactors, we demonstrate that direct regeneration of catalytically active reduced flavins is an efficient and convenient approach. Reducing equivalents are provided from simple sacrificial electron donors such as ethylenediaminetetraacetate (EDTA), formate, or phosphite via photocatalytic oxidation. This novel photoenzymatic reaction scheme was characterized. Up to 65% rates of the NADH‐driven reaction were obtained while preserving enantioselectivity. The chemoselectivity of the novel approach was exclusive. Even when using crude cell extracts as biocatalyst preparations, only CC bond reduction was observed while ketone and aldehyde groups remained unaltered. Overall, a simple and practical approach for photobiocatalytic reductions is presented.  相似文献

beta-Lactam acylases are crucial for the synthesis of semisynthetic cephalosporins and penicillins. Unfortunately, there are no cephalosporin acylases known that can efficiently hydrolyse the amino-adipic side chain of Cephalosporin C. In a previous directed evolution experiment, residue Asn266 of the glutaryl acylase from Pseudomonas SY-77 was identified as being important for substrate specificity. In order to explore the function of this residue in substrate specificity, we performed a complete mutational analysis of position 266. Codons for all amino acids were introduced in the gene, 16 proteins that could be functionally expressed in Escherichia coli were purified to homogeneity and their catalytic parameters were determined. The mutant enzymes displayed a broad spectrum of affinities and activities, pointing to the flexibility of the enzyme at this position. Mutants in which Asn266 was changed into Phe, Gln, Trp and Tyr displayed up to twofold better catalytic efficiency (k(cat)/K(m))than the wild-type enzyme when adipyl-7-aminodesacetoxycephalosporanic acid (adipyl-7-ADCA) was used as substrate, due to a decreased K(m). Only mutants SY-77(N266H) and SY-77(N266M) showed an improvement of both catalytic parameters, resulting in 10- and 15-times higher catalytic efficiency with adipyl-7-ADCA, respectively. Remarkably, the catalytic activity (k(cat)) of SY-77(N266M) when using adipyl-7-ADCA as substrate was as high as when glutaryl-7-aminocephalosporanic acid (glutaryl-7-ACA) was used, and approaches commercially interesting activity. SY-77(N266Q), SY-77(N266H) and SY-77(N266M) mutants showed a modest improvement in hydrolysing Cephalosporin C. Since these mutants also have a good catalytic efficiency when adipyl-7-ADCA is used and are still active towards glutaryl-7-ACA, they can be regarded as broad substrate acylases. These results demonstrate that the combination of directed evolution for the identification of important positions, together with saturation mutagenesis for finding the optimal amino acid, is a very effective method for finding improved biocatalysts.  相似文献

This paper presents a hysteretic on–off control scheme with optimization algorithms for temperature regulation in refrigerated transport systems. A nonlinear dynamic refrigerated transport system model, which consists of a refrigeration unit and a cargo space, is developed. The model serves as an analytical tool for control design. An optimal on–off control strategy is developed based on the time domain analysis of the temperature oscillations. A cost function involving temperature variations from the set-point, energy consumption and average compressor on–off cycling frequency is introduced for minimization. Choices of weighting values in the cost function give the flexibility of the control approach to meet different requirements. Simulation examples are presented that demonstrate the ability and robustness of the proposed method in achieving tighter temperature regulation and higher system efficiency control in transport refrigeration systems. The simulations are augmented by experimental validation via a novel hardware-in-the-loop load emulation approach.  相似文献
