Organelle proteomics describes the study of proteins present in organelle at a particular instance during the whole period of their life cycle in a cell. Organelles are specialized membrane bound structures within a cell that function by interacting with cytosolic and luminal soluble proteins making the protein composition of each organelle dynamic. Depending on organism, the total number of organelles within a cell varies, indicating their evolution with respect to protein number and function. For example, one of the striking differences between plant and animal cells is the plastids in plants. Organelles have their own proteins, and few organelles like mitochondria and chloroplast have their own genome to synthesize proteins for specific function and also require nuclear‐encoded proteins. Enormous work has been performed on animal organelle proteomics. However, plant organelle proteomics has seen limited work mainly due to: (i) inter‐plant and inter‐tissue complexity, (ii) difficulties in isolation of subcellular compartments, and (iii) their enrichment and purity. Despite these concerns, the field of organelle proteomics is growing in plants, such as Arabidopsis, rice and maize. The available data are beginning to help better understand organelles and their distinct and/or overlapping functions in different plant tissues, organs or cell types, and more importantly, how protein components of organelles behave during development and with surrounding environments. Studies on organelles have provided a few good reviews, but none of them are comprehensive. Here, we present a comprehensive review on plant organelle proteomics starting from the significance of organelle in cells, to organelle isolation, to protein identification and to biology and beyond. To put together such a systematic, in‐depth review and to translate acquired knowledge in a proper and adequate form, we join minds to provide discussion and viewpoints on the collaborative nature of organelles in cell, their proper function and evolution. © 2010 Wiley Periodicals, Inc., Mass Spec Rev 30:772–853, 2011  相似文献

Exposure to trihalomethanes (THMs) through inhalation and dermal contact during showering and bathing may pose risks to human health. During showering and bathing, warm water (35 °C-45 °C) is generally used. Warming of chlorinated supply water may increase THMs formation through enhanced reactions between organics and residual chlorine. Exposure assessment using THMs concentrations in cold water may under-predict the possible risks to human health. In this study, THMs concentrations in warm water were estimated by developing a THMs formation rate model. Using THMs in warm water, cancer and non-cancer risks to human health were predicted for three major cities in Ontario (Canada). The parameters for risk assessments were characterized by statistical distributions. The total cancer risks from exposure to THMs during showering were predicted to be 7.6 × 10− 6, 6.3 × 10− 6 and 4.3 × 10− 6 for Ottawa, Hamilton and Toronto respectively. The cancer risks exceedance probabilities were estimated to be highest in Ottawa at different risk levels. The risks through inhalation exposure were found to be comparable (2.1 × 10− 6-3.7 × 10− 6) to those of the dermal contact (2.2 × 10− 6-3.9 × 10− 6) for the cities. This study predicted 36 cancer incidents from exposure to THMs during showering for these three cities, while Toronto contributed the highest number of possible cancer incidents (22), followed by Ottawa (10) and Hamilton (4). The sensitivity analyses showed that health risks could be controlled by varying shower stall volume and/or shower duration following the power law relationship.  相似文献

The aim of this study is to examine the reasons why firms use ICTs at varying rates, by making a distinction between the two stages of their diffusion: their adoption and the intensity of their use. What are the differences between Internet adopters (in terms of their internal organization and external environment) that explain the intensity with which they use this technology? Furthermore, do these processes vary according to the type of area in which firms are located (urban vs. rural)? A model of technological diffusion is constructed that merges two types of models: those that concentrate on epidemic effects and the so-called equilibrium models that consider the decision to adopt new technologies as a result of an economic calculation by firms. To test this model, we use data drawn from a recent French national survey of 5,200 industrial firms (“ICT and E-commerce”, 2002). One striking result of this study is that we do not obtain a significant positive correlation between firm’s size and Internet intensity of use. Moreover, though spatial disparities related to ICT adoption are no longer significant in France, they remain very important in the processes of ICT appropriation and use by firms. The results also indicate that the determinants of the intensity of Internet use vary significantly according to the firms’ location: epidemics effects play an essential role in the case of urban the firms, while rank effects are essential in low density areas.  相似文献

