Primary bone-derived cell colonization of unconditioned and pre-conditioned Bioglass 45S5 surfaces in vitro 总被引:1,自引:0,他引:1
Samples of Bioglass 45S5, a bioactive glass that reacts chemically on immersion in an aqueous environment and following implantation can bond with bone, were immersed in culture medium with foetal calf serum as a source of proteins for periods of 1–11 days. Energy dispersive spectroscopy (EDS) revealed that over 11 days immersion, the Bioglass surface was rich in Si relative to Ca and P and scanning electron microscopy (SEM) showed that the Bioglass developed surface reaction layers. Samples conditioned in culture medium and previously untreated samples were used as substrates for primary osteoblast or periosteal cell cultures, to allow comparison of the effects of different Bioglass surface development, on subsequent cell attachment. SEM examination revealed that pre-conditioned Bioglass was a more suitable substrate for osteoblast colonization than previously undeveloped Bioglass, whereas periosteal cells colonized all samples equally well and more rapidly than osteoblasts. 相似文献

The influence of water on the physical properties of a hydrogel is important for understanding natural tissues and in designing synthetic materials to replace them. In this study, poly (2-hydroxyethyl methacrylate) (pHEMA) was used as a model system to understand how water interacts with the polymer of a hydrogel. Thermal analysis methods (thermogravimetric analysis coupled to mass spectrometry and differential scanning calorimetry) were used to determine: (i) the total water content of pHEMA gels; (ii) how this water was lost during heating; (iii) the relationship between water content of the gel and its glass transition temperature; and (iv) the behavior of the water in the gel on cooling. Previous researchers have invoked various models to describe the organization of water in a hydrogel. In this study, the simplest model which could explain all of the results from the different thermal analysis techniques was one which consisted of three classes of water: (i) hydration water in close proximity to the polymer; (ii) interstitial water in regions or cavities surrounded by polymer chains; and (iii) bulk water. 相似文献

Bioabsorbable materials have been widely used in the repair of damaged tissue as well as in the controlled release of drugs and as a supports for cultured cells. The degradation time of poly-L-(lactic acid) (PLLA) may be controlled by altering the polymer porosity through the addition of the plasticizer triethylcitrate. This in turn influences the extent cellular infiltration. In this study, we examined the degradation of PLLA membranes containing different concentrations of plasticizer. PLLA discs were implanted subcutaneouly in rats and withdrawn 2, 14 and 60 days after implantation. The samples were processed for light microscopy and scanning electron microscopy (SEM). Polymer degradation was proportional to the concentration of plasticizer, indicating that triethylcitrate could affect the degradation time of the implants, without damaging the polymer biocompatibility. 相似文献

Poly(2-hydroxyethyl methacrylate) (pHEMA) hydrogels have been used, or suggested for use, in a wide range of biomedical applications. In many of these applications, the mechanical properties of the gel are important for its proper functioning. These properties are influenced by a number of factors, including water content. In this study the storage and loss shear moduli were measured as a function of frequency for gels with water contents ranging from 22% to 48% at a temperature of 37 °C. At low frequencies and high water contents, deformation frequency had little effect. However, at higher frequencies and lower water contents, both moduli increased markedly with increasing frequency. This can be explained by the gels approaching a glass transition. The curves describing the behavior of each gel were combined to form a master curve, using a method analogous to the time–temperature superposition principle. This master curve can be used to predict the shear moduli for gels with a wide range of water contents and loading frequencies. For example, for a gel with a water content of 47.8% (as a percentage of the mass of gel), the curve provides shear moduli values over a frequency range of 10–2–104 Hz. 相似文献

