DNA topoisomerases have been proposed as the proteins involved in the formation of the DNA-protein cross-links detected after ultraviolet light (UV) irradiation of cellular DNA. This possibility has been investigated by studying the effects of UV-induced DNA damage on human DNA topoisomerase I action. UV lesions impaired the enzyme's ability to relax negatively supercoiled DNA. Decreased relaxation activity correlated with the stimulation of cleavable complexes. Accumulation of cleavable complexes resulted from blockage of the rejoining step of the cleavage-religation reaction. Mapping of cleavage sites on the pAT153 genome indicated UV-induced cleavage at discrete positions corresponding to sites stimulated also by the topoisomerase I inhibitor camptothecin, except for one. Subsequent analysis at nucleotide level within the sequence encompassing the UV-specific cleavage site revealed the precise positions of sites stimulated by camptothecin with respect to those specific for UV irradiation. Interestingly, one of the UV-stimulated cleavage sites was formed within a sequence that did not contain dimerized pyrimidines, suggesting transmission of the distortion, caused by photodamage to DNA, into the neighboring sequences. These results support the proposal that DNA structural alterations induced by UV lesions can be sufficient stimulus to induce cross-linking of topoisomerase I to cellular DNA.  相似文献

Lattice-matched InAlAs-InGaAs HEMTs with dry etched and wet etched gate recesses have been fabricated and both high-frequency and noise measurements have been carried out. The highly selective dry etching process ensures uniform device parameters. The small signal and noise performance shows only minor differences between the two transistor types. There is no evidence of detrimental effects caused by dry etching that reduce the electrical and noise performance of the device at high frequencies. These results show that dry etched InP HEMT's have suitable characteristics for the fabrication of MM-wave integrated circuits  相似文献

In this paper, the results of a laboratory investigation conducted with heat evolution of high-volume fly ash (HVFA) concrete are presented. Heat evolution of concrete was studied by measuring the temperature increase in concrete under adiabatic curing condition. Characteristic of heat evolution of fly ash concrete was found to be strongly dependent on the replacement level of fly ash and dosage of superplasticizer used to maintain workability. It was also found that using fly ash as cement replacement resulted in a reduction on the maximum temperature rise. Increasing the replacement level of fly ash caused lower temperature rise in concrete. Superplasticizer caused a delay in peak temperature rise time; this is taken as an indicator that high-dosage superplasticizer used in concrete caused retardation in hydration of cement. Concretes having similar ingredients showed similar peak temperature rise whether they are superplasticized or not.  相似文献

Submicronic powders of Bi4Ti3O12 with different morphologies were prepared by both the oxalate coprecipitation and the conventional mixing oxides methods. Compacts of the two calcined powders were sintered at 850–1100 °C in air, and the densification process was studied by non-isothermal and dilatometric experiments. A rapid densification (> 97% theoretical density) below 875 °C took place in the Bi4Ti3O12 oxalate powder which was attributed to an extremely uniform pore-size distribution in the green compact. The possible formation of a transient liquid which promotes densification also was taken into account. The development of plate-like morphology in the conventional Bi4Ti3O12 powder, broad pore-size distribution, and the plate-like colony formation, hindered rapid densification of the green compacts at low temperature. Microstructural development was studied; preliminary dielectric and electrical results are also reported.  相似文献

Three studies investigated conditions in which perceivers view dispositions and situations as interactive, rather than independent, causal forces when making judgments about another's personality. Study 1 showed that perceivers associated 5 common trait terms (e.g., friendly and shy) with characteristic if...then... (if situation a, then the person does x, but if situation b, then the person does y) personality signatures. Study 2 demonstrated that perceivers used information about a target's stable if...then... signature to infer the target's motives and traits; dispositional judgments were mediated by inferences about the target's motivations. Study 3 tested whether perceivers draw on if...then... signatures when making judgments about Big Five trait dimensions. Together, the findings indicate that perceivers take account of person-situation interactions (reflected in if...then... signatures) in everyday explanations of social behavior and personality dispositions. The computer industry has evolved very rapidly from single-user computers to computer networks where users are able to share both local and remote files. Networks of microcomputers facilitate the integration of all information processing for distributed applications such as database processing and electronic mail. One management application of promising potential for computer networks is distributed simulation. Simulation analysis can be useful to essentially all problem-solving and decision-making on the job.

To implement a particular distributed application, computer communication between processors must be considered. Unlike expensive multiprocessor computers, networks of less-expensive microcomputers do not have pre-established communication paths between processors. This paper addresses how this obstacle may be overcome by using communication protocols based on the Open Systems Interconnection (OSI) reference model. Protocol services needed to support a distributed simulation environment will be identified, and their implementation through a prototype will then be investigated and evaluated.  相似文献

The biosynthesis of fatty acids in the diatomPhaeodactylum tricornutum was studied. The diatom was incubated with sodium [114C] acetate and the acids [1-14C] palmitic, [1-14C] stearic, [1-14C] linoleic and [1-14C] α-linolenic. The distribution of radioactivity in the products was determined by gas liquid radiochromatography. The diatom synthesized “de novo” not only saturated and monounsaturated fatty acids, but also linoleic, α-linolenic and other fatty acids including the highly polyunsaturated 20∶5ω3 and 22∶6ω3. When labeled acetate, stearic, α-linolenic or even linoleic acid were incubated with the diatom, the polyunsaturated C20 fatty acids synthesized belonged predominantly to the ω 3 family. The existence of Δ9, Δ6, Δ5, Δ4, ω6 and possibly ω3 desaturases inP. tricornutum is suggested. Two important issues in computational modelling in cognitive neuroscience are: first, how to formally describe neuronal networks (i.e. biologically plausible models of the central nervous system), and second, how to analyse complex models, in particular, their dynamics and capacity to learn. We make progress towards these goals by presenting a communicating automata perspective on neuronal networks. Specifically, we describe neuronal networks and their biological mechanisms using Data-rich Communicating Automata, which extend classic automata theory with rich data types and communication. We use two case studies to illustrate our approach. In the first case study, we model a number of learning frameworks, which vary in respect of their biological detail, for instance the Backpropagation (BP) and the Generalized Recirculation (GeneRec) learning algorithms. We then used the SPIN model checker to investigate a number of behavioral properties of the neural learning algorithms. SPIN is a well-known model checker for reactive distributed systems, which has been successfully applied to many non-trivial problems. The verification results show that the biologically plausible GeneRec learning is less stable than BP learning. In the second case study, we presented a large scale (cognitive-level) neuronal network, which models an attentional spotlight mechanism in the visual system. A set of properties of this model was verified using Uppaal, a popular real-time model checker. The results show that the asynchronous processing supported by concurrency theory is not only a more biologically plausible way to model neural systems, but also provides a better performance in cognitive modelling of the brain than conventional artificial neural networks that use synchronous updates. Finally, we compared our approach with several other related theories that apply formal methods to cognitive modelling. In addition, the practical implications of the approach are discussed in the context of neuronal network based controllers.  相似文献