Energy-storing prosthetic feet are designed to store energy during mid-stance motion and to recover it during late-stance motion. Gait analysis is the most commonly used method to characterize prosthetic foot behaviour during walking. In using this method, however, the foot is generally modelled as a rigid body. Therefore, it does not take into account the ability of the foot to deform. However, the way this deformation occurs is a key parameter of various foot properties under gait conditions. The purpose of this study is to combine finite element modelling and gait analysis in order to calculate the strain, stress and energy stored in the foot along the stance phase for self-selected and fast walking speeds. A finite element model, validated using mechanical testing, is used with boundary conditions collected experimentally from the gait analysis of a single transtibial amputee. The stress, strain and energy stored in the foot are assessed throughout the stance phase for two walking speed conditions: a self-selected walking speed (SSWS), and a fast walking speed (FWS). The first maximum in the strain energy occurs during heel loading and reaches 3J for SSWS and 7J for FWS at the end of the first double support phase. The second maximum appears at the end of the single support phase, reaching 15J for SSWS and 18J for FWS. Finite element modelling combined with gait analysis allows the calculation of parameters that are not obtainable using gait analysis alone. This modelling can be used in the process of prosthetic feet design to assess the behaviour of a prosthetic foot under specific gait conditions.  相似文献

International organisations, such as International Atomic Energy Agency, United Nations Scientific Committee on the Effects of Atomic Radiation and World Health Organisation, together with committees of experts such as Biological Effects of Ionising Radiation and Committee on Medical Aspects of Radiation in the Environment, have assessed the effects of radiation on large exposed populations (Chernobyl accident, and Hiroshima/Nagasaki atomic bombings) and on nuclear energy workers and people living near nuclear facilities. Childhood and in utero exposure to moderate and high levels of ionizing radiation, such as those experienced during the atomic bombings of Japan, or from radiotherapy, is an established cause of leukaemia and solid cancer. There is no evidence of increase in solid cancers (excluding thyroid cancer) or leukaemia in the children from Chernobyl, and no evident link between worker's exposure to radiation and leukaemia in their offspring or with the presence of leukaemia clusters around nuclear power plants. It has also not been possible to demonstrate the evidence of radiation hereditary effects in human populations. In accordance with international guidance, Canadian Nuclear Safety Commission recommends optimisation of protection strategies to reduce doses to children. The development of credible radiological/nuclear event scenarios would assist in identifying probable sources of radioactivity and pathways of exposure for children. Such scenarios should then be used to identify protection strategies appropriate for children.  相似文献

We are developing an instrument based on Kinetic Inductance Detectors (KID) known as the Néel IRAM KID Array (NIKA). Leveraging the experience gained from the first generation NIKA in 2009, an improved, dual-band (150?GHz and 240?GHz) instrument has been designed and tested at the Institut of RadioAstronomie Millimetrique (IRAM) 30-meter telescope in October 2010. The performances, in terms of sensitivity on-the-sky at 150?GHz, are already comparable to existing state-of-the-art bolometer-based instruments. NIKA represents thus the first real proof that KID are a viable technology for ground-based Astronomy. We will describe the instrument, the most recent results and the future plans for building a large resident mm-wave camera.  相似文献

The use of biological-probe-modified solid-state pores in biosensing is currently hindered by difficulties in pore-wall functionalization. The surface to be functionalized is small and difficult to target and is usually chemically similar to the bulk membrane. Herein, we demonstrate the contactless electrofunctionalization (CLEF) approach and its mechanism. This technique enables the one-step local functionalization of the single pore wall fabricated in a silica-covered silicon membrane. CLEF is induced by polarization of the pore membrane in an electric field and requires a sandwich-like composition and a conducting or semiconducting core for the pore membrane. The defects in the silica layer of the micropore wall enable the creation of an electric pathway through the silica layer, which allows electrochemical reactions to take place locally on the pore wall. The pore diameter is not a limiting factor for local wall modification using CLEF. Nanopores with a diameter of 200 nm fabricated in a silicon membrane and covered with native silica layer have been successfully functionalized with this method, and localized pore-wall modification was obtained. Furthermore, through proof-of-concept experiments using ODN-modified nanopores, we show that functionalized nanopores are suitable for translocation-based biosensing.  相似文献

HIPERLAN will provide a 23.5 Mbps data rate transmission to radio channel user. The medium access control is based on carrier sense (CSMA) and on active signaling. Active signaling is an original feature which makes it possible to merge priority contention and collision detection into an elegant and efficient channel access mechanism. In particular it is expected to boost the performance of HIPERLAN above the level of IEEE 8802.3 standard (Ethernet). It also gives the opportunity to mix time constraint traffics (like voice) with non-time constraint traffics (data) by making use of access priorities.  相似文献
