We investigated the effect of chronic exposure (3 days) with low-density lipoprotein (LDL) and oxidized (Ox)-LDL on the unstimulated and stimulated formation of prostacyclin (6-keto-prostaglandin [PG]F1 alpha) and total inositol phosphates (IPs) by cultured bovine aortic endothelial cells. Neither basal nor bradykinin-stimulated (1 to 10 nmol/L) formation of 6-keto-PGF1 alpha was affected by LDL, except at the highest concentration of bradykinin tested (100 nmol/L). In the presence of the antioxidants N-acetyl-L-cysteine (NAC, 10 mumol/L) or vitamin E (100 mumol/L), basal and bradykinin-stimulated formation of 6-keto-PGF1 alpha was potentiated by 20 micrograms protein/mL of LDL. Ox-LDL decreased unstimulated formation of the eicosanoid from 3.1 +/- 0.2 pg/micrograms protein in control cells to 1.6 +/- 0.1 and 0.5 +/- 0.1 pg/microgram protein after 3-day incubation with 5 and 20 micrograms protein/mL of Ox-LDL, respectively (P < .05). As in the basal state, Ox-LDL decreased bradykinin-induced 6-keto-PGF1 alpha formation. NAC or vitamin E did not influence Ox-LDL-induced endothelial cell changes in eicosanoid production. IPs formation by endothelial cells increased to a similar extent in the presence of 20 micrograms protein/mL of either LDL or Ox-LDL. However, no change was apparent in the bradykinin (10 mumol/L)-induced increase in total IPs formation after incubation with the lipoproteins. The data indicate that chronic exposure to Ox-LDL abolishes the production of prostacyclin by cultured endothelial cells. This paper is concerned with the problem of pipe whip, the dynamic response of a high pressure piping system subjected to an end force as the result of a pipe break which releases a jet of fluid from the broken section. Both experimental and theoretical results are presented concerning the dynamic elastic-plastic behaviour of cantilever pipes subjected to a transverse force pulse at the free end. Comparisons between experimental data and theoretical predictions are made for mild-steel pipes with outer diameter-to-thickness ratios of 19.5, 28 and 32. It is demonstrated that, for these geometries, the whipping pipes display three characteristically different responses, viz. elastic, plastic hardening behaviour for thick pipes, elastic, plastic hardening-softening behaviour for moderately thick pipes and elastic, plastic hardening-softening-collapse behaviour for thinner pipes. The experimental data taken from a series of high-speed films are compared with the predictions of the instantaneous shapes of the whipping pipes derived from both a rigid, perfectly-plastic, large deflection, dynamic beam model and a more comprehensive model which incorporates the effects of elasticity and plastic hardening and softening, the details of which are presented in the paper.  相似文献

Large scale use of lysozyme for periplasmic release has been impeded by the cost of the pure enzyme and its subsequent presence as a contaminant in later downstream processing steps. In this paper, we discuss the use of lysozyme for pilot scale recovery of a periplasmic enzyme from E. coli. The effects of concentration of sucrose, lysozyme and cells on periplasmic enzyme release were examined. Lysozyme concentration can be reduced 5-fold from previous reports and a reduction in sucrose concentration from 20 to 15% (w/v) allows an improvement in centrifugal harvesting by reducing viscosity. High levels of release were still achieved using this technique and further improvements in yield were obtained by optimising other components of the releasing mixture. Results show that some release is still achieved in circumstances where no lysozyme use is possible. Results also indicate that a substantial proportion (up to 70%) of lysozyme remains bound to the cellular debris after its action and is removed with this material.  相似文献

BACKGROUND AND OBJECTIVES: We operated on three patients with leiomyosarcoma of the inferior vena cava. METHODS: Complete excision was possible in all three patients. RESULTS: One patient developed widespread metastasis at 23 months, one patient is alive with no evidence of disease at 70 months, and one patient is alive at 15 months. The third patient had subcutaneous and pulmonary metastases at the time of presentation, which are radiologically nondetectable at present following postoperative chemotherapy. CONCLUSIONS: The clinicopathologic features, prognostic factors, and treatment of 130 cases found in a comprehensive literature search and our three cases are reported.  相似文献

Charybdotoxin (ChTX), a venom protein, suppresses Ca2+-activated K+ (K+(Ca)) currents in the glomus cell of neonatal rat carotid body. If it works similarly for cat carotid body chemoreceptors, charybdotoxin is expected to stimulate the chemosensory discharge during normoxia, and particularly hypoxia and hypercapnia. We studied the effects of charybdotoxin (20-40 nM) in vitro (perfused/superfused) on the cat carotid chemosensory discharge, and simultaneously tissue PO2 (PtiO2), as a measure of positive control. ChTX (20 nM) only increased PtiO2 and decreased carotid chemosensory discharge during hypoxia, indicating vasodilation. We conclude that K+(Ca) channels do not appear to play a significant role in chemotransduction in the cat carotid body.  相似文献

Librarians, like most professionals, may arrive at a stage in their careers when they start questioning themselves about their job satisfaction and importance. The reasons are many, from not being challenged by their work to abrupt changes in the workplace dictated by management and the economy. At this stage, professionals may begin to feel unappreciated by management and not in control of their work environment. In this piece, I offer some suggestions that could help mid-career librarians to re-energize themselves and survive in a workplace that is in constant flux. How you react to the anxiety of "job disconnect" will affect the rest of your career life. Should you become more proactive at your job, increasing your responsibility and value to your institution? What does your personality have to do with how you adjust to management changes? Does tenure fit into this equation? Should you even think about changing your job at this stage of your career?
