Proximate composition, amino acid and fatty acid profiles, and mineral and vitamin contents of by‐products from Atlantic sea cucumber (Cucumaria frondosa) and green urchin (Strongylocentrotus droebachiensis) processing plants were investigated. Results showed that sea cucumber viscera, urchin digestive tract (UDT) and non‐commercial grade gonads contained 4.5–14.4% total proteins and 1.9–4.6% lipids (wet weight basis). They were rich in essential amino acids with values of about 36% of total amino acids. Samples were also rich in polyunsaturated fatty acids (44% of total fatty acids) and showed high content in EPA (~17% of total fatty acids), but low DHA level (0.2%) was observed. In terms of vitamins content, UDT showed highest retinol and alpha‐tocopherol contents when compared to other marine samples. Results showed that by‐products from echinoderm processing plants contain various nutritious components with levels generally comparable even better than their edible counterparts, and efforts should be invested to valorise these biological wastes in a world context of rarefaction of nutritional marine resources.  相似文献

We report on the case of a girl with an immune deficiency characterized by recurrent infections of the upper and lower respiratory tract, low IgG and IgA serum levels as well as deficiency of the in vivo antibody response. Since this patient is the sister of a boy affected with a hyper-IgM syndrome due to a defect in CD40 ligand (CD40L) expression, the involvement of CD40L in this phenotypic expression was investigated. A very low fraction of activated T cells (5%) in this female patient expressed CD40L. This resulted from the presence of a heterozygous CD40L nonsense mutation associated with a skewed pattern of X chromosome inactivation as determined by methylation pattern analysis. Although carriers of X-linked hyper-IgM are considered to be asymptomatic, this study indicates that extreme lyonization of the normal X can lead to a mild expression of the hyper-IgM syndrome which is similar to common variable immune deficiency (CVID). Therefore, it is possible that some cases of CVID in females represent partial deficiency of CD40L expression in carriers of the CD40L mutation.  相似文献

Non-specific X-linked mental retardation (MRX) is a very common disorder which affects approximately 1 in 600 males. Despite this high frequency, little is known about the molecular defects underlying this disorder, mainly because of the clinical and genetic heterogeneity which is evident from linkage studies. Recently, a collaborative study using the candidate gene approach demonstrated the presence of mutations in GDIalpha, a Rab GDP-dissociation inhibitor encoded by a gene localized in Xq28, associated with non-specific mental retardation. GDIalpha is mainly a brain-specific protein that plays a critical role in the recycling of Rab GTPases involved in membrane vesicular transport. The study presented here was designed to assess the prevalence of mutations in the GDIalpha in mentally retarded patients and to discuss the clinical phenotypes observed in affected individuals. Mutation screening of the whole coding region of the GDIalpha gene, using a combination of denaturing gradient gel electrophoresis and direct sequencing, was carried out in 164 patients found negative for expansions across the FRAXA GCC repeat. In addition to the nonsense mutation recently reported in MRX48, we have identified a novel missense mutation in exon 11 of the GDIalpha gene in one familial form of non-specific mental retardation. In this family (family R), all affected males show moderate to severe mental retardation, and the X-linked semidominant inheritance is strongly suggested by the severe phenotypes in males with respect to mildly affected females or unaffected obligatory carriers. This study showed that the prevalence of GDIalpha mutations in non-specific mental retardation could be estimated to be 0.5-1%, and molecular diagnosis and genetic counselling in some cases of non-specific mental handicap can now be provided.  相似文献

