Until now, attention has been focused solely in the drying of squid mantle from an experimental point of view, neglecting the transport phenomenon studies of water in squid muscle. This work studies the drying of squid mantle (Loligo brasiliensis), previously salted and smoked by liquid smoking (hickory extract), using a tubular dryer in closed cycle with a silicagel fixed bed in series. The mass transfer phenomenon during drying was studied, based on the Fick's second law, with the effective diffusivity supposed constant, which enabled an analytical solution to the problem. The drying curve calculated with the resulting equation was compared with experimental data. The model was applied to a hollow cylinder geometry (round squid mantle), with its internal surface isolated from the drying environment by a plastic film. The latter was given a tubular form and introduced inside the mantle cavity (impervious wall boundary condition).  相似文献

Methanol-induced conformational transitions of hen egg white lysozyme were investigated with a combined use of far- and near-UV CD and NMR spectroscopies, ANS binding and small-angle X-ray scattering. Addition of methanol induced no global change in the native conformation itself, but induced a transition from the native state to the denatured state which was highly cooperative, as shown by the coincidence of transition curves monitored by the far- and near-UV CD spectroscopy, by isodichroic points in the far- and near-UV CD spectra and by the concomitant disappearance of individual 1H NMR signals of the native state. The ANS binding experiments could detect no intermediate conformer similar to the molten globule state in the process of the methanol denaturation. However, at high concentration of methanol, e.g., 60% (v/v) methanol/water, a highly helical state (H) was realized. The H state had a helical content much higher than the native state, monitored by far-UV CD spectroscopy, and had no specific tertiary structure, monitored both by near-UV CD and NMR spectroscopy. The radius of gyration in the H state, 24.9 angstroms, was significantly larger than that in the native state (15.7 angstroms). The Kratky plot for the H state did not show a clear peak and was quite similar to that for the urea-denatured state, indicating a complete lack of globularity. Thus we conclude that the H state has a considerably expanded, flexible broken rod-like conformation which is clearly distinguishable from the "molten globule" state. The stability of both N and H states depends on pH and methanol concentration. Thus a phase diagram involving N and H was constructed.  相似文献

Plant class I glucan endo-1,3-beta-glucosidases (beta-1,3-glucanase; 1,3-beta-D-glucan glucanohydrolase, EC 3.2.1.39) have been implicated in development and defense against pathogen attack. Nevertheless, beta-1,3-glucanase deficiencies generated by antisense transformation of Nicotiana sylvestris and tobacco have little biological effect. We report here that another beta-1,3-glucanase activity is induced in these deficient mutants after infection with necrotizing viruses. Induction of class I beta-1,3-glucanase was markedly inhibited in leaves of N. sylvestris and tobacco antisense transformants infected with tobacco necrosis virus and tobacco mosaic virus, respectively. A serologically distinct beta-1,3-glucanase activity was present in the infected antisense transformants but was absent in both healthy and infected control plants and in antisense transformants treated with the stress hormone ethylene. Immunoblot analyses, localization studies, and measurements of antibody specificity indicate that this compensatory beta-1,3-glucanase activity is an intracellular enzyme different from known tobacco beta-1,3-glucanases. Therefore, plants can compensate for a deficiency in enzyme activity by producing a functionally equivalent replacement--i.e., "ersatz"--protein or proteins. The fact that compensation for beta-1,3-glucanase activity occurs in response to infection argues strongly for an important role of these enzymes in pathogenesis.  相似文献

Previous studies have focused on the extension of the Berenger's (1994) absorbing boundary condition (ABC)-the perfectly matched layer (PML)-to anisotropic and dispersive media. We describe how the PML can be extended for anisotropic and/or dispersive media (or even bianisotropic media) with little analytical effort, in a simple and general conceptual setting, by using an analytical continuation of the coordinate space of the Maxwell's equations to a complex variables domain. The impact of parametric variations on digital circuit performance is increasing in nanometer Integrated Circuits (IC), namely of Process, power supply Voltage and Temperature (PVT) variations. Moreover, circuit aging also impacts circuit performance, especially due to Negative Bias Temperature Instability (NBTI) effect. A growing number of physical defects manifest themselves as delay faults (at production, or during product lifetime). On-chip, on-line delay monitoring, as a circuit failure prediction technique, can be an attractive solution to guarantee correct operation in safety–critical applications. Safe operation can be monitored, by predictive delay fault detection. A delay monitoring methodology and a novel delay sensor (to be selectively inserted in key locations in the design and to be activated according to user’s requirements) is proposed, and a 65 nm design is presented. The proposed sensor is programmable, allowing delay monitoring for a wide range of delay values, and has been optimized to exhibit low sensitivity to PVT and aging-induced variations. Two MOSFET models—BPTM and ST—have been used. As abnormal delays can be monitored, regardless of their origin, both parametric variations and physical defects impact on circuit performance can be identified. Simulation results show that the sensor is effective in identifying such abnormal delays, due to NBTI-induced aging and to resistive open defects.  相似文献

The purpose of this paper is to present a methodology and tools for the design and test of an EN298 compliant ASIC chip for a safety-critical gas burner control system. Safe operation, as far as the critical variable is concerned, is guaranteed in the presence of two simultaneous faults. We report on an extension of finite-difference time-domain (FDTD) subgridding (SG) algorithms incorporating digital filters and domain-overriding to three-dimensional (3-D) simulations and to problems involving materials traversing the SG interfaces. The field of Systems Biology is a rapidly evolving area of research. It follows on from the previous experimental and theoretical 'omics' revolution in biology. Now that we have through the use of these tools many 'indices' of biological systems available the next step is to actually start composing the systems that these indices specify. In this paper we will discuss the developments in the field of Systems Biology as they pertain to predictive food microbiology and give an example of state of the art current approaches. The data discussed in the case study deal with the resistance of the yeast Saccharomyces cerevisiae towards environmental temperature changes through adaptation of its metabolism, protein signalling and gene-expression. The results are integrated and its implications for the definition of new experiments discussed; the iteration between experiment driven model definition and model driven experimentation being characteristic for contemporary Systems Biology approaches. The stress condition discussed represents in no way a practical situation in food microbiology but what it teaches may well be applied in such cases. We will indicate how the latter may be achieved.  相似文献
