Cis-9,trans-11 and trans-7,cis-9 CLA are the most prevalent CLA isomers in milkfat. The majority of cis-9,trans-11 CLA is synthesized endogenously by Δ9-desaturase. We tested the hypothesis that trans-7,cis-9 CLA originates from endogenous synthesis by inhibiting Δ9-desaturase with a source of cyclopropene FA (sterculic oil: SO) or with a trans-10,cis-12 CLA supplement. Experiment 1 (four cows; Latin square) involved four treatments: control, SO, partially hydrogenated vegetable oil (PHVO), and PHVO+SO. Milk, plasma, and rumen fluid were collected. Experiment 2 treatments (four cows) were 0 or 14.0 g/d of 10,12 CLA supplement; milk and plasma were collected. Samples were analyzed by GC and Ag+-HPLC to determine FA. In Experiment 1, SO decreased milkfat content of trans-7,cis-9 CLA by 68 to 71% and cis-9,trans-11 CLA by 61 to 65%. In Experiment 2, the 10,12 CLA supplement decreased milkfat content of trans-7,cis-9 CLA and cis-9,trans-11 by 44 and 25%, respectively. Correcting for the extent of treatment-induced inhibition of Δ9-desaturase based on changes in myristic and myristoleic acids, endogenous synthesis of trans-7,cis-9 CLA represented 85 and 102% in Experiments 1 and 2, respectively. Similar corrected values were 77 and 58% for endogenous synthesis of cis-9,trans-11 CLA. Thus, milkfat cis-9,trans-11 CLA was primarily from endogenous synthesis with a minor portion from rumen escape. In contrast, trans-7,cis-9 CLA was not present in rumen fluid in significant amounts. Results indicate this isomer in milkfat is derived almost exclusively from endogenous synthesis via Δ9-desaturase.  相似文献

Quiescent mammalian fibroblasts can be induced to reenter the cell cycle by growth factors and oncoproteins. We studied the pathway(s) through which v-Src, the oncogenic tyrosine kinase encoded by the v-src oncogene of Rous sarcoma virus, forces serum-starved NIH3T3 cells to enter S-phase. To this purpose, we isolated and characterized a polyclonal population of NIH3T3 cells transformed by the MR31 retroviral vector, encoding G418 resistance and the v-src temperature-sensitive allele from the mutant ts LA31 PR-A. NIH(MR31) cells displayed a temperature-conditional transformed phenotype and could be made quiescent by serum deprivation at the restrictive temperature. Serum stimulation or thermolabile v-Src reactivation induced entry into S-phase to a comparable extent, although with different kinetics. The data suggest that v-Src mitogenic activity involves early activation of the Erk1/Erk2 MAP kinases with very little tyrosine phosphorylation of the Shc adaptor proteins at least during the early stages of v-Src reactivation and that v-Src-induced S-phase entry was strongly inhibited by drugs affecting MEK or PI 3-kinase. Our results also suggest that down-regulation of gas1 gene expression plays an important role in regulating the efficiency of entry into S-phase triggered by reactivated v-Src and that Gas1 down-regulation does not require PI 3-kinase dependent signals.  相似文献

Nucleophilic reactions of hexafluorobenzene and related polyfluorobenzenes were studied in detail. Reaction of hexafluorobenzene with hydroxides, alcoholates, aqueous amines, and organolithium compounds led to the substitution of one or more fluorine atoms. The structures of the products were determined, using near infrared and nuclear magnetic resonance spectra. Fluorine is replaced more readily than chlorine, bromine, iodine, or other groups. In the majority of the products in which two of the fluorines in hexafluorobenzene were replaced, the substituting groups were para to each other. However, depending on the reagents other orientation effects were noted. The reaction mechanisms were a function of reagents and conditions. The most prevalent mechanism is presumably the displacement of a fluoride anion by another anion, probably via the formation of transition complexes of different lifetimes. However, simple ionization or attack by neutral species may occur under some conditions. The diazotization and oxidation of pentafluoroaniline were also investigated.  相似文献