Biologically active sand filters within water treatment plants (WTPs) are now recognised as an effective barrier for the removal of geosmin. However, little is known regarding the actual microbiological processes occurring or the bacteria capable of degrading geosmin. This study reports the enrichment and isolation of a Gram-negative bacterium, Geo48, from the biofilm of a WTP sand filter where the isolate was shown to effectively degrade geosmin individually. Experiments revealed that Geo48 degraded geosmin in a planktonic state by a pseudo-first-order mechanism. Initial geosmin concentrations ranging from 100 to 1000 ng/l were shown to directly influence geosmin degradation in reservoir water by Geo48, with rate constants increasing from 0.010 h−1 (R2 = 0.93) to 0.029 h−1 (R2 = 0.97) respectively. Water temperature also influenced degradation of geosmin by Geo48 where temperatures of 11, 22 and 30 °C resulted in rate constants of 0.017 h−1 (R2 = 0.98), 0.023 h−1 (R2 = 0.91) and 0.019 h−1 (R2 = 0.85) respectively. Phylogenetic analysis using the 16S rRNA gene of Geo48 revealed it was a member of the Alphaproteobacteria and clustered with 99% bootstrap support with an isolate designated Geo24, a Sphingopyxis sp. previously described as degrading geosmin but only as a member of a bacterial consortium. Of the previously described bacteria, Geo48 was most similar to Sphingopyxis alaskensis (97.2% sequence similarity to a 1454 bp fragment of the 16S rRNA gene). To date, this is the only study to report the isolation and characterisation of a Gram-negative bacterium from a biologically active sand filter capable of the sole degradation of geosmin.  相似文献

The Rural Utilities Services (RUS) and the National Rural Electric Cooperative Association (NRECA) collaborated to form the Transmission and Distribution Engineering Committee (T&DEC). This committee was primarily formed to assist in updating RUS Bulletins to current standards and practices being used by electric cooperatives. This article highlights the two recent distribution reliability activities of the committee: the NRECA reliability online study and the revision of RUS Bulletin 161-1-"interruption reporting and service continuity objectives for electric distribution systems". An online reliability reporting practices survey of NRECA distribution cooperatives have conducted for the past two years and the results are discussed here. Electrochemistry and membrane ultrafiltration methods (electro-oxidation and liquid phase polymer based retention technique, LPR, respectively) were off-line coupled to remove As(III) inorganic species from aqueous solutions. Our main objective was to achieve an efficient extraction of arsenic species by associating a polymer-assisted liquid phase retention procedure, based on the As(V) adsorption properties of cationic water-soluble polymers, with an electrocatalytic oxidation process of As(III) into its more easily removable analogue As(V). The electrocatalytic oxidation of As(III) to As(V) was performed in the presence of different water-soluble poly(quaternary ammonium) salts acting also as supporting electrolyte, i.e. poly(vinylbenzyl)trimethyl ammonium chloride, P(ClVBTA), poly[3-(methacryloylamine)propyl]trimethyl ammonium chloride, P(ClMPTA), and poly(4-vinyl-1-methylpyridinium bromide), P(BrVMP). After complete electrocatalytic conversion of As(III) into As(V), the mixtures were introduced into an LPR cell to remove the As(V)-polymer adducts. Using P(ClMPTA), P(ClVBTA), or P(BrVMP) ammonium salts in a 20:1 polymer:As(III) mol ratio at pH 8, complete (100%) retention of the arsenic was achieved. Moreover, the As(V) retention efficiency turned out to be directly related to the net charge consumed during the electrochemical conversion of As(III) to As(V).  相似文献

Mechanosensitive ion channels of the archaeon Haloferax volcanii   总被引:1,自引:0,他引:1  
Mechanosensitive (MS) ion channels have been documented in a variety of cells belonging to Eukarya and Eubacteria. We report the novel finding of two types of MS ion channels in the cell membrane of the halophilic archaeon Haloferax volcanii, a member of the Archaea that comprise the third phylogenetic domain. The two channels, MscA1 and MscA2, differed in their kinetic properties with MscA1 exhibiting more frequent open-closed transitions than MscA2. Both channels have large conductances that rectify between -40 mV and +40 mV where the conductance of MscA1 ranged from 380 to 680 picosiemens, whereas MscA2 ranged from 850 to 490 picosiemens. Both channels were blocked by submillimolar gadolinium. In addition, the channels of either membrane vesicles or detergent-solubilized membrane proteins remained functional upon reconstitution into artificial liposomes, a result that indicates that these channels are activated by mechanical force transmitted via the lipid bilayer alone. Subsequently a 37-kDa protein corresponding to the MscA1 channel activity was purified. With the possible functional similarity to bacterial MS channels, our finding of MS channels in Archaea emphasizes the ubiquity and importance of these channels in all domains of the evolutionary tree.  相似文献