X-ray microradiographs of small biological objects, such as animals and plant materials at micrometric resolution, are currently performed by various methods, all of which are limited by the resolution or the dynamic range of the image detectors. Here a novel X-ray image detector is discussed, in which the previous limitations have been overcome. The objectives of this study were to determine the effects of ovariectomy and growth hormone on mammary epithelial cell proliferation and estrogen receptor alpha (ER alpha) expression within the bovine mammary gland. Two experiments were performed. In the first experiment, eight Holstein heifer calves aged between 1 and 3 mo were ovariectomized, while six calves served as controls. At 6 mo of age, calves were treated with bromodeoxyuridine (BrdU) to label proliferating cells and sacrificed 2 h later. Coinciding with reduced mammary mass (304 +/- 25 vs. 130 +/- 21 g), proliferation of mammary epithelial cells was significantly lower in ovariectomized heifers compared to control heifers (2.24 vs. 0.25%). ER alpha expression was restricted to mammary epithelial cells and was not observed within intra-lobular stroma of parenchymal tissue. The proportion of ER alpha positive cells was significantly higher in ovariectomized heifers than in controls (36.1% +/- 2.2 vs. 46.7% +/- 2.4). In the second experiment, mammary biopsies were taken from five 6-mo-old heifers, immediately preceding and 7 d following a single injection of bovine growth hormone. Mammary epithelial cell proliferation (assessed by incorporation of 3H-thymidine) was increased by growth hormone. The proportion of ER alpha positive mammary epithelial cells was not increased by growth hormone. In conclusion, reduced mammary epithelial cell proliferation following ovariectomy was associated with an increase in ER alpha expression, whereas increased proliferation caused by bovine growth hormone was not associated with changes in the proportion of ER alpha positive cells. 相似文献

The effect of inherent location uncertainty on the detection of stationary targets was determined in noisy image sequences. Targets were thick and thin projected cylinders mimicking arteries, catheters, and guide wires in medical imaging x-ray fluoroscopy. With the use of an adaptive forced-choice method, detection contrast sensitivity (the inverse of contrast) was measured both with and without marker cues that directed the attention of observers to the target location. With the probability correct clamped at 80%, contrast sensitivity increased an average of 77% when the marker was added to the thin-cylinder target. There was an insignificant effect on the thick cylinder. The large enhancement with the thin cylinder was obtained even though the target was located exactly in the center of a small panel, giving observers the impression that it was well localized. Psychometric functions consisting of d' plotted as a function of the square root of the signal-energy-to-noise-ratio gave a positive x intercept for the case of the thin cylinder without a marker. This x intercept, characteristic of uncertainty in other types of detection experiments, disappeared when the marker was added or when the thick cylinder was used. Inherent location uncertainty was further characterized by using four different markers with varying proximity to the target. Visual detection by human observers increased monotonically as the markers better localized the target. Human performance was modeled as a matched-filter detector with an uncertainty in the placement of the template. The removal of a location cue was modeled by introducing a location uncertainty of approximately equals 0.4 mm on the display device or only 7 microm on the retina, a size on the order of a single photoreceptor field. We conclude that detection is affected by target location uncertainty on the order of cellular dimensions, an observation with important implications for detection mechanisms in humans. Peripheral neural stimulation is a major problem in current gradient coil designs. Induced current problems in patients relate
directly to gradient strength and modulation frequency. Current designs of gradient coil tend to limit ultra-high-speed imaging
methods such as echo-planar imaging through the effect of induced currents which produce tingling sensations and involuntary
muscle twitch. Neural stimulation could also trigger epileptic fits and/or cardiac fibrillation. For reduction of induced
currents, an important aspect is the coil geometry. It is desirable to design the gradient coil in such a way as to prevent
closed loop circulating currents within the body. Preliminary results using a four-sector gradient coil with rectangular geometry,
operating in a low mutual coupling mode, indicate significant reduction in the E-field within the subject volume of the coil. Reduction in induced currents in the patient allows safer operation at higher
magnetic field strengths together with faster scans currently prohibited through neural stimulation effects in standard coil

Since cyclodextrin gas chromatography columns became popular for chiral separations, many researchers have noticed high enantiomeric ratios [ER: (+)-enantiomer/(-)-enantiomer] for alpha-HCH in the brains of wildlife. This investigation used the laboratory rat as a model for these phenomena. Rats were either pretreated with phenobarbital (PB) or left untreated and then dosed with alpha-HCH. Animals were sacrificed after 1 or 24 h. The ER averaged 0.95 +/- 0.01 in blood, 1.29 +/- 0.02 in fat, and 0.77 +/- 0.004 in liver. ERs in brain ranged from 2.8 +/- 0.5 to 13.5 +/- 0.4. Both the tissue concentration distribution and the ERs agree well with those previously reported in wildlife. To determine whether high brain ERs were due to enantioselective metabolism or transport through the blood-brain barrier, alpha-HCH exposed brain and liver tissue slices were compared. Concentrations in the brain slices did not decrease with PB pretreatment but did decrease in the liver slices. Enantiomeric ratios in the brain slices averaged 1.11 +/- 0.02 and were 0.76 +/- 0.03 in liver slices for the PB pretreated rats. These data indicate that the enantioselective metabolism of alpha-HCH by the brain is not the mechanism responsible for high ERs in this tissue. 相似文献