The regulation of macrophage lipoprotein lipase (LPL) secretion and mRNA expression by atherogenic lipoproteins is of critical relevance to foam cell formation. LPL is present in arterial lesions and constitutes a bridging ligand between lipoproteins, proteoglycans, and cell receptors, thus favoring macrophage lipoprotein uptake and lipid accumulation. We investigated the effects of native and of oxidized lipoproteins on the expression of LPL in an in vitro human monocyte-macrophage system. Exposure of mature macrophages (day 12) to highly copper-oxidized human low density lipoprotein (LDL) (100 microg protein per milliliter) led to marked reduction in the expression of LPL activity (-62%, P<0.01) and mRNA level (-47%, P<0.05); native LDL, acetylated LDL, and LDL oxidized for <6 hours were without effect. The reduction in LPL activity became significant at a threshold of 6 hours of LDL oxidation (-31%, P<0.05). Among the biologically active sterols formed during LDL oxidation, only 7beta-hydroxycholesterol (5 microg/mL) induced a minor reduction in macrophage LPL activity, whereas 25-hydroxycholesterol was without effect. By contrast, lysophosphatidylcholine, whose LDL content increased in parallel with the degree of oxidation, induced significant reductions in LPL activity and mRNA levels at concentrations of 2 to 20 micromol/L (-34% to -53%, P<0.01). Our results demonstrate that highly oxidized LDL (>6-hour oxidation) exerts negative feedback on LPL secretion in human monocytes-macrophages via a reduction in mRNA levels. By contrast, native LDL and mildly oxidized LDL (<6-hour oxidation) did not exert a feedback effect on LPL expression. We speculate that the content of lysophosphatidylcholine and, to a lesser degree, of 7beta-hydroxycholesterol in oxidized LDLs is responsible for the downregulation of LPL activity and mRNA abundance in human monocyte-derived macrophages and may therefore modulate LPL-mediated pathways of lipoprotein uptake during conversion of macrophages to foam cells.  相似文献

This study examines L. L. Carstensen's (see records 1993-98639-005 and 1996-22608-001) socioemotional selectivity theory within and across three cohorts spanning 4 decades. Socioemotional selectivity theory predicts that as individuals age, they narrow their social networks to devote more emotional resources to fewer relationships with close friends and family. Data from 3 cohorts of nationally representative samples were analyzed to determine whether respondents' satisfaction with the size of their social networks differed by age, cohort, or both. Results support socioemotional selectivity theory: More older adults than younger adults were satisfied with the current size of their social networks rather than wanting larger networks. These findings are consistent across all cohorts. Results are discussed with respect to social relationships across the life course. The oxidation of L-aminoethylcysteine (AEC) by L-amino acid oxidase has been compared with that of the respective sulfoxide (AEC-SO) and sulfone (AEC-SO2). Spectral and HPLC analyses of the incubates reveal the formation of the respective cyclic ketimines. While the ketimine coming from AEC is subjected to autooxidation yielding the sulfoxide and other products, the ketimines produced from AEC-SO and AEC-SO2 are more stable and account for almost the total conversion of the substrate in the product. Spectrophotometric and HPLC properties of the ketimine produced from AEC-SO are identical to those reported earlier for the main product of the autooxidation of AEC ketimine, thus confirming its identification. These results could explain the presence of chondrine in biological materials as a product of reduction of AEC-SO ketimine.  相似文献

The hydrogenation of acetone to isopropanol has been studied in the vapour phase over Pt/Al2O3 catalysts. The rate law obtained at a total pressure of 1 atm and temperatures between 303 and 363 K is of the form V=kP0aP1/2H exp (-44 kJ mol?1 RT?1). The kinetic results are consistent with a Langmuir-Hinshelwood hydrogenation mechanism involving a half hydrogenated species and a non-competitive chemisorption of acetone and hydrogen on the platinum surface. The specific activity (calculated per platinum surface atom) has been found to be scarcely dependent on the platinum particle size. It is suggested that the chemisorption sites are made of a very small ensemble of platinum atoms.  相似文献
